Persistent retrograde memory deficit after transient global amnesia.

Abstract

A 64-year-old woman suddenly had an attack of confusion and amnesia that suggested transient global amnesia. However, her loss of memory for recent events lasted more than ten days and was accompanied by psychomotor agitation and transient alteration of sexual behavior. The patient had no other neurologic signs during the episode. She recovered completely from the recent memory deficit, but was left with a persistent retrograde amnesia for a period of five to ten years and total amnesia for the acute episode. The EEG was suggestive of a left medial temporal lobe lesion.

The 5-HT 1A receptor antagonist WAY 100635 improves rats performance in different models of amnesia evaluated by the object recognition task.

The effects of the 5-HT(1A) receptor antagonist WAY 100635 on recognition memory were investigated in two different amnestic models in the rat by using the object recognition task. WAY 100635 at 1 mg/kg, but not at 0.3 mg/kg, counteracted scopolamine-induced performance deficits in the acquisition version of this behavioral paradigm. At the same dose, WAY 100635 antagonized extinction of recognition memory in the normal rat, suggesting that it affected acquisition, storage and retrieval of information. These results support and extend prior findings that interactions between the serotonergic and cholinergic systems are relevant to cognition and indicate that WAY 100635 modulates different aspects of recognition memory.

Unexpected amnesia: are there lessons to be learned from cases of amnesia following unilateral temporal lobe surgery?

Cases of amnesia following unilateral temporal lobe surgery are rare, but they may provide important insights into human brain functioning. Such cases are reconsidered here in the light of recent developments in clinical and cognitive neuroscience. Descriptions of preoperative seizure activity in these cases indicate the potentially valuable role of ictal semiology in localizing the source of epileptiform discharges. Cases of amnesia after unilateral temporal lobectomy illustrate the complexity of intra- and inter-hemispheric propagation of epileptiform discharges and highlight possible neurophysiological mechanisms underlying false localization of abnormal EEG activity. This review points to the value of preoperative neuropsychological assessment in providing information on the likely primary locus of pathology and in predicting outcome after surgery. The analysis of cases upholds the benefits of the Wada procedure, but it highlights the variability in Wada test procedures and the fact that Wada test scores themselves may be open to varying interpretation. These cases of postoperative amnesia are further considered in the context of the cognitive neuroscience of human memory and, in particular, mechanisms underlying the human amnesic syndrome. They confirm the critical role of bilateral medial temporal lobe structures in anterograde memory, but they also highlight the complexity in teasing apart neural mechanisms underlying remote memory loss.

1-[2-(4-nitrophenoxy)acetyl]pyrrolidin-2-one: an antiamnesic agent.

The title compound, C(12)H(12)N(2)O(5), is a potential antiamnesic agent. The pyrrolidinone ring has an envelope conformation, and the central moiety is almost coplanar with the planes of the phenyl and pyrrolidinone rings. In the crystal structure, weak intermolecular C--H...O interactions link the molecules into a complex network that can be described by R(2)(2)(X) rings (X = 16, 20 and 26) and a C(12) chain.
