Circulating 5-hydroxytryptamine concentrations in preterm newborns.

Abstract

Alterations in circulating 5-hydroxytryptamine (5-HT) concentrations play a role in the pathophysiology of respiratory failure in adults. We undertook a study to develop a micromethod and measure circulating free 5-HT concentrations in preterm newborns with and without respiratory distress. Forty-six samples of platelet-poor plasma were obtained from 29 preterm newborns with varying degrees of respiratory distress. Samples were taken on days 2-3 and 6-7 of life. For measuring 5-HT concentrations we used a precolumn sample enhancement technique followed by ion exchange HPLC with electrochemical detection. The assay allowed detection of extremely small (50 pg) amounts of 5-HT from small (0.2 ml) amounts of blood. The mean 5-HT concentration on days 2-3 was 1.77 +/-0.74 ng/ml (mean +/- 95% confidence limits) and on days 6-7 was 0.69 +/- 0.23 ng/ml. This represented a significant fall in 5-HT concentrations (P = 0.01). All of 16 paired serial samples fell with time (P = 0.006). We conclude that platelet-poor plasma 5-HT concentrations in premature newborns are low, that there is a significant decline in these values over the first week of life, and that, in contrast with adults, the presence of respiratory failure is not associated with increased free 5-HT concentrations. The low 5-HT concentrations seen in newborns may reflect the ability to increase pulmonary uptake.

Determination of gibberellins in fermentation broth produced by Fusarium verticilliodes MTCC 156 by high-performance liquid chromatography tandem mass spectrometry.

A method for the detection of gibberellins produced by Fusarium verticilliodes is described using HPLCMS/MS (HPLC tandem MS). A Hypersil (5 microm) octadecylsilane column with methanol/water as eluent in the ratio 3:1 at a flow rate of 0.5 ml/min was used. In the HPLCMS, GA(3) (gibberellic acid; m / z 346.3) eluted at retention time tr=3.08 min, with the corresponding mass chromatogram having peaks at m / z 346.7 and 328.8 corresponding to the M+ and M+-H2O ions respectively. The ethyl acetate extract from the broth, subjected to HPLCMS analysis under similar conditions, showed a constituent with tr=2.13 min, the mass chromatogram of which exhibited peaks at m / z 348.9 and 331.9 corresponding to the MH+ and MH+-H2O ions respectively. Comparison of the MS and MS/MS results (direct infusion) of an authentic sample of GA3 and the ethyl acetate extract from the broth revealed the formation of reduced GA3 in the broth. The present study, utilizing HPLCMS/MS, describes an improved methodology for the unambiguous determination and estimation of gibberellins from fermentation broth.

Gelatin microparticles containing propolis obtained by spray-drying technique: preparation and characterization.

Gelatin microparticles containing propolis extractive solution (PES) were prepared by spray-drying technique. The optimization of the spray-drying operating conditions and the proportions of gelatin and mannitol were investigated. Regular particle morphology was obtained when mannitol was used, whereas mannitol absence produced a substantial number of coalesced and agglomerated microparticles. Microparticles had a mean diameter of 2.70 microm without mannitol and 2.50 microm with mannitol. The entrapment efficiency for propolis of the microparticles was upto 41% without mannitol and 39% with mannitol. The microencapsulation by spray-drying technique maintained the activity of propolis against Staphylococcus aureus. These gelatin microparticles containing propolis would be useful for developing intermediary or eventual propolis dosage form without the PES' strong and unpleasant taste, aromatic odour, and presence of ethanol.

Measurement of free fatty acids in cerebrospinal fluid from patients with hemorrhagic and ischemic stroke.

Free fatty acid (FFA) concentrations in cerebrospinal fluid (CSF) from patients with ischemic and hemorrhagic stroke (n=25) and in contemporary controls (n=73) were examined using HPLC. Concentrations of CSF FFAs from ischemic and hemorrhagic stroke patients obtained within 48 h of the insult were significantly greater than in control patients. Higher concentrations of polyunsaturated fatty acids (PUFAs) in CSF obtained within 48 h of insult were associated with significantly lower (P<0.05) admission Glasgow Coma Scale scores and worse outcome at the time of hospital discharge, using the Glasgow Outcome Scale (P<0.01).
