A rapid multicolor Western blot.

Abstract

A multicolor Western blotting technique was developed, by which different kinds or different subtypes of interferon were identified with different colors on a single Western blot. This was achieved by sequentially applying different sets of probing antibodies, enzyme-conjugated developing antibodies and enzyme substrates to detect each of the two or more types of interferon on the Western blot. An improved and much faster method of obtaining the same result by the simultaneous application of more than one kind of probing antibodies, the simultaneous application of a mixture of different enzyme-conjugated developing antibodies followed by successive application of different substrates was also described. In addition, a combination of the sequential and simultaneous techniques was used to produce a triple color Western blot.

Membrane filtration - atomic absorption spectrometry combination for copper, cobalt, cadmium, lead and chromium in environmental samples.

Cellulose nitrate membrane filter was used for the preconcentration-separation of Cu, Co, Cd, Pb and Cr ions. The analyte ions were collected on the membrane filter by the aid of carmine. Then membrane filter was dissolved by using nitric acid. The levels of the analytes in the final solutions were determined by flame atomic absorption spectrometry (FAAS). The analytical parameters including pH, amounts of carmine, sample volumes etc. have been optimized. No influences have been observed from the matrix ions. The detection limits for analytes were in the range of 0.08 microg/l-0.93 microg/l. The validation of the procedure was checked by the analysis of standard reference sediment (GBW 07309). The present method has been successfully applied for the FAAS determinations of analyte ions in real samples including black tea and magnesium salts.

An approach for precise three-dimensional modeling of the human inner ear.

For further morphological and physiological research, it is vital to establish precise three-dimensional models of the whole inner ear including the details of the membranous components. With the system of a projector and a high-resolution digital camera, 2 complete serial unstained celloidin sections of fresh human temporal bones were digitized as high-resolution images which were then sorted, calibrated, aligned and segmented using the 3D-Doctor software. Finally, 2 precise three-dimensional models of the inner ear were generated by simple surface rendering. The contours of tiny structures such as the crista ampullaris, the macula utriculi and the macula sacculi could be observed clearly. Our study suggests that it is technically feasible to employ complete serial unstained celloidin sections for precise three-dimensional reconstruction and that this helps reduce errors and laboratory workload. Moreover, the use of a high-resolution digital camera and the autoalignment function of 3D-Doctor further increase the accuracy of the models.
