[New method for recording human cochlear microphonic potentials].

Abstract

The use of invasive methods to record cochlear microphonic potentials preclude its use in routine clinical diagnosis in humans. We present an instrument and method used to obtain cochlear microphonics recordings using external electrodes in human beings. Biological and technical tests were performed before application. Stimulation with a pure, continuous and defined tone yielded cochlear microphonics that can be interpreted easily. This new method allows human cochlear physiology and pathophysiology to be studied non-invasively.

The intensity of masking noise affects the mismatch negativity to speech sounds in human subjects.

The present study investigated the effects of decreased audibility of the speech sounds 'ba' and 'da' on the mismatch negativity (MMN). Audibility was altered by varying the intensity of the masking noise to be either 0 (no noise), 65, 70 or 75 dB speech perception level. Cortical event-related potentials were recorded while normal listeners (n=7) were instructed to ignore the stimuli. In a separate condition, subjects were asked to signal detection of the deviant stimulus by button-press. As audibility of speech sounds was decreased, MMN peak amplitude decreased and MMN peak latency increased. Behavioral responses were in accordance with the MMN data. Hit rates decreased as audibility decreased. These results suggest that both pre-attentive and attentive speech discrimination deteriorate following a reduction in audibility.

Anti-epileptiform effects of audiogenic seizure priming on in vitro kindling in rat hippocampus.

The effect of priming for audiogenic seizures (AGS) on the development of epileptiform activity in the hippocampus was studied using in vitro kindling (IVK) in Long-Evans rats. AGS priming consists of intense auditory stimulation during a critical period of auditory development, resulting in sound-induced clonic convulsions upon subsequent testing. Between postnatal day (PND) 28 and 50, slices from subjects primed and sham-primed for AGS on PND 18 were used for recording responses in area CA1 of hippocampus following Schaffer collateral stimulation from stratum radiatum of area CA2/CA3. The developmental priming procedure, which enhances auditory brainstem excitability, resulted in fewer afterdischarges in slices from primed subjects across initial IVK stimulation sequences. These results suggest that changes in excitability that occur with acoustic priming can initially diminish selective epileptiform response characteristics in forebrain areas such as the hippocampus.

L-NAME precipitates catatonia during ethanol withdrawal in rats.

The effect of N(G)-nitro-L-arginine methyl ester (L-NAME), a non-specific inhibitor of nitric oxide (NO) synthase, on catatonia in ethanol dependent rats was investigated. Ethanol was given to rats by a modified liquid diet. An isocaloric liquid diet without ethanol was also given to control rats. L-NAME (50, 100 and 200 mg/kg) and saline were injected intraperitoneally to ethanol-dependent rats 30 min before ethanol withdrawal. Then, catatonia was evaluated by vertical wire test at the 30th min, 2nd, 4th and 6th h of ethanol withdrawal. The injections were repeated 30 min before the observation of 6 h. Locomotor activity was also recorded for 5 min in the same observation intervals. L-NAME (200 mg/kg) or saline were also injected to ethanol non-dependent control rats. L-NAME (50 and 100 mg/kg) inhibited both incidence and intensity of the audiogenic seizures which appeared at 6 h of ethanol withdrawal. L-NAME (200 mg/kg) produced a significant augmentation in both incidence and intensity of the catatonia in ethanol dependent rats. This dose of L-NAME also reduced the locomotor activity of both ethanol dependent and non-dependent rats. The locomotor inhibitory effect was more prominent in ethanol-dependent group. The catatonia precipitating effect of L-NAME was not prevented by L-arginine (1 g/kg, i.p.), a NO precursor, pretreatment. In the naive rats, L-NAME also did not produce catatonia. These results indicate that L-NAME has a catatonia precipitating effect during ethanol withdrawal in rats and this effect seems to be independent from NO mediated mechanisms.
