Abstract

The fetal adrenal cortex is the central steroid modulator in the fetal placental complex. Its anatomic structure and physiology clearly identify it as a uniquely fetal organ that undergoes atrophy to a fraction of its intrauterine size after birth and assumes markedly different steroidogenic functions on the assumption of extrauterine life. Its importance as a regulator of maturation and parturition in normal gestation is just being understood. The fetal adrenal cortex no doubt plays major roles in the pathogenesis of some of the most important clinical problems currently faced in contemporary obstetrics. Continued research that provides a more complete understanding of this organ will serve as the major base for innovative diagnostic and therapeutic advancements of the future.

The -104G nucleotide of the human CYP21 gene is important for CYP21 transcription activity and protein interaction.

CYP21 gene encodes the steroid 21-hydroxylase (P450c21) that is involved in steroidogenesis in the adrenal cortex. Mutations occurring on CYP21 which convert it to the neighboring pseudogene, CYP21P, are found in patients with congenital adrenal hyperplasia (CAH), an autosomal recessive disease. We previously reported that the CYP21P pseudogene had lower transcription activity when compared with the active CYP21 gene. The sequences determining the basal transcription activity of the human CYP21 gene are located within the 166 bp region upstream from the transcriptional start site. Within this region, only 4 nucleotides are different between the active CYP21 and the CYP21P pseudogene; they are located at the -117, -104, -101 and -94 positions from the start site of the gene. Here, we report that the CYP21 gene-specific G nucleotide sequence at the -104 position is required for its basal transcription activity driven by the native TATA box of the human CYP21 gene. When this single nucleotide is changed to the CYP21P sequence, the basal transcription activity decreases by approximately 80% in transient transfection assay. In addition, our data from gel retardation assay show that this sequence is also critical for interaction with nuclear proteins from adrenal cells. These results therefore suggest that the single G sequence of the human CYP21 gene is crucial for the expression of its basal transcription activity, and this may be influenced by the interaction with specific nuclear proteins from the adrenal gland.

Adrenocortical lipid depletion gene (ald) in AKR mice is associated with an acyl-CoA:cholesterol acyltransferase (ACAT) mutation.

ald, a recessive allele in AKR inbred mice, is responsible for complete adrenocortical lipid depletion in postpubertal males, which appears to be androgen dependent. Two recent observations (adrenocortical lipid depletion in acyl-CoA:cholesterol acyltransferase-deficient (Acact-/-) mice and the mapping of Acact to a region of chromosome 1 containing the ald locus) prompted us to ask whether adrenocortical lipid depletion in AKR mice results from an Acact mutation. Refined genetic mapping of Acact and ald was consistent with colocalization of these loci. Crossing Acact-/- with AKR (ald/ald) mice yielded postpubertal male offspring characterized by adrenocortical lipid depletion, indicating that these loci are not complementational and are therefore allelic. Immunoblotting of preputial gland homogenates demonstrated that AKR mice had an ACAT protein with a lower molecular mass than other mouse strains. Analysis of Acact cDNA from AKR mice revealed a deletion of the first coding exon and two missense mutations. Despite these coding sequence differences, the ACAT protein from the ald allele catalyzed cholesterol esterification activity at levels similar to that of wild-type protein. We speculate that the adrenocortical lipid depletion resulting from the ald mutation is caused by an altered susceptibility of the mutant protein to modifying factors, such as androgen production at puberty, in an as yet undetermined manner.

The caudal neurosecretory system and its afferent synapses in the goldfish, Carassius auratus: morphology, immunohistochemistry, and fine structure.

Morphological features of the goldfish caudal neurosecretory system were investigated by means of immunohistochemical localization of urotensins I and II (UI and UII) and electron microscopic examination of the caudal neurosecretory neurons, the urophysis, and the synaptic neuropil. The aim of the work is to provide a detailed morphological description of the afferent synapses to the caudal neurons and to analyze their distribution through the rostrocaudal extension of the caudal neurosecretory system. Three morphologically different types of neurosecretory cells have been identified according to size and shape: large, medium, and small Dahlgren cells. The three different-sized cells share similar patterns of immunoreactivity with the UI (or oCRF) and the UII antisera. Electron microscopic examination of the synaptic neuropil throughout the caudal system revealed the presence of four types of terminals: dense-cored-vesicle end bulbs (DC), spherical-vesicle end bulbs (S), flattened-vesicle end bulbs (F), and granular-vesicle end bulbs (G). The present study demonstrates that the small Dahlgren cells receive different synaptic inputs from the large and the medium neurosecretory cells. Indeed, G terminals are only found on the small Dahlgren cells, whereas DC, S, and F terminals are distributed on the large, medium, and small Dahlgren cell bodies and proximal processes.
