Multivariate optimization approach for chiral resolution of drugs using human serum albumin in affinity electrokinetic chromatography-partial filling technique.

The enantiomeric resolution of chiral compounds using HSA by means of affinity EKC (AEKC)-partial filling technique is the result of a delicate balance between different experimental variables such as protein concentration, running pH (background electrophoretic buffer, protein and compound solutions) and protein solution plug length. In this paper multivariate optimization approaches for chiral separation of four basic drugs (alprenolol, oxprenolol, promethazine and propranolol) using HSA as chiral selector in AEKC-partial filling technique are studied. The experimental conditions to achieve maximum resolution are optimized using the Box-Behnken experimental design. Partial least squares and pareto charts are used to analyse the main effects on the resolution. The experimental resolutions observed for all compounds studied in optimum conditions agree with the estimated values based on response surface models. The results obtained show that the range of experimental conditions that provided enantioresolution narrows as hydrophobicity of analytes decreases. This fact can be explained by assuming that hydrophobicity controls the interaction of basic compounds with HSA.

Determination of dissociation constants by competitive binding in partial filling capillary electrophoresis.

The determination of dissociation constands (K(d)) by competitive ligand binding in partial filling capillary electrophoresis is demonstrated. Two different strategies were applied, one of which only uses a single reporter ligand and a more elaborated one which suppresses systemic disturbances by using a racemic mixture as reporter. The dissociation constants obtained by both alternatives were virtually identical and in good agreement with those previously reported.

Chiral separation of nanomole amounts of alprenolol with cITP/NMR.

On-line cITP-NMR with chiral selectors separates and concentrates analytes and identifies host-guest interactions of analytes with selectivity enhancers in the electrolyte. An NMR microcoil designed for a 200 microm i.d. capillary creates a high-mass-sensitivity 30 nL NMR cell and is used as an on-line detector for cITP. Using a mixture of 2 nmol racemic alprenolol in acetate buffer with alpha-cyclodextrin and sulfated beta-cyclodextrin at pD 6.0, cITP-NMR successfully separates and concentrates both R- and S-alprenolol. The concentration enhancement for the R isomer is 224-fold and for the S isomer is 200-fold. The estimated concentration at peak maximum for R-alprenolol is approximately 28 mmol L(-1) and a slightly lower concentration, 25 mmol L(-1) is achieved for S-alprenolol. These concentrations convert to placing 76% of the injected S-alprenolol and 84% of the R-alprenolol into the 30 nL detection cell at peak maximum. With on-flow cITP-NMR, intermolecular interactions between the cyclodextrins and the alprenolol are observed in the NMR spectra. Aromatic and methyl moieties of R- and S-alprenolol are identified as two important sites that bind with these particular cyclodextrins.
