Pseudaquabacterium prasiolae sp. nov., Isolated from the Freshwater Green Alga Prasiola japonica, and Rubrivivax soli sp. nov., Isolated from Soil, with Reclassification of Aquabacterium humicola as Pseudaquabacterium humicola comb. nov.

Abstract

Two Gram-stain-negative, catalase- and oxidase-positive, strictly aerobic, non-flagellated rod-shaped bacteria, designated OR-4T and RP6-9T, were isolated from the freshwater green alga Prasiola japonica and soil in Republic of Korea, respectively. Strain OR-4T exhibited gliding motility, whereas strain RP6-9T lacked gliding motility. Strain OR-4T grew at 10-30 °C, pH 6.0-9.0, and 0-1.5% (w/v) NaCl, while strain RP6-9T grew at 20-35 °C, pH 6.0-9.0, and 0-1.0% (w/v) NaCl. Both strains contained ubiquinone-8 as the sole respiratory quinone and phosphatidylethanolamine, phosphatidylglycerol, and diphosphatidylglycerol as major polar lipids; strain OR-4T additionally possessed an unidentified phospholipid and an unidentified polar lipid. The predominant fatty acids of OR-4T were C12:0, C16:0, summed feature 3 (C16:1ω6c and/or C16:1ω7c), and summed feature 8 (C18:1ω7c and/or C18:1ω6c), whereas RP6-9T contained C12:0, C16:0, and summed feature 3 as major components. The genomic DNA G + C content of both strains was 71.0 mol%. Whole-genome-based phylogenomic analyses placed OR-4T and RP6-9T within the genera Pseudaquabacterium and Rubrivivax, respectively, forming distinct lineages. Comparative analyses of average nucleotide identity, digital DNA-DNA hybridization, and average amino acid identity further supported their assignment to these genera while confirming their separation from previously described species. Based on combined phenotypic, chemotaxonomic, and genomic evidence, strains OR-4T and RP6-9T represent novel species, for which the names Pseudaquabacterium prasiolae sp. nov. (type strain OR-4T =KACC 22752T =NBRC 116024T) and Rubrivivax soli sp. nov. (type strain RP6-9T =KACC 24055T =DSM 119932T) are proposed. Phylogenomic analyses also support the reclassification of Aquabacterium humicola as Pseudaquabacterium humicola comb. nov. (type strain RJY3T =KCTC 92105T =NBRC 115831T).

Curtobacterium uvaldense sp. nov., Falsirhodobacter allii sp. nov., Pseudomonas texasensis sp. nov., Three Novel Species Isolated from Onions in Texas, USA.

Bacterial strains 20TX0008T and 20TX0035T were isolated from the symptomatic onion foliage in 2020, and a bacterial strain 21TX0197T was isolated from the symptomatic onion bulb in 2021. Pathogenicity assays confirmed the two strains isolated from foliage to be non-pathogenic, while the strain 21TX0197T was found to be pathogenic. Phylogenetic analysis using the 16&#xa0;S rRNA gene revealed the closest resemblance of 20TX0008T, 20TX0035T, and 21TX0197T to the genera Curtobacterium, Falsirhodobacter, and Pseudomonas, respectively. For the respective genus, strains 20TX0008T showed the greatest 16&#xa0;S rRNA sequence similarity with Curtobacterium caesinilyticum GDMCC 1.2667T (99.7%), 20TX0035T with Falsirhodobacter deserti W402T (99.1%), and 21TX0197T with Pseudomonas uvaldensis 20TX0172T (99.4%). In the phylogenetic trees based on whole genome sequences, all three strains formed a distinct, well-supported branch that separated them from their closely related species within the same genus. The genomic similarity, based on average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH), revealed that the novel strains had values below the species delineation thresholds (ANI&#x2009;<&#x2009;95% and dDDH&#x2009;<&#x2009;70%), confirming the novelty of all these strains. Based on genomic and phenotypic characteristics, we propose three novel species, Curtobacterium uvaldense sp. nov. (type strain 20TX0008T =LMG 33207T =NCIMB 15480T), Falsirhodobacter allii sp. nov. (type strain 20TX0035T =LMG 33208T =NCIMB 15477T), and Pseudomonas texasensis sp. nov. (type strain 21TX0197T =LMG 33210T =NCIMB 15479T).

Molecular characterization of a new emaravirus infecting Clerodendrum thomsoniae plants.

High-throughput sequencing revealed a new emaravirus named clerodendrum yellow blotch virus (ClYBV), infecting Clerodendrum thomsoniae plants in S&#xe3;o Paulo State, Brazil. Four genomic RNA segments (RNA1, RNA3, RNA4, and RNA5) were recovered, whereas RNA2 remained undetected. Phylogenetic analysis and pairwise comparisons based on amino acid sequences showed ClYBV is most closely related to redbud yellow ringspot-associated virus (RYRaV, Emaravirus cercidis). No ClYBV particles were observed by transmission electron microscopy in leaf tissues from infected plants. These results support the identification of ClYBV as a new Emaravirus member, though the undetectable RNA2 suggests the isolate may be defective, lacking this segment and unable to produce virus particles. The binomial species name "Emaravirus clerodendri" is proposed.

Molecular characterization of a novel partitivirus harboring an additional third dsRNA segment from Trichoderma harzianum.

We report the complete genome sequence of a novel partitivirus identified from Trichoderma harzianum NFCF092 strain, designated Trichoderma harzianum partitivirus 4 (ThPV4). Unlike canonical members of the family Partitiviridae, which possess a bipartite genome consisting of two double-stranded RNA (dsRNA) segments encoding an RNA-dependent RNA polymerase (RdRP) and a capsid protein (CP), ThPV4 harbors a third dsRNA segment encoding a protein of unknown function. The complete genome consists of dsRNA1 (1,950&#xa0;bp; encoding the RdRP), dsRNA2 (1,772&#xa0;bp; encoding the CP), and dsRNA3 (1,629&#xa0;bp; encoding a protein with unknown function). Sequence analysis shows that each segment possesses a single open reading frame (ORF). The deduced amino acid sequence of the RdRP shows the highest similarity (90.5% identity) to that of Trichoderma gamsii alphapartitivirus 1. Phylogenetic analyses based on the RdRP indicate that ThPV4 clusters within the genus Alphapartitivirus of the family Partitiviridae. To our knowledge. ThPV4 is the first member of the genus Alphapartitivirus identified from T. harzianum to possess an additional, conserved third dsRNA segment.
