Specificity and effector-target interaction.

Abstract

When effector cells were tested on a number of target cells, the most obvious effect observed was a nonselective cytotoxicity that is now known as natural cell-mediated cytotoxicity. The apparent nonselectivity was a result of specific cytotoxic activity against many different antigens on target cells. This specificity was demonstrated in the cross-competition assay in which target cells sharing common antigens selectively inhibited natural cytotoxicity.

An indirect sandwich ELISA for LP(a) in serum and dried blood spots.

We have established a reproducible and inexpensive indirect sandwich ELISA for Lp(a) quantitation in both serum and dried blood spot samples. All reagents used in the assay are available commercially. The intra-assay CVs were 3.8 +/- 0.9% for serum and 4.5 +/- 1.7% for dried blood spots on filter paper. The inter-assay CVs were 6.3 +/- 2.3% and 4.5 +/- 0.1% for serum and dried blood spot, respectively. Lp(a) concentrations measured by the ELISA and a commercial RIA were highly correlated (r = 0.989, n = 60, P less than 0.001). However concentrations measured by RIA were 34.3% +/- 9.7% higher than those by ELISA. Lp(a) concentrations in serum and in dried blood spots were also highly correlated (r = 0.966, n = 40, P less than 0.001). This indirect ELISA is suitable for assaying large numbers of serum or dried blood spot samples. However, the differences between the concentrations measured by ELISA and RIA stress the need for standardization of Lp(a) measurements.

Carbonylmetalloimmunoassay (CMIA) a new type of non-radioisotopic immunoassay. Principles and application to phenobarbital assay.

A new non-radioisotopic immunoassay procedure, which we have termed carbonylmetalloimmunoassay (CMIA), is described. The tracers used in this approach are organometallic carbonyl complexes that can be detected at femtomole levels (300-700 fmol) by Fourier transform infrared (FT-IR) spectroscopy. The validity of the technique has been tested in a phenobarbital assay using as the marker a cyclopentadienylmanganese (I) tricarbonyl (cymantrene) moiety, ethyl acetate extraction to separate the free and bound organometallic fractions, and FT-IR spectroscopy to detect the CO stretching modes of the organometallic label. Typical dilution and standard curves obtained with this CMIA procedure are presented. The method was of comparable sensitivity to a [14C] radioimmunoassay (RIA) for the detection of phenobarbital. A comparison of the results for phenobarbital assays by both CMIA and RIA showed that higher titres were obtained using the CMIA method. The standard curves suggest that CMIA is a reliable and reproducible immunoassay procedure for phenobarbital.

An enzyme immunoassay and immunoblot analysis for curculin, a new type of taste-modifying protein: cross-reactivity of curculin and miraculin to both antibodies.

We have developed an enzyme immunoassay method for curculin, a new type of taste-modifying protein. This method can accurately quantify 0.05-20 ng of curculin, a sensitivity about 3000-times that of the psychometric method. The content of curculin in the fruit of Curculigo latifolia increased gradually until 3 weeks after artificial pollination and dramatically at 4 weeks, to finally reach 1.3 mg per fruit. Immunoblot analysis indicated that antiserum to curculin was faintly reactive with miraculin, but not with thaumatin or monellin.
