Solid-state magic-angle spinning 31P-NMR studies of native casein micelles.

Solid-state magic-angle spinning 31P-NMR spectroscopy was used to characterize the structure and composition of native casein micelles. The features of the magic-angle spinning 31P-NMR spectra, including overlapping resonances from mobile/immobile phosphorylated serine residues and inorganic calcium phosphates, have been determined using different experimental techniques and assigned by comparison with spectra of the presumed constituents within the casein micelle. Comparison with 31P-NMR spectra of alpha s1-, alpha s2-, and beta-caseins in dissolved and freeze-dried forms demonstrated that a major fraction of the phosphoserines in these proteins was in an immobilized state within the micelle. Likewise, from 31P-NMR spectra of the C-terminal part of kappa-casein, it was shown that this region of the micelle has a considerable conformational mobility. Finally, magic-angle spinning 31P-NMR spectra for a series of inorganic calcium phosphates and mineralized bone tissue revealed that the micellar inorganic calcium phosphates exhibit structural similarities to hydroxyapatite and hence resemble mineralized bone tissue.

A single phosphotyrosine residue of the prolactin receptor is responsible for activation of gene transcription.

Members of the cytokine/growth hormone/prolactin (PRL) receptor superfamily are associated with cytoplasmic tyrosine kinases of the Jak family. For the PRL receptor (PRLR), after PRL stimulation, both the kinase Jak2 and the receptor undergo tyrosine phosphorylation. To assess the role of tyrosine phosphorylation of the PRLR in signal transduction, several mutant forms of the PRLR in which various tyrosine residues were changed to phenylalanine were constructed and their functional properties were investigated. We identified a single tyrosine residue located at the C terminus of the PRLR to be necessary for in vivo activation of PRL-responsive gene transcription. This clearly indicates that a phosphotyrosine residue in the cytoplasmic domain of a member of the cytokine/growth hormone/PRL receptor superfamily is directly involved in signal transduction.

Selective determination of phosphopeptide beta-CN(1-25) in a beta-casein digest by adding iron: characterization by liquid chromatography with on-line electrospray-ionization mass spectrometric detection.

A beta-casein tryptic digest has been analysed by reversed-phase high-performance liquid chromatography (RP-HPLC) with on-line electrospray-ionization mass spectrometry (ESI-MS). Analyses of peptides were carried out before and after addition of iron(II) to the peptides in solution. In both cases, the majority of peptides were identified by the determination of molecular masses by ESI-MS and by prior knowledge of the amino acid sequence of beta-casein, and thus of its corresponding tryptic peptides. In the presence of iron(II), only phosphopeptide beta-CN(1-25) was able to bind iron to form different complexes that have increased retention times on the RP-HPLC column and that also absorbed at 280 nm. The method presented here appears to be selective for peptides containing phosphoseryl cluster(s).
