The authors found that, by continuously bubbling pure nitrogen within a dilute Hb solution, the latter releases its oxygen with an intensity which is quite proportional to the duration of bubbling. Such a procedure can be employed instead of the common tonometric technique, to measure functional properties of Hb. In fact, not only dissociation curves can be obtained, but also a correct evaluation of the affinity changes upon pH variations (Bohr effect). The results obtained by this technique are not far from those recently reported by using tonometry, although the method is less precise and still not suitable for routine purposes. Nevertheless, it can be substantially improved.
HemoCue, an instrument for measuring B-haemoglobin, is described as correcting for the turbidity caused by high white cell counts or hyperlipaemia. It was evaluated for its ability to correct haemoglobin measurement for hyperlipaemia against a previously published method and found to perform adequately, but with a small constant positive bias.
The influence of lipid and high concentrations of leukocytes on the haemoglobin determination by six different instruments was examined. The increase in the determination of haemoglobin concentration with elevated amounts of lipid in the samples was greatest with the Coulter Counter S plus III and the Technicon H 1 followed by the LK 540 and the Technicon H 6000. There was no effect of lipid on the haemoglobin determination using the Reflotron. High concentrations of leukocytes increased the haemoglobin determination most by Coulter Counter S plus III and Hemocap. There were no changes in the haemoglobin determinations by the LK 540 and the Technicon H 6000 when the samples contained increased amounts of leukocytes.
