Microbial transformation studies on arteannuin B.

Abstract

The microbial transformation of the sesquiterpene lactone arreannuin B [3] using Aspergillus flavipes produced dihydroarteannuin B [4] as the main transformation product. Preparative-scale fermentation of 3 with Beauveria bassiana, on the other hand, has resulted in the production of two metabolites, 3 beta-hydroxyarteannuin B [5] and 13-hydroxy-11-epi-dihydroarteannuin B [6]. The structure of these metabolites, all of which are new compounds, was established using chemical and spectroscopic techniques. The isomeric dihydrocompound, 11-epi-dihydroarteannuin B [7] and an isomer of arteannuin B [8] were also prepared chemically. All compounds were subjected to 2D-nmr experiments and full 1H- and 13C-nmr assignments were made.

Synthesis and immunosuppressive activity of new artemisinin derivatives. Part 2: 2-[12(beta or alpha)-dihydroartemisinoxymethyl(or 1'-ethyl)]phenoxyl propionic acids and esters.

Another series of novel dihydroartemisinin derivatives were synthesized and assessed for their cytotoxicity of lymphocyte, inhibitory activity on mitogen-induced spleen lymphocyte proliferation in vitro. Some of the compounds exhibited inhibitory effects on ConA-induced T cell and LPS-induced B cell proliferation comparable to or more potent than parent artemisinin.

Biosynthesis and engineering of isoprenoid small molecules.

Isoprenoid secondary metabolites are a rich source of commercial products that have not been fully explored. At present, there are isoprenoid products used in cancer therapy, the treatment of infectious diseases, and crop protection. All isoprenoids share universal prenyl diphosphate precursors synthesized via two distinct pathways. From these universal precursors, the biosynthetic pathways to specific isoprenoids diverge resulting in a staggering array of products. Taking advantage of this diversity has been the focus of much effort in metabolic engineering heterologous hosts. In addition, the engineering of the mevalonate pathway has increased levels of the universal precursors available for heterologous production. Finally, we will describe the efforts to produce to commercial terpenoids, paclitaxel and artemisinin.

Gold(III) Salen complex-catalyzed synthesis of propargylamines via a three-component coupling reaction.

[reaction: see text] Propargylamines have been synthesized by a gold(III) salen complex-catalyzed three-component coupling reaction of aldehydes, amines, and alkynes in water in excellent yields at 40 degrees C. With chiral prolinol derivatives as the amine component, excellent diastereoselectivities (up to 99:1) have been attained. This coupling reaction has been applied to the synthesis of propargylamine-modified artemisinin derivatives with the delicate endoperoxide moieties remaining intact. Cytotoxicities with IC(50) values up to 1.1 microM against a human hepatocellular carcinoma cell line (HepG2) were exhibited by these artemisinin derivatives.
