Characterization of representative enzymes from a sulfate reducing bacterium implicated in the corrosion of steel.

This communication reports the isolation, purification and characterization of key enzymes involved in dissimilatory sulfate reduction of a sulfate reducing bacterium classified as Desulfovibrio desulfuricans subspecies desulfuricans New Jersey (NCIMB 8313) (Ddd NJ). The chosen strain, originally recovered from a corroding cast iron heat exchanger, was grown in large scale batch cultures. Physico-chemical and spectroscopic studies of the purified enzymes were carried out. These analyses revealed a high degree of similarity between proteins isolated from the DddNJ strain and the homologous proteins obtained from Desulfomicrobium baculatus Norway 4. In view of the results obtained, taxonomic reclassification of Desulfovibrio desulfuricans subspecies desulfuricans New Jersey (NCIMB 8313) into Desulfomicrobium baculatus (New Jersey) is proposed.

Metal versus polyethylene wear particles in total hip replacements. A review.

Research has recently been focused on the development of hip replacements with alternative bearing surfaces with cobalt chrome alloy, to avoid the production of polyethylene wear particles in hip replacements and polyethylene wear debris mediated bone lysis. Cobalt chrome on cobalt chrome bearing surfaces are being reevaluated. Characterization of wear particles and studies on the reaction of the body to these particles, have played an important role in the determination of the factors that cause aseptic loosening and will therefore play an important role in the comparison of metal on polyethylene and metal on metal hip prostheses. In this paper, a comparison between the different aspects of metal and polyethylene wear particles is made using data from the literature and the authors' experience. The authors conclude that techniques need to be optimized to isolate and characterize individual metal wear particles from periprosthetic tissues and they advocate the performance of in vitro studies with these in vivo generated wear particles or comparable particles.

Complexation of nicotinamide adenine dinucleotide with ferric and ferrous ions.

Motivated by the observed influence of stainless steel and ferric and ferrous ions on the behavior of the peroxidase/oxidase oscillator, the mechanism and kinetics of interaction of 1,4-dihydronicotinamide adenine dinucleotide (NADH) with iron ions in 0.1 M acetic acid/sodium acetate buffer with pH 5.1 and with the solution/stainless steel interface were extensively studied. The character of a possible mutual influence of NADH/acetate buffer solution and Type 316 stainless steel has been investigated. We also suggest the mechanism of stainless steel corrosion inhibition by NADH. It was determined that fast complexation of ferric and ferrous ions with NADH occurred with rate constant kcompl = 4.0 x 10(9) +/- 0.2 x 10(9) M-1 s-1. The composition of the product complex is [Fe-(NADH)2] for both Fe2+ and Fe3+. A previously unreported complex of ferrous ion and NADH was discovered, determined, and separately investigated. Kinetic and equilibrium constants for reactions of iron ions-NADH complexation and following redox processes of the complex decomposition were determined from spectrophotometric and electrochemical experiments.
