Transplanted ossicles after two decades.

Abstract

The authors now report on the histological appearance of two very long-term ossicles, an autologous incus removed from the middle ear at revision surgery 21 years after repositioning and a homologous incus removed from the middle ear after 20 years. Having been placed in formalin immediately after removal from the ear, both specimens were later decalcified by ethylene diamine tetra-acetic acid, embedded in paraffin wax, serially sectioned and stained with haematoxylin and eosin.

A synergistic approach to the design, fabrication and evaluation of 3D printed micro and nano featured scaffolds for vascularized bone tissue repair.

3D bioprinting has begun to show great promise in advancing the development of functional tissue/organ replacements. However, to realize the true potential of 3D bioprinted tissues for clinical use requires the fabrication of an interconnected and effective vascular network. Solving this challenge is critical, as human tissue relies on an adequate network of blood vessels to transport oxygen, nutrients, other chemicals, biological factors and waste, in and out of the tissue. Here, we have successfully designed and printed a series of novel 3D bone scaffolds with both bone formation supporting structures and highly interconnected 3D microvascular mimicking channels, for efficient and enhanced osteogenic bone regeneration as well as vascular cell growth. Using a chemical functionalization process, we have conjugated our samples with nano hydroxyapatite (nHA), for the creation of novel micro and nano featured devices for vascularized bone growth. We evaluated our scaffolds with mechanical testing, hydrodynamic measurements and in vitro human mesenchymal stem cell (hMSC) adhesion (4 h), proliferation (1, 3 and 5 d) and osteogenic differentiation (1, 2 and 3 weeks). These tests confirmed bone-like physical properties and vascular-like flow profiles, as well as demonstrated enhanced hMSC adhesion, proliferation and osteogenic differentiation. Additional in vitro experiments with human umbilical vein endothelial cells also demonstrated improved vascular cell growth, migration and organization on micro-nano featured scaffolds.

Transfer of apatite coating from porogens to scaffolds: uniform apatite coating within porous poly(DL-lactic-co-glycolic acid) scaffold in vitro.

Strategies to bone tissue engineering have focused on the use of synthetic or natural degradable materials as scaffolds for cell transplantation to guide bone regeneration. Biocompatibility, biodegradability, biomechanical integrity, and osteoconductivity are important requirements for the scaffold materials. This study explored a new approach of apatite coating to enhance the osteoconductivity of a synthetic degradable poly(DL-lactic-co-glycolic acid) (PLGA) scaffold. The new approach was developed to ensure a relatively uniform apatite coating on the interior pore surfaces deep inside a scaffold, even for a relatively thick scaffold with small pores. Apatite was first coated on the surface of paraffin spheres of the desirable sizes. The paraffin spheres were then molded to form a foam. PLGA/pyridine solution was cast into the interspaces among the paraffin spheres. After the paraffin spheres were dissolved and removed by cyclohexane, PLGA scaffold with controlled pore size, good interconnectivity and high porosity was obtained with apatite left on the pore surface uniformly throughout the whole scaffold. The scaffold and apatite coating were characterized using thermogravimetry analysis, scanning electron microscopy, energy dispersive X-ray spectroscopy, and X-ray diffractometry.

Electrospinning biomedical nanocomposite fibers of hydroxyapatite/poly(lactic acid) for bone regeneration.

Development of fibrous matrices of bioceramic-biopolymer nanocomposite offers great potential in the field of bone regeneration and tissue engineering. However, in order to produce electrospun fibers with homogeneous structure, it is essential for the ceramic powder to be fine and to remain stable in suspension. Herein, we developed a novel method whereby the bioceramic hydroxyapatite (HA) was kept in suspension in biopolymer poly(lactic acid) (PLA). The strategy was to introduce a surfactant hydroxysteric acid (HSA) between the hydrophilic HA powder and the hydrophobic chloroform-dissolved PLA. The HA nanopowder was dispersed effectively in HSA and mixed homogeneously with PLA. Continuous and uniform fibers were generated successfully with diameters of approximately 1-2 microm, and featured a well-developed nanocomposite structure of HA nanopowder-dispersed PLA. Initial cellular assays showed excellent cell attachment and proliferation and also enhanced expression of alkaline phosphatase at 7 days of culturing. The HA-PLA nanocomposite fibers may be potentially useful in tissue engineering applications, particularly as three-dimensional substrates for bone growth.
