Ultrasonographic and pathological endometrial findings in asymptomatic postmenopausal women taking antihypertensive drugs.

OBJECTIVES: To assess whether abnormal sonographic endometrial findings are more frequent in asymptomatic postmenopausal women receiving anti-hypertensive drugs than in normotensive women. METHODS: A prospective study on 187 postmenopausal asymptomatic normotensive (group A) and 182 postmenopausal asymptomatic women receiving anti-hypertensive drugs (group B) was performed. All patients were evaluated initially by transvaginal ultrasound and endometrial sampling (Pipelle device). Those women with abnormal sonographic findings (endometrial thickness > or =5 mm or heterogeneous echogenicity) or abnormal endometrial sampling results underwent hysteroscopy and guided biopsy. RESULTS: Patients in group B had significantly higher percentage of endometrial thickness > or =5 mm (26.9 vs. 12.8%, P<0.001) and heterogeneous endometria (23.1 vs. 12.8%, P<0.001) than in group A. Patients in group B had a significantly higher incidence of endometrial polyps (17.6 vs. 9.6%, P<0.001) too. These findings were not associated with the type of anti-hypertensive drug taken and were independent of the body mass index. CONCLUSIONS: Postmenopausal hypertensive asymptomatic women receiving pharmacological anti-hypertensive therapy show a higher percentage of abnormal sonographic endometrial findings than normotensive women.

A study of the determination of the hypertensive drug captopril by square wave cathodic adsorptive stripping voltammetry.

In this work, the determination of captopril (CPL) was studied by square wave cathodic adsorptive stripping voltammetry (SWCAdSV) on a hanging mercury drop electrode (HMDE). CPL was adsorptively preconcentrated on the mercury surface as a sparingly soluble mercury salt under stirring of the solution and then the accumulated species was reduced by a cathodic square wave voltammetric scan. The reduction current was related to the CPL concentration in the sample. The chemical and instrumental parameters affecting the response were investigated and optimized for the CPL determination. The calibration curve was linear from 0.5 to 180 microg l(-1) of CPL (depending on the preconcentration time), the limit of detection at a S/N ratio of 3 was 0.5 microg l(-1) with 300 s of preconcentration and the relative standard deviation was 3.2% at the 20 microg l(-1) level (with 120 s of preconcentration, n=8). The method was applied to the determination of CPL in two pharmaceutical formulations with recoveries of 97.9 and 98.8%. Finally, the potential for applying the proposed method to the determination of CPL in biological media is briefly discussed.

Development and validation of an UV derivative spectrophotometric determination of Losartan potassium in tablets.

Development and validation of an analytical UV derivative spectrophotometric method to quantify Losartan potassium used as a single active principle in pharmaceutical forms were done. Pharmacopeias have not yet provided an official method for its quantification. A study was carried out of all the parameters established by USP XXIV to validate an analytical method for a solid pharmaceutical form, i.e. linearity, range, accuracy, precision and specificity. All these parameters were found in accordance with the acceptance criteria of Comité de Guías Oficiales de Validación de la Dirección General de Control de Insumos para la Salud de México. Based on the spectrophotometric characteristics of Losartan potassium, a signal at 234 nm of the first derivative spectrum (1D234) was found adequate for quantification. The linearity between signal 1D234 and concentration of Losartan potassium in the range of 4.00-6.00 mg l(-1) in aqueous solutions presents a square correlation coefficient (r2) of 0.9938. The mean recovery percentage was 100.7+/-1.1% and the precision expressed as relative standard deviation (R.S.D.) 0.88%. In addition, the proposed method is simple, easy to apply, low-cost, does not use polluting reagents and requires relatively inexpensive instruments. Then, it is a good alternative to existing methods for determining Losartan potassium in tablets provided that the pharmaceutical dosage form does not contain hydrochlorothiazide as second drug.
