Antiphlogistic aryloxypropionic acids: configurational study.

Abstract

Configurational relationships of a series of antiphlogistic 2-aryloxypropionyl derivatives by means of 1H-NMR and HPLC methods were demonstrated. NMR spectra of racemic mixtures and optically active esters were recorded by adding suitable quantities of Eu(hfc)3 chiral shift reagent. The chemical shift values of the non-equivalent signals were unambiguously assigned to R and S enantiomers and the sign of the delta delta parameter was shown to be the same for all compounds. Chiral resolution of (1-naphthyl)methylamides on R-DNBPG and S-DNBL analytical columns was carried out. The alpha and K' values of the chromatographic separations by means of S-DNBL phase were generally better than those using R-DNBPG. The elution order could be determined: S isomers are generally eluted last from both columns. For all compounds the solute-CSP interaction was studied by molecular models to verify if the interacting conformation is the same for the enantiomers which have equal configuration. This control is essential to validate the configurational assignment method by means of HPLC analysis. Finally, we hypothesized the interactions of the compounds showing inversion of the elution order or lack of chiral resolution.

Ion selectivity in the Chlamydomonas reinhardtii flagellar regeneration system.

Extracellular Ca2+ mediates the cellular and molecular responses to cell stimulation of Chlamydomonas reinhardtii. Extracellular Ca2+ concentrations ([Ca2+]e) must exceed certain threshold values to support flagellar excision by acid shock and to stimulate flagellar outgrowth following mechanical shear of the flagella. Also, the magnitude and duration of flagellar RNA accumulations following acid shock or mechanical shear increase with increasing [Ca2+]e. To better understand the role of Ca2+ in flagellar excision, RNA induction, and outgrowth, we have performed a survey of the ion selectivity of each of these responses to acid shock. We found that flagellar excision in vivo following acid shock was supported by Sr2+ and Ca2+, but no other ion tested. LaCl3 and neomycin prevented flagellar excision following acid shock of cells in Ca2+- or Sr2+-containing buffer. Sr2+ addition to detergent-permeabilized cell models, however, failed to elicit flagellar excision in vitro. Cells failed to regrow flagella following flagellar excision in Sr2+-containing buffer unless exogenous Ca2+ was added. Flagellar RNA accumulations of lower magnitude and shorter duration were measured in cells acid-shocked in Sr2+-containing buffer than in Ca2+-containing buffer. These results demonstrate that a Sr2+ influx can evoke flagellar excision following acid shock, but cannot directly activate the machinery for flagellar excision, suggesting that a Sr2+ influx induces excision by stimulating an intracellular Ca2+ release. Furthermore, they suggest that flagellar outgrowth and normal flagellar RNA induction have a strict requirement for Ca2+, which is not satisfied by the proposed intracellular Ca2+ release.

Concerning speed, cost and on-line capabilities, biosensors offer attractive alternatives to existing methods for food analysis. They make monitoring and control of manufacturing processes possible. Furthermore, portable biosensors could be used for monitoring in manufacturing, retail and distribution of foods. An overview is given about existing biosensors for foodstuffs that could find applications in food industry.

Spectral features of central pattern generation in the in vitro brain stem spinal cord preparation of the newborn rat.

In the present investigation, brain stem spinal cord preparations of 0-4-day-old rats were used to determine whether inspiratory-related discharges were modulated by a central pattern generator either during baseline conditions or during conditions of increased chemical drive. Spectral analyses were carried out on pairs of nerve activities during superfusion with normal solutions (pH = 7.4) and during superfusion with acidic solutions (pH = 6.8-7.0). Autopower spectra of nerve discharges in normal pH solution revealed the presence of two peaks: one in the 2-6 Hz band and the other in the 20-39 Hz band. Peaks occurring over both frequency ranges were highly correlated as revealed by coherence spectral analysis. Acidic stimulation produced no systematic changes in spectral features, for example, shifting peaks to other frequency regions, or increasing the values of coherence. The 2-6 Hz peak is most likely due to the arrival of depolarizing inputs from the brain stem that generate a ramp of activity at recording sites. On the other hand, activity in the 20-39 Hz region represents the discharge frequency of inspiratory motoneurons. The fact that coherence is present in this latter band provides evidence for short-time scale (ms) synchronization of functionally and anatomically distinct inspiratory motoneurons by a central pattern generator.
