Fibronectin during thyroid hormone replacement therapy.

Abstract

Hemostasis may be affected by thyroid function in various ways. We studied plasma concentration of Fibronectin in 13 untreated patients after total thyroidectomy due to thyroid carcinoma. Fibronectin levels were significantly decreased compared to an equal numbered group of healthy volunteers (p 0.0005). The same patients were studied again after an oral thyroid replacement of 200 micrograms L-thyroxine/d over at least six weeks. Fibronectin had increased significantly, compared to untreated patients and controls resp. Furthermore, a significant positive correlation was found between plasma concentrations of Fibronectin and total T4 (r = 0.92; y = 6.617 + 0.52x). In contrast Factor V activity was low in untreated patients but normal during high dose replacement therapy. No correlation was found between Factor V and thyroid hormone concentrations. We discuss the relation between fibronectin plasma levels and thyroid hormones.

[Inhibitory effect of ulinastatin on the alpha-thrombin activation of factors V and VIII].

Ulinastatin is a remedy of urine serinprotease inhibitors and also it inhibits enzymatic activities of several blood coagulation factors. The action of ulinastatin on alpha-thrombin shows noncompetitive inhibition with the formation of enzyme-inhibitor complex. In this study, the effects of ulinastatin on the alpha-thrombin activation of factors V and VIII are observed by 5-20% gradient gel SDS PAGE with or without Western blotting method. The addition of ulinastatin to the mixture of factor V and alpha-thrombin inhibits the production of active peptides proceeded in the alpha-thrombin activation process of factor V. Furthermore, using anti-factor VIII 43KDa peptide monoclonal antibody with Western blotting method, the addition of ulinastatin to the mixture of alpha-thrombin and factor VIII indicates to inhibit the release of 43KDa peptide from factor VIII in the process of factor VIII activation induced by alpha-thrombin.

Factor V

Factor V: a prototype pro-cofactor for vitamin K-dependent enzyme complexes in blood clotting.

The relative abundance of factor V, factor X and prothrombin has enabled detailed analyses of the prothrombinase complex. Determination of the primary structure for factor V has provided the basis for examination of structure-function relationships. The imminent in vitro expression of recombinant factor V will provide the opportunity for site-specific mutagenesis and a verification of these structure-function relationships. A comparison of the physical properties and primary structures for factors V and VIII has revealed extensive similarities in these two cofactor proteins. This observation indicates that a direct application of the technology developed for the analysis of prothrombinase will lead to an equal understanding of the factor Xase complex. Whether similar relationships exist for other blood coagulation enzyme complexes remains to be determined.

Factor V
