Forensic DNA analysis of biological specimens such as blood and semen often yields vital evidence in contemporary criminal investigations. Appropriate, correctly performed physical examination in cases of sexual offences is of crucial importance in providing the forensic analyst with genital and extra-genital samples, for example. Despite the availability of standard guidelines for procedures to be followed by the examining physician during sampling, many cases submitted to the National Laboratory of Forensic Science at Linköping lack adequate material for forensic analysis. A recently modernised 'sexual offence kit' is now available, containing the equipment and instructions needed to facilitate standard forensic sampling for the general physician.
The haematopoietic stem cells remaining in the placenta after the umbilical cord has been cut can substitute for bone marrow in transplantations. It is argued that these cells should be retrieved and kept frozen as the child's personal biological spare part. A pilot, population-based collection of cord blood was carried out in a small provincial in Denmark. It was found that the pregnant women supported the concept (89%), that the midwives and technicians could carry out sampling and freezing after a short instruction period, and that the cost of sampling and storing for 75 years approx. DKK 12,000 per child. The project group is now seeking partners for further development of the concept.
Literature values for human plasma GSH vary over 10-fold despite the use of apparently valid analytical procedures for GSH measurement. The purpose of this study was to develop a procedure to minimize error in sample collection, processing and storage that could contribute to such differences. HPLC with fluorescence detection of dansyl derivatives was used for quantification. The results show that collection of blood with a butterfly needle and syringe reduces overestimation due to limited hemolysis and that use of a preservation solution designed to inhibit autooxidation and enzymatic degradation allows quantitative recovery of both GSH and GSSG. Stability tests showed that non-derivatized samples were stable for at least 2 months at - 80 degrees while dansyl derivatives were stable in the dark at 0-4 degrees for 12 months. Results from 59 healthy individuals (20-43 years) provided a mean (+/-1 SD) GSH value of 2.09+/-1.14 micromolar.
