Electrophoretically uniform fluorescent dyes for automated DNA sequencing.

A class of dyes, BODIPY fluorophores, has been identified for automated DNA sequencing that has improved spectral characteristics compared with conventional fluorescein and rhodamine dyes. Single and double BODIPY dye primers were characterized in commercially available DNA sequencers and showed uniform electrophoretic mobilities and high fluorescence intensities. The improved physical properties of BODIPY dye primers were demonstrated by direct base-calling from the unprocessed fluorescent signals and improved heterozygote analyses of mixed-base populations. The high sensitivity of BODIPY dye primers requires at least 33 percent less reagent consumed per reaction than conventional dye primers, which should affect the costs of large genome-sequencing efforts.

Automated solid-phase extraction coupled to gas chromatography with electron-capture detection: a combination of extraction and clean-up of pyrethroids in the analysis of surface water.

The combination of automated solid-phase extraction (SPE) and large-volume introduction gas chromatography electron-capture detection (GC-ECD) is used for the determination of synthetic pyrethroids in surface and drinking water. The selectivity that is needed for the use of GC-ECD of environmental samples is achieved by addition of a polarity modifier to the sample. In this study, sufficient methanol was added to 19 ml of sample to obtain a level of 30% of modifier prior to SPE. The cartridge is washed with one additional ml of 30% methanol in water. After drying, the cartridge is eluted with toluene. GC-ECD analysis is performed by on-column introduction of 100 microliters of the toluene extract. The whole procedure was automated by using an automated solid-phase extraction apparatus. Limits of detection (LODs) for the pyrethroids in surface water are in the range of 1 to 8 ng/1 (signal-to-noise ratio = 3); even lower limits of detection are possible for drinking water.

An automated preconcentration-derivatization system for the determination of cocaine and its metabolites in urine and illicit cocaine samples by gas chromatography/mass spectrometry.

Although gas chromatography/mass spectrometry (GC/MS) is the most powerful technique for analysing cocaine and its hydrolysis products (benzoylecgonine, ecgonine methyl ester and ecgonine) in urine, the sample pretreatment required is rather laborious and involves a large number of steps. This paper reports a continuous method for the separation (based on the retention properties of cocaine and its major metabolites on a reversed phase C18 column), preconcentration (evaporation under nitrogen) and derivatization (collection of the residue in a few microlitres of derivatizing reagent) of cocaine and its metabolites in urine samples for subsequent analysis by GC/MS. Calibration graphs are linear from 5 to 3000 ng/mL; the detection limits are 1 ng/mL (cocaine and ecgonine methyl ester), 20 ng/mL (benzoylecgonine) and 200 ng/mL (ecgonine). The method was applied to illicit cocaine samples and to urine from a drug addict.
