[Occurrence of beta-phenylethylamine and its derivatives in cocoa and cocoa products].

2-Phenylethylamine was extracted from cocoa nibs and chocolates and analysed by coupled gas chromatography-mass spectroscopy. The amine concentration increases in fermentation of cocoa and decreased during roasting and alkalization. Its concentration in chocolates is dependent on non-fat cocoa contents. Previously unreported aldimines were found in cocoa powders, which arise from the condensation of phenylethylamine and aldehydes. The main component of these products is N-phenylmethyl-N-phenylmethylene amine (CAS 3240-95-7).

Confirmation method for the identification and determination of some organophosphorus and organochlorine pesticides in cocoa beans by gas chromatography-mass spectrometry.

The main objective of this work is to develop a routine quality control method for pesticide residues in cocoa beans, using gas chromatography-mass spectrometry. The investigated pesticides, which are used to control pests in the growing of cacao, are: Acephate, Propoxur, HCH, Heptachlor, Fenitrothion, Pirimiphos-methyl, Aldrin, Dieldrin, pp'-DDE, op-DDE and DDT. Two extraction methods were tested. The first was based on strong attack by concentrated sulphuric acid and later extraction with n-hexane: the investigated residues were Acephate, HCH, Fenitrothion and DDT; recoveries were 68-95% and the detection limits 0.5-10 ppb. The second extraction method was based on the Universal Trace Residue Extractor (UNITREX), which consists of a distillation system for organophosphorus and organochlorine pesticides in fatty samples. The investigated residues were Heptachlor, Pirimiphos-methyl, Aldrin, Propoxur, Dieldrin, op-DDE and pp'-DDE; recoveries were 67-88% and the detection limits 1-10 ppb.

Collaborative study of the International Office of Cocoa, Chocolate and Sugar Confectionery on Salmonella detection from cocoa and chocolate processing environmental samples.

A comparative collaborative study was performed in 13 laboratories to evaluate the use of motility enrichment on Modified Semisolid Rappaport-Vassiliadis medium for rapid Salmonella detection from food-processing environmental samples. Artificially contaminated chocolate scrapings and naturally contaminated cocoa bean dust samples were used in the study. Pre-enrichment was performed in buffered peptone water with added casein and malachite green oxalate. Motility enrichment was compared with a conventional cultural procedure using Rappaport-Vassiliadis broth and selenite cystine broth as selective enrichment. The productivity of motility enrichment was 93.5% compared to a productivity of the cultural procedure of 92%. Statistical analysis showed that there was no significant difference between the two procedures. Modified Semisolid Rappaport-Vassiliadis medium is a sensitive and simple diagnostic tool for the microbiological safety evaluation of food-processing environments.
