A modified aortic multiple-ring preparation for functional studies.

Abstract

A comparison is made of four rat aortic preparations, that is, single ring, spiral strip, zig-zag strip, and multiple-ring, on the basis of the responses to norepinephrine (NE) and acetylcholine (ACh). The single ring preparation was suitable under isometric, but not under isotonic conditions, because of the small isotonic tension which developed in response to the contractile agonist. The spiral and the zig-zag strips showed a discontinuity in the relaxant activity of the ACh, probably because of the removal, or not, of the endothelium from the preparation. The reproducibility of both contracturant and relaxing responses in the multiple-ring preparation makes this the most suitable of all four for the study of vasoactive drugs.

Norepinephrine facilitates the development of the murine sweat response but is not essential.

During development, the sympathetic neurons innervating sweat glands undergo a neurotransmitter switch from noradrenergic to cholinergic between postnatal day (P) 4, when the sympathetic neurons first contact the sweat glands, and P21. Several in vitro experiments suggest that norepinephrine (NE), produced by sympathetic neurons, stimulates sweat glands to produce a factor that then induces the phenotypic switch. We tested this hypothesis in vivo using dopamine beta-hydroxylase-deficient mice (DBH -/-), which are unable to synthesize NE and epinephrine, and tyrosine hydroxylase-deficient mice (TH -/-), which are unable to synthesize any catecholamines. The cholinergic agonist pilocarpine and electrostimulation of the sciatic nerve both elicited a sweat response in adult DBH -/- mice that was indistinguishable from the response of controls, and the cholinergic antagonist atropine effectively blocked these responses. We did note, however, a 1- to 2-week delay in the acquisition of the sweat response in DBH -/- mice. Although diminished in magnitude, a sweat response to pilocarpine was also noted in TH -/- mice at P21. Immunohistochemistry demonstrated that TH and vasoactive intestinal peptide were detectable at P14 and increased to adult levels by P21 in DBH +/- and DBH -/- mice. These observations indicate that NE is not essential for the acquisition of the cholinergic phenotype, but it may facilitate its postnatal development.

Differential control of cortical activity by the basal forebrain in rats: a role for both cholinergic and inhibitory influences.

Using microdialysis and high-performance liquid chromatography, we measured acetylcholine (ACh) release simultaneously from two cortical sites in anesthetized rats. One site was always in the somatosensory cortex, and the other was in either the visual or the motor cortex. After baseline measurements were obtained, selected sites in the basal forebrain (BF) were stimulated to increase ACh release. Some BF sites provoked more release in one microdialysis probe than in the other, suggesting some degree of corticotropic organization of the cholinergic projections from the BF. BF sites optimal for release from the visual cortex were separated from optimal sites for release from the somatosensory cortex by greater distances than were the best sites for release from the somatosensory and the motor cortex. Stimulation of a single BF site often provoked similar release from the latter two cortical areas. Electrical stimulation of the BF also modified cortical neuronal activity. Activation of some BF sites provoked an intense discharge of many neurons in the vicinity of the cortical recording electrode, and the same stimulus site in the BF provoked release of large amounts of ACh in the cortex. Stimulation of other BF sites produced strong inhibition of ongoing cortical activity and no increase in cortical ACh release. When other sites were stimulated, they had no effect or they generated stereotyped bursting patterns in the cortex without any observable effect on ACh release. BF sites that generated inhibition of cortical neural activity were generally located near the sites that activated the cortex and provoked release of ACh. These data suggest an elaborate control of the sensory cortex by a mechanism involving both gamma-aminobutyric acid-containing and cholinergic neurons of the BF.

Effect of neostigmine on concentration and extraction fraction of acetylcholine using quantitative microdialysis.

A quantitative microdialysis method was used to determine the effect of local perfusion of 0, 100, 200, and 300 nM neostigmine (NEO) on acetylcholine (ACh) extracellular concentration and microdialysis extraction fraction (E(d)) in the striatum of the rat. Because of the efficiency of AChE, the inhibition of this enzyme is expected to result in a substantial increase in ACh levels and a decrease in the E(d) of ACh. The extracellular concentration of ACh increased linearly with increasing concentrations of NEO. The control ACh concentration was determined to be 18.4 +/- 11.8 nM (n = 10; mean +/- S.E.M.) The ACh extracellular concentration for the remaining groups was determined to be 173 +/- 14 nM (n = 5), 329 +/- 52.5 nM (n = 13), and 581 +/- 109 nM (n = 10) for the 100, 200, and 300 nM NEO groups, respectively. Perfusion with 300 nM NEO resulted in a significant reduction in the E(d) of ACh (64.5 +/- 3.5% vs. 43.6 +/- 7.5%, P < 0.05). In contrast to ACh, perfusion with 0, 1, and 10 microM hemicholinium-3, an inhibitor of high-affinity choline uptake, increased choline levels but did not affect the E(d) of choline. The effects on E(d) are consistent with E(d) being influenced by rapid clearance mechanisms.
