Acanthocytosis and cholesterol enrichment decrease lipid fluidity of only the outer human erythrocyte membrane leaflet.

Abstract

The structure and functions of the human erythrocyte are influenced by the composition and organization of the membrane lipids. The outer (exofacial) and inner (endofacial) leaflets of the erythrocyte membrane differ in lipid composition, and recent studies using a group of membrane-impermeant pyrene fluorophores have demonstrated that the lipid fluidity of the outer leaflet exceeds that of the inner. Using one of these probes, pyrene butyryl hydrazide linked to the tetrasaccharide stachyose (SPBH), we have compared the lipid fluidity of the outer and inner leaflets in normal human erythrocytes treated experimentally to alter membrane cholesterol content and in acanthocytes, erythrocytes of altered morphology found in individuals with the genetic disorder abetalipoproteinaemia. The results, reported here, demonstrate that hemileaflet fluidity can be altered selectively: acanthocytosis and experimental cholesterol enrichment decrease the lipid fluidity of the outer but not the inner hemileaflet.

Acanthocytosis, retinitis pigmentosa, pallidal degeneration. Report of two cases without serum lipid abnormalities.

We describe two unrelated patients with Hallervorden-Spatz, disease characterized by prominent facio-bucco-lingual dyskinesia. Acanthocytosis and retinitis pigmentosa were additional findings. Brain MRI showed the typical 'tiger's eye' image of the globus pallidus. This phenotype closely resembled the so-called HARP syndrome (hypoprebetalipoproteinemia, acanthocytosis, retinitis pigmentosa and pallidal degeneration), but extensive serum lipid study failed to demonstrate any lipoprotein abnormality. Our results raise the question whether HARP syndrome is an autonomous entity or a particular phenotype of Hallervorden-Spatz disease.

The pattern of cognitive impairments in neuroacanthocytosis. A frontosubcortical dementia.

OBJECTIVE: To identify the neuropsychological profiles of patients with neuroacanthocytosis. PATIENTS: The medical files of 19 previously reported cases of neuroacanthocytosis were reviewed. Ten of the patients involved had undergone comprehensive cognitive assessments, and their neuropsychological records formed the basis of the present study. DATA EXTRACTION: Data on discrepancies between estimated optimal and obtained measures of general intelligence and scores on focal cognitive tests of memory, language, visual perception, and frontal lobe executive skills were tabulated and interpreted. RESULTS: Eight of the patients had evidence of general intellectual deterioration. Five patients presented with memory impairment, two of whom showed visuoperceptual deficits. None of the patients showed any significant high-level language deficits. The most consistent findings across cases was evidence of impairment in frontal lobe executive skills and psychiatric morbidity. CONCLUSION: The cognitive and psychiatric features of the patients suggests that neuroacanthocytosis is a frontosubcortical type of dementia.

A novel point mutation in the McLeod syndrome gene in neuroacanthocytosis.

McLeod syndrome is an X-linked recessive disorder, characterized by neuromuscular and hematopoietic dysfunction. Two cases of McLeod syndrome were reported in a family with neuroacanthocytosis and, remarkably, 1 of them was female. Direct sequence analysis of the McLeod gene in 12 members of the family revealed a novel point mutation in exon 2 that creates a frameshift and results in premature termination of translation. There was marked skewing of X inactivation in the severely affected female.
