Computerized tridimensional reconstruction of vegetable structures.

Abstract

By means of a "Personal Computer", a technique has been worked out for the acquisition and processing of data, for the tridimensional reconstruction of vegetable structures starting out from information, in the form of photographic images, obtained by optical or electronic microscope. Examples of the application of this program to vegetable drugs (corolla of Lavandula angustifolia Mill flowers) are given.

Objective measurements of image quality.

Tissue harmonic imaging (THI) and compound imaging have been reported clinically to improve contrast resolution, tissue differentiation and overall image quality. However, there have been limited studies to date to quantify objectively the improvements in image quality achieved with these new imaging techniques. The aim of this study was to quantify differences in image quality that exist between conventional B-mode imaging, harmonic imaging, compound imaging and harmonic compound imaging. An ATL HDI 5000 scanner with three probes (C5-2, L7-4 and L12-5) was tested with two different types of test object, the Gammex-RMI model 404 GS LE and the Gammex-RMI 403 GS LE. The measurement limitations associated with subjective analysis methods were not present in this study because an automated image analysis program was used to determine the image quality parameters. Therefore, subtle differences between the four imaging modes could be detected. Significant improvements in lateral resolution and slice thickness as a function of depth were found with THI. Contrast resolution and anechoic target detection improved with compound imaging, and harmonic compound imaging improved lateral resolution, slice thickness as a function of depth and contrast resolution.

Fast reconstruction of four-dimensional NMR spectra from plane projections.

Four-dimensional NMR spectroscopy can be speeded up by a large factor by a projection-reconstruction technique related to that used in X-ray tomography. The limited amount of information recorded in a few suitably-tilted projection planes suffices to recreate the entire four-dimensional spectrum. The method is demonstrated by reference to the 500 MHz spectrum of 1 millimolar aqueous solution of ubiquitin by dispersing the responses of the three-dimensional HNCO spectrum into the C(alpha) dimension of a four-dimensional HNCOCA spectrum obtained by projection-reconstruction.

Innovative instrumentation for microarray scanning and analysis: application for characterization of oligonucleotide duplexes behavior.

Accuracy in microarray technology requires new approaches to microarray reader development. A microarray reader system (optical scanning array or OSA reader) based on automated microscopy with large field of view, high speed 3 axis scanning at multiple narrow-band spectra of excitation light has been developed. It allows fast capture of high-resolution, multi-fluorescence images and is characterized by a linear dynamic range and sensitivity comparable to commonly used photo-multiplier tube (PMT)-based laser scanner. Controlled by high performance software, the instrument can be used for scanning and quantitative analysis of any type of dry microarray. Studies implying temperature-controlled hybridization chamber containing a microarray can also be performed. This enables the registration of kinetics and melting curves. This feature is required in a wide range of on-chip chemical and enzymatic reactions including on-chip PCR amplification. We used the OSA reader for the characterization of hybridization and melting behaviour of oligonucleotide:oligonucleotide duplexes on three-dimensional Code Link slides.
