[Palatal myoclonus associated with a decrease in 5-hydroxy-indole acetic acid in cerebrospinal fluid and responding to clonazepam].

Abstract

A patient representing with palatal myoclonus was diagnosed as suffering from possible multiple sclerosis. A reduction of cerebrospinal 5-hydroxy-indole-acetic acid suggested a decreased central serotonergic turnover. A therapeutic trial with clonazepam, a serotonergic agonist, improved the myoclonus symptomatically. The possible involvement of serotonergic inhibitory systems in the expression of palatal myoclonus is discussed.

Molecular modelling of asperlicin derived cholecystokinin A receptor antagonists.

The C3-substituted benzodiazepines derived from asperlicin, e.g. devazepide (L-364,718, MK-329), constitute the most potent class of cholecystokinin A-type (CCKA) receptor antagonists. In order to gain insight into the prerequisites for binding, we examined the conformational properties of both potent and weak representatives of this class with computer assisted molecular modelling (CAMM) techniques. The CAMM results indicate that the binding site for the C3-substituents is a planar slot on the CCKA receptor surface and, in addition, allow the proposal of a model which describes the relative binding mode of the less potent R isomers versus that of the S isomers. The latter model illustrates the unique spatial properties of the benzodiazepine moiety, which we suggest functions primarily as an invertible core which assures an optimal arrangement of attached substituents.

Application of preparative liquid chromatography to the isolation of enantiomers of a benzodiazepinone derivative.

The isolation of milligram amounts of the enantiomers of a benzodiazepinone derivative was performed on an analytical cellulose tribenzoate-based column by multiple repetitive injections. An enantiomeric purity greater than 98% was required. First, an analytical method was developed to maximize the resolution by adjusting the mobile phase composition, flow-rate and most importantly the column temperature. Then the preparative separation was optimized by adjusting the sample size and detecting the sample where its UV absorbance was low. The locations of the cut points were determined by use of detector response levels. The method development, preparative separations and analytical assays of the fractions obtained were all performed on analytical columns.

Determination of L-365,260, a new cholecystokinin receptor (CCK-B) antagonist, in plasma by liquid chromatography/atmospheric pressure chemical ionization mass spectrometry.

L-365,260 is a novel cholecystokinin receptor antagonist. A sensitive and specific liquid chromatographic/mass spectrometric assay has been developed for the determination of the drug in plasma using the CD3-labeled species as the internal standard. Plasma extracts were separated on a 3 cm C18 reverse-phase high-performance liquid chromatography column. The column eluate passed, by means of a heated nebulizer interface, into a corona discharge atmospheric chemical ionization source. The mass spectrometer was operated in the positive ion tandem mass spectrometric mode. The method has sufficient sensitivity, specificity, precision, accuracy and selectivity for the determination of drug concentrations in clinical samples. The chromatographic run time is less than 2 min.

Benzodiazepinones
