A classification of replicators and lambda-calculus models of biological organization.

W. Fontana & L.W. Buss (Proc. Natn. Acad. Sci. U.S.A. 91, 757 (1994) and Bull. math. Biol. 56, 1 (1994) have put forward a scheme for a theory of biological organization based on the lambda-calculus. Their key innovation was to represent, with the aid of this calculus, a certain minimal chemistry. Although this idea is very promising, their concrete formulation could be improved if suggestions for the following items were incorporated: (i) a better coding of chemical reactions; (ii) a reinterpretation of the evolutionary behaviour of autocatalytic chemical networks; (iii) a better appreciation of morphological and genetic factors; (iv) a more complete embedding of the theory into the background of relevant earlier contributions. Confusion can be stopped by the application of a proper classification of replicators (important categories being: processive and modular, limited and unlimited hereditary replicators). Suggestions to facilitate improvement are made explicit in this paper. The most challenging task would be to model the transition from processive, limited hereditary replicators to modular replicators with limited and unlimited heredity.

Microbiological quality in Finnish public swimming pools and whirlpools with special reference to free living amoebae: a risk factor for contact lens wearers?

To assess the possible risk of microbial keratitis associated with swimming or bathing in public pools, the microbiological quality as well as the presence of free living amoebae in 16 halogenated swimming pools and whirlpools, located in Helsinki, Finland, was determined. Five additional whirlpools situated in the ferries cruising from Finland to Sweden were included in the study. Other parameters investigated were the total bacterial count, identification of Pseudomonas aeruginosa and Staphylococcus aureus, measurement of free residual and combined chlorine, potassium permanganate index, urine, pH, and turbidity. Amoebae were detected in 41% of the pool water samples studied. Seven of 11 whirlpools and four of 10 swimming pools were shown to contain amoebae. An Acanthamoeba species was isolated from only one outdoor swimming pool; the other amoebae belonged to the genera Vexillifera, Flabellula, Hartmannella, and Rugipes. Although not a single verified case of Acanthamoeba keratitis has been found in Finland, the findings show that there is a theoretical risk of amoebic and bacterial keratitis associated with swimming or bathing in properly cleaned public pools. Consequently, we do not recommend swimming or bathing with contact lenses.

Intercellular guanosine-5'-0-(3-thiotriphosphate) blocks chemotactic motility of Dictyostelium discoideum amoebae.

Starving amoebae of the cellular slime mold Dictyostelium discoideum react chemotactically towards the attractant cAMP. In this study, the effect of nonhydrolyzable analogs of GTP and GDP on the chemotactic behavior was analyzed with light microscopic techniques. Guanosine-5'-0-(2-thiotriphosphate) (GTP gamma S) or guanosine-5'-0-(2-thiodiphosphate) (GDP beta S) was scrape-loaded into the cytoplasm of cells, together with a fluorescent marker. Stimulation with a cAMP-filled glass capillary revealed a reduced capacity of loaded cells to migrate towards the capillary tip. Most cells still protruded filopods in the direction of the capillary tip, but full extension of pseudopods was inhibited in a dose-dependent and reversible manner. This indicates that in the presence of the analogs, chemotactic sensing still occurs, and that a more distal step of the cascade of events leading to the formation of the pseudopod is impaired. In cells loaded with the analogs together with the calcium indicator fura-2, stimulation with 10 microM cAMP led to a transient change in the intracellular free calcium concentration ([Ca2+]i), which was detectable in 28% of the cells. Furthermore, large vacuoles were found containing high amounts of calcium. On the other hand, clamping of [Ca2+]i at low levels with 1,2-bis(2-aminophenoxy) ethane N,N,N',N'-tetraacetic acid (BAPTA) also inhibited motility, with neither filopods nor pseudopods formed. The data suggest that chemotactic migratory activity involves GTP-dependent processes that participate in the regulation of the Ca2+ homeostasis of the cell and in the regulation of membrane traffic that contributes to the directed locomotion.
