Retinal adhesion.

Abstract

The development of retinal adhesion after coagulation has been investigated by the use of an experimental model of detachment in which fluid is injected into the retinal pigment interface. This adhesion, which varied from day to day in a predictable manner was low on the third day. The possible reasons for this are discussed.

The metabolic dependency of retinal adhesion in rabbit and primate.

OBJECTIVE: To determine the oxygen and glucose dependency of retinal adhesion in primate and rabbit. METHODS: Experiments were performed on Dutch rabbits and monkeys. Retinal adhesiveness was measured by peeling the retina from the retinal pigment epithelium in vitro, under different conditions of PO2 and glucose supply, and by observing the amount of adherent pigment. In vivo ischemia was produced by raising the intraocular pressure. RESULTS: Retinal adhesion failed quickly at low oxygen tensions, but a well-oxygenated solution preserved strong retinal adhesion in vitro for 15 to 20 minutes in rabbit tissue and up to 50 minutes in primate tissue. Ischemic adhesive failure was reversible on raising the PO2. Glucose levels did not affect adhesiveness. Ischemia in vivo for more than 1 minute caused rabbit retina to lose its adhesiveness. CONCLUSIONS: Retinal adhesion is continually and reversibly dependent on oxygenation, and probably on aerobic metabolism. Primate tissue is more resistant to metabolic adhesive failure than is rabbit tissue, but the metabolic requirements appear qualitatively similar.

An autologous fibrin tissue adhesive with greater bonding power.

OBJECTIVE: To describe and evaluate an autologous fibrin tissue adhesive (AFTA) that uses a combination of ethanol and freezing to precipitate fibrinogen (AFTA-E). DESIGN: The bonding power of AFTA-E was compared with that of a conventional AFTA based on ammonium sulfate precipitation of fibrinogen (AFTA-A). In this study, Silastic, porcine dermis, and human dura mater blocks were bonded together for 10 and 30 minutes with AFTA-E or AFTA-A. The blocks were then separated and the bonding power was measured. The efficacy of AFTA-E was also evaluated after a 24-hour refrigeration. SETTING: The Department of Otolaryngology Research Laboratory at the University of Illinois Eye and Ear Infirmary, Chicago, Ill. PARTICIPANTS: Blood was drawn from 86 healthy volunteers and AFTA-E and/or AFTA-A was manufactured. RESULTS: The AFTA-E was shown statistically to bond stronger than the AFTA-A. In addition, it was found that the efficacy of AFTA-E was unchanged after a 24-hour refrigeration. CONCLUSIONS: The improved AFTA, AFTA-E, is a superior alternative to the conventional AFTA-A. Furthermore, AFTA-E can be manufactured before surgery and stored, thus minimizing preparation time during surgery.

Effects of a structural change in collagen upon binding to conditioned dentin studied by 13C NMR.

To develop a better adhesive functional monomer, it is imperative to understand the adhesion mechanisms of the resin to the dentin surface. Bond strength to the decalcified dentin surface pretreated with dentin primer, an aqueous ethanol solution of N-methacryloyl glycine, increases by increasing the water content in the primer. The aqueous primer could increase the thickness of the hybrid layer. The structural change of dentinal collagen with NM alpha A was studied by using a model compound for collagen,--(1Pro-2Pro-3Gly)10--, by 13C nuclear magnetic resonance (NMR). The model compound was aggregated in ethanol but dissociated in water. It was found that NM alpha A effectively dissociated the aggregated model compound in water. The dissociation of decalcified dentin was essential to create a thick hybrid layer that could afford a higher bond strength to dentin.
