Low-frequency modulation sensors using nanosecond fluorophores.

Abstract

We describe a new approach to fluorescence sensing based on a mixture of fluorophores, one of which is sensitive to the desired analyte. If a long-lifetime analyte-insensitive fluorophore is mixed with a short-lifetime analyte-sensitive fluorophore, the modulation of the emission at conveniently low frequencies becomes equal to the fractional fluorescence intensity of the sensing fluorophore. Under these conditions, the modulation can be used to determine the analyte concentration. This can be used with any fluorophore that changes intensity in response to analyte and does not require the sensing fluorophore to display a change in lifetime. The feasibility of modulation-based sensing was demonstrated using mixtures of 6-carboxyfluorescein and [Ru 2,2'-(bipyridyl)3]2+ as a pH sensor and of the calcium probe Fluo-3 and [Ru 2,2'-(bipyridyl)3]2+ as a calcium sensor.

Luminescent Eu(III) and Gd(III) trisbipyridine cryptates: experimental and theoretical study of the substituent effects.

Synthesis, absorption spectra and luminescebce properties of a series of lanthanide trisbipyridine cryptates Ln within R-Bpy x R-Bpy x R-Bpy, where Ln = Eu, Gd and R = H, COOH, COOCH3, CONH(CH2)2NH2 are described. Comparison of the unsubstituted parent compound with the substituted compounds shows that bipyridine substitution doesn't alter significantly the photophysical properties of the lanthanide cryptate. The absorption maximum is slightly red-shifted when three bipyridines are substituted, whereas substituting one bipyridines has a negligible effect on the absorption spectra. The experimental triplet state energy is between 21600 and 22 100 cm(-1) for the series of compounds and the luminescence lifetimes at 77 K are between 0.5 and 0.8 ms in HO2 and equal to 1.7 ms in D2O. The experimental characterizations are completed by DFT and TD-DFT calculations to assess the ability of these approaches to predict absorption maxima, triplet state energies and structural parameters of lanthanide cryptates and to characterize the electronic structure of the excited states. The calculations on the unsubstituted parent and substituted compounds show that absorption maxima and lowest 3pipi* triplet state energies can be accurately determined from density functional theory (DFT) and time-dependent (TD) DFT calculations.

Electrogenerated chemiluminescence. 83. Immunoassay of human C-reactive protein by using Ru(bpy)3(2+)-encapsulated liposomes as labels.

Liposomes ( approximately 100-nm diameter) containing Ru(bpy)32+ (bpy = 2,2'-bipyridine) were prepared as an electrogenerated chemiluminescent (ECL) tag for a sandwich-type immunoassay of human C-reactive protein (CRP). Polyclonal human CRP antibodies were introduced onto liposomes and magnetic beads through biotin-streptavidin interaction. The antigen-antibody conjugates formed on addition of a CRP-containing sample were separated from unreacted species magnetically. Addition of 0.1 M tri-n-propylamine and 0.1 M phosphate buffer (pH 7.6) containing 0.1 M NaCl and 1% (v/v) Triton X-100 caused liberation of the Ru(bpy)32+ from the liposome. ECL obtained in this medium showed a detection limit of 100 ng/mL for human CRP with good linearity of ECL intensity versus antigen concentration over the range 100 ng/mL-10 microg/mL.

Controlling the helicity of 2,2'-bipyridyl ruthenium(II) and zinc(II) hemicage complexes.

Two enantiomers of a new 4,5-pineno-2,2'-bipyridine ligand were synthesized and subsequently incorporated into hemicage ligands through a phenyl linker to yield ligands (+)-L1 and (-)-L1 or through a mesityl linker to yield ligands (+)-L2 and (-)-L2. Complexation of these ligands to Ru(II) afforded diastereomerically pure Delta and Lambda isomers, as verified through circular dichroism and circularly polarized luminescence spectroscopy. Ligands (+)-L2 and (-)-L2 were further coordinated to Zn(II) to form a complex with intriguing photophysical properties. Whereas Zn(bpy)32+ was shown to be a fluorescent emitter outside the visible spectrum, the caging process provided an unprecedented enhancement of intersystem crossing and subsequent switching to the phosphorescent emission of blue light. Additionally, the chiroptical properties of the Zn(II) complexes were also studied.
