Determination of vitamins in food based on supercritical fluid extraction prior to micellar electrokinetic capillary chromatographic analyses of individual vitamins.

The separation of 14 water-soluble vitamins and vitamin cofactors was investigated by micellar electrokinetic capillary chromatography and diode array detection using sodium cholate as the micellar phase. The method was optimised with respect to the effect of buffer composition, capillary temperature and applied voltage resulting in separation of all compounds in about 25 min. With the current method it is possible to predict the eluting order of the individual compounds from their net charges of each compound because of a low ion pairing between solutes and micelles. The linearities within concentration ranges of up to two-orders of magnitudes were good with correlation coefficients from 0.971 to 0.997. The separation efficiency was satisfactory with a good resolution ranging from 2 to 45 and a theoretical number of plates varying from 200,000 to 480,000. The repeatability of the developed method showed relative standard deviations on migration time in the range from 0.5% to 1.2% (n = 15) and for normalised peak areas, relative standard deviations were approximately 6%.

Caffeine intake and low birth weight: a population-based case-control study.

The authors conducted a matched case-control study to investigate the effects of caffeine intake during pregnancy on birth weight. From January to November 1992, in the first 24 hours after delivery, 1,205 mothers (401 cases and 804 controls) were interviewed and their newborns were examined to assess birth weight and gestational age by means of the method of Capurro et al. (J Pediatr 1978;93:120-2). The cases were children with birth weight < 2,500 g and gestational age > or = 28 weeks. Cases and controls were matched for time of birth and hospital of delivery and were recruited from the four maternity hospitals in Pelotas, southern Brazil. Daily maternal caffeine intake during pregnancy for each trimester was estimated. To assess caffeine intake, 10% of the mothers were reinterviewed at their households and samples of reported information on drip coffee and maté (a caffeine-containing drink widely used in South America) were collected and sent to the laboratory for caffeine determination through liquid chromatography. When instant coffee was reported, the weight of powder was measured using a portable scale, and caffeine intake was estimated from a reference table. Caffeine intake from tea, chocolate, soft drinks, and medicines was estimated from a reference table. Analyses were performed by conditional logistic regression. Crude analyses showed no effect of caffeine on low birth weight, preterm births or intrauterine growth retardation. The results did not change after allowing for confounders.

Simultaneous separation of flavanone glycosides and polymethoxylated flavones in citrus juices using liquid chromatography.

We present a simultaneous liquid chromatographic method for the separation of two flavonoid compound families, flavanone glycosides (FGs) and polymethoxylated flavones (PMFs), which are usually found in citrus fruits species and varieties. This technique permits the quantitation of six FGs (narirutin, naringin, hesperidin, neohesperidin, didymin, poncirin) and six PMFs (sinensetin, hexamethoxyflavone, nobiletin, scutellarein, heptamethoxyflavone and tangeretin). This technique, to be used to characterize a citrus juice by its polyphenolic profile, has been applied to the determination of flavonoid compounds in grapefruit- and orange juice. Differentiation of orange juice varieties and mixtures containing tangor juice using polyphenolic profiles and flavonoid content has been achieved.
