Relationships between cotransducible plasmids in Staphylococcus aureus.

Abstract

Cotransduction of two or even three plasmids was observed when a Staphylococcus aureus strain, carrying five distinct, compatible plasmids, was used as donor. An active host recombination system did not seem to be indispensable for plasmid cotransduction, since RecA+ and RecA- donors gave similar cotransduction frequencies. Analysis of plasmids carried by cotransductant clones demonstrated that a genetic interaction can take place between cotransduced plasmids, leading to new plasmids. Some of the properties of these new plasmids are discussed. Another set of experiments tested the ability of a cotransducible plasmid to allow a significant degree of multiplication of a temperature-sensitive plasmid at restrictive temperature. In an attempt to explain the results obtained, a working hypothesis suggesting a transient and reversible association of cotransducible plasmids is presented.

Interactions between chromosomal omnipotent suppressors and extrachromosomal effectors in Saccharomyces cerevisiae.

Chromosomal omnipotent suppressor mutations recovered in psi+ strains of Saccharomyces cerevisiae were brought into psi- cytoplasm. SUP46, SUP138 and SUP139 acted as dominant omnipotent suppressors in the psi- cytoplasm though their suppressor activity was substantially reduced. SUP46 and SUP138 conferred recessive thermosensitivity and antibiotic sensitivity in psi- cytoplasm as in psi+ cytoplasm. On the other hand, sup111 through sup115, which acted as recessive omnipotent suppressors in the psi+ cytoplasm, manifested no, or very low, suppressor activity in the psi- cytoplasm. They, however, still enhanced the efficiency of the SUP29 tRNA suppressor in psi- cytoplasm. A multicopy plasmid carrying the wild-type SUP35 gene enhanced the efficiency of sup111 in psi- cytoplasm.

Extrachromosomal Inheritance

In search of non-random X inactivation: studies of fetal membranes heterozygous for glucose-6-phosphate dehydrogenase.

Extraembryonic membranes and fetal tissues were obtained from 55 specimens of 5--11 weeks conceptual age. The glucose-6-phosphate dehydrogenase (G6PD) electrophoretic phenotype was determined and correlated with that of maternal blood. Fifteen specimens were heterozygous for G6PD A, and for nine of these the maternal allele could be determined. In none of these specimens did the isozyme pattern of the membraneous chorion or chorionic villi differ significantly from that of fetal tissue. We have obtained no evidence of non-random inactivation in extraembryonic membranes of human fetal specimens at this stage of development.
