Dielectrophoretic dynamic light-scattering (DDLS) spectroscopy is presented. DDLS identifies macromolecules based on their dielectric, or polarizability, properties. DDLS measurements are carried out in an oscillating, nonuniform electric field. The field induces macromolecules to undergo dielectrophoretic motion, which is detected by the modulation in the dynamic light-scattering autocorrelation function. The DDLS experimental setup, data analysis, and data on latex particles and yeast cells are presented.
The literature on polymer latex film formation has grown enormously in recent times--driven by the need to find alternatives for solvent-based systems with their adverse environmental impacts. Although greater insight has been shown by the use of modern instrumental techniques such as small angle neutron scattering, direct non-radiative energy transfer and atomic force microscopy, the actual mechanisms involved in deforming spherical particles into void-free films are still the subject of controversy and debate. Surfactant-free homopolymer model colloid latices, favoured in academic studies, together with latices containing surfactants whose redistribution can influence film properties, and the more complex copolymer, blended, core-shell and pigmented systems needed to satisfy a full range of film properties are all considered.
A major constraint in the enzymatic assays for determination of linamarin in cassava is the preparation of purified linamarase. Cassava latex, which exhibits high linamarase activity, was tried as an alternate source of the enzyme. Enzyme yield from latex was compared with that from rind and leaf. Preparations from latex had significantly higher linamarase activity (approximately 300-fold) compared to leaf and rind. The purification of the enzyme was easier since homogenization of large quantities of tissues could be avoided. A 1 g amount of latex could yield enough enzyme for >3000 assays.
