Defining erythrocyte internal labeling by phosphorylation.

Abstract

When erythrocytes are incubated with 32Pi, incorporation of label into phosphoproteins is a gradual process, increasing for at least 2 hours. Membrane phospholipids also are labeled. Exogenous protein kinase substrates are unlabeled in these incubations. This suggests that labeling by 32Pi occurs into polypeptides inside the erythrocytes. When erythrocytes are incubated with [gamma-32P]ATP and active protein kinase, membrane polypeptides are not labeled. Only exogenously added protein kinase substrates and the regulatory subunit of protein kinase (and its contaminants) are labeled. This suggests that labeling from [gamma-32P]ATP and active protein kinase occurs in the compartment outside the erythrocytes. Apyrase (EC 3.6.1.5) eliminates such labeling, demonstrating that it was occurring in the compartment external to the erythrocytes. However, in incubations of cells with 32Pi, apyrase has no effect on the incorporation into membrane polypeptides and phospholipids, demonstrating that this labeling occurs on the inside of the membrane. Thus, additions of apyrase to intact particles incubated with protein kinase substrates and 32Pi provides a method for identifying internally exposed polypeptides in the plasma membranes of a variety of systems.

The history of pyrosequencing.

One late afternoon in the beginning of January 1986, bicycling from the lab over the hill to the small village of Fullbourn, the idea for an alternative DNA sequencing technique came to my mind. The basic concept was to follow the activity of DNA polymerase during nucleotide incorporation into a DNA strand by analyzing the pyrophosphate released during the process. Today, the technique is used in multidisciplinary fields in academic, clinical, and industrial settings all over the word. The technique can be used for both single-base sequencing and whole-genome sequencing, depending on the format used. In this chapter I will give my personal account of the development of Pyrosequencing--beginning on a winter day in 1986, when I first envisioned the method--until today, nearly 20 yr later. I will conclude with future prospects for the method.

Association kinetics of wild- and mutant-type Ynd1p in relation to quality of grown crystals.

The intermolecular interaction and association dynamics of the Ynd1p protein were investigated using dynamic and time-resolved static light scattering measurements. The mutual diffusion coefficients of wild- and mutant-type (a single amino acid substitution) Ynd1p monomer were measured in 50 mM 2-(N-morpholino)ethanesulfonic acid (MES) buffer with 5 mM MnCl2 and 7.5% (v/v) ethylene glycol. Both translational diffusion coefficients at a zero protein concentration were (40.3 +/- 0.2) x 10(-12) m2/s at 20 degrees C and a pH of 7.0, so the hydrodynamic radius of the monomers was 4.1 +/- 0.1 nm. The measured intermolecular interaction between monomers, however, showed that the mutant-type Ynd1p had a stronger attractive force. Time-resolved static light scattering measurements showed that the association of mutant-type Ynd1p yielded a larger number of aggregates than that of wild-type Ynd1p. The time dependence of aggregate gyration radius differed between the two types. Fractal dimension analysis using scattering intensity data suggested that the inner structure of the aggregates changed from loose to rigid with time. Although this phenomenon is common for wild and mutant types, the differences in the number of aggregates yielded in the initial stages and in the intermolecular interaction affected the quality of the final grown crystals. That is, single crystals of Ynd1p grew in the mutant-type protein solution and polycrystals of Ynd1p grew in the wild-type protein solution.

Apyrase, the product of the virulence plasmid-encoded phoN2 (apy) gene of Shigella flexneri, is necessary for proper unipolar IcsA localization and for efficient intercellular spread.

The role in virulence of the Shigella flexneri ospB-phoN2 operon has been evaluated. Here we confirm that OspB is an effector and show that apyrase, the product of phoN2, may be a virulence factor, since it is required for efficient intercellular spreading. Apyrase may be important in a deoxynucleoside triphosphate-hydrolyzing activity-independent manner, suggesting that it may act as an interaction partner in the process of IcsA localization.
