Multiple-time-point dissolution specifications for a sampling acceptance plan.

Abstract

In dissolution testing, multiple dissolution measurements at specific time points are used to obtain the dissolution characteristics for most extended-release and some immediate-release drug products. This paper presents a general procedure for defining specifications by a multivariate confidence region or by simultaneous confidence limits on the dissolution values of individual time points. The confidence regions and simultaneous confidence limits were estimated using two approaches: the first approach assumed a multivariate normal distribution on the multiple dissolution values and the second approach used the bootstrap resampling method. The multivariate confidence region was constructed using the Hotelling's T2 statistic (equivalently, Mahalanobis distance D2), and the simultaneous confidence limits were based on the Mahalanobis statistic as well as on the Bonterroni adjustment. The Mahalanobis simultaneous confidence limits specification criteria have the overall false out-of-specification rate too low in both parametric and bootstrap approaches.

Education of medicinal chemists in Departments of Medicinal Chemistry (U.S.A.).

The present state of faculties, student bodies, and curricula in departments of medicinal chemistry have been surveyed by questionnaire and analyzed in the context of perceptions of quality and content. The results reveal a healthy diversity of educational objectives and a broader range of educational objectives than those uncovered in previous surveys of the perceived needs of industrial departments of medicinal chemistry in their search for drug discovery personnel.

A computer-based expert system designs and analyzes a 2(k - p) fractional factorial design for the formulation optimization of novel multicomponent liposomes.

A computer-based technique based on a 2(k - p) fractional factorial design was applied for the optimization of recently described multicomponent protective liposomal formulations. These formulations contain sodium ascorbate (vitamin C) as a model drug sensitive to photochemical oxidation, as well as oil red O and/or oxybenzone as oil soluble light absorbers, incorporated into the lipid bilayers and sulisobenzone as a water soluble light absorber incorporated into the aqueous phase of liposomes. The three light absorbers (present or absent) incorporated in multilamellar liposomes and the drug in free or in complexed with alpha-cyclodextrin form comprised the four factors of the system. The stabilization ratio and the percentage entrapment in the liposomes of the vitamin were the two response variables of the system to be optimized. The entrapment values were calculated for all the materials either spectrophotometrically or by using second order derivative spectrophotometry. The response variables were predicted by multiple regression equations comprising combinations of the four formulation factors. Both the higher entrapment and the higher protection for the drug should characterize the optimum formulation.

Influence and correction of temperature perturbations on NIR spectra during the monitoring of a polymorph conversion process prior to self-modelling mixture analysis.

The influence of temperature variations on the rank of a NIR dataset, has been investigated by comparing the results of principal component analysis (PCA) and evolving factor analysis (EFA), applied to two datasets measured at constant temperature and varying temperature. After temperature correction, the concentration profiles and spectra were obtained with PCA, SIMPLISMA and the orthogonal projection approach (OPA). The same resolution methods were used on the dataset measured at constant temperature.
