Factors affecting adrenocortical hormone function.

Abstract

This paper will briefly outline those elements which must be considered in assessing the effects of any given compound which might impinge on the actions of adrenocortical steroids. Considerations that ought to be taken into account include the following: the delivery to the affected cells of corticosteroids, the uptake of the hormone by the cells, the metabolism of the hormone by the cells, the intracellular actions of the hormone, the possible secondary interactions between cells, and multihormonal actions on the final target cell.

Adrenocortical suppression blocks the enhancement of memory storage produced by exposure to psychological stress in rats.

Several reports have indicated that various stress stimuli modulate learning and memory processes. In the present study, the effects of adrenocortical suppression with the 11beta-hydroxylase inhibitor metyrapone on the psychological stress-induced changes in memory storage in inhibitory avoidance training and in serotonin turnover in various brain regions were investigated in rats. Retention of one-trial inhibitory avoidance and the plasma corticosterone level were significantly enhanced by post-training exposure to psychological stress for 1 h. Pretreatment with metyrapone (12.5 or 25 mg/kg, s.c.) 90 min beforehand dose-dependently blocked the enhancement of memory storage and of the plasma corticosterone level produced by psychological stress. These results suggest that the adrenocortical system may contribute to the memory-enhancing effect of psychological stress. In a neurochemical study, a significant increase in serotonin turnover in the hippocampus and limbic forebrain, including the nucleus accumbens, were observed in rats that were exposed to psychological stress. In contrast to the behavioral experiments, these changes in serotonin turnover produced by exposure to psychological stress were not antagonized by pretreatment with metyrapone; instead, a further increase in serotonin turnover was observed only in the hippocampus. These results suggest that the serotonergic system in the hippocampus might be selectively regulated by adrenal steroids in response to stress, and imply the existence of negative feedback mechanisms via a hippocampal serotonergic system in the memory enhancement associated with corticosterone and psychological stress.

Hypothalamic oxytocin in the developing ovine fetus: interaction with pituitary-adrenocortical function.

Oxytocin (OT) stimulates corticotroph function in adult sheep, however, there is little information on OT synthesis and its potential involvement in hypothalamo-pituitary-adrenal (HPA) function in the fetus. The objectives of this study were to examine developmental changes in hypothalamic OT synthesis and to investigate the actions of OT on fetal corticotroph function. Hypothalami were removed at various stages of pre- and post-natal development. OT mRNA levels were measured using in situ hybridization. For in vitro studies, fetal pituitaries were removed on days 129 and 138 of gestation. Anterior pituitary cells were dispersed and cells were treated with different concentrations and combinations of OT, corticotrophin-releasing hormone (CRH), vasopressin (AVP) and cortisol. OT mRNA was present in the paraventricular nucleus (PVN) and supraoptic nucleus (SON) by day 60 of gestation, and levels significantly increased at term. OT mRNA was present in parvocellular and magnocellular fields of the PVN. In vitro, OT stimulated adrenocorticotropin (ACTH) output in a dose-dependent fashion, but had no effect on cellular pro-opiomelanocortin (POMC) mRNA levels. There was no significant difference in corticotroph responsiveness to secretagogues between cells harvested at gestation day 129 or gestation day 138. Simultaneous exposure to CRH and OT stimulated increases in ACTH output that were significantly greater than for OT or CRH alone. However, no similar synergistic interaction existed between OT and AVP. Cortisol attenuated OT-stimulated ACTH output. In conclusion, hypothalamic OT mRNA increases at term and OT can stimulate ACTH output from fetal corticotrophs. Together, these data indicate that OT may be involved in the regulation of ACTH secretion in fetal sheep in late gestation.

The importance of being dehydroepiandrosterone sulfate (in the blood of primates): a longer and healthier life?

The general aging sequence in tissues of healthy human beings is proposed to be: capillary endothelial cell damage --> arteriosclerosis --> decreased blood flow --> metabolic dysregulation --> secondary tissue damage. Molecular O2 is an obligatory substrate for the successive syntheses of 17alpha-OH pregnenolone and dehydroepiandrosterone (DHEA) by cytochrome P450c17 in the zona reticularis of the adrenal cortex, in which it is suggested that arteriosclerosis --> decreased blood flow --> O2 and glucose deficit --> decreased O2-requiring synthesis of DHEA --> eventual decrease in number of DHEA-synthesizing cells. Aging changes in the zona reticularis synergize with those in the hypothalamo-hypophyseal machinery that controls it neurally and hormonally, with ACTH-evoked pulsatile floods of cortisol coming from the adrenal zona fasciculata, with the onslaught of free radicals generated by the metabolism of catecholamines released from interdigitating cells of the adrenal medulla, and with age-correlated disabilities of erythrocytes to bind and release O2 to decrease the viability of the DHEA and dehydroepiandrosterone sulfate (DHEAS)-forming cells. One of the chief functions of serum DHEAS in the male may be to act as an allosteric facilitator of the binding of testosterone (T) to serum albumin, thereby helping target T to specific receptors and to allosteric sites for rapid and efficient action at the cellular level. There is reason to consider combining O2 therapy with appropriate administration of DHEA and T to optimize steroid functionality in the healthy aging male, and thus, possibly, to alleviate some of the age-related cognitive and physical decrements that occur.
