Activation of the phosphatidylinositol 3-kinase/Akt pathway protects against interleukin-3 starvation but not DNA damage-induced apoptosis.

Baf-3 cells are dependent on interleukin-3 (IL-3) for their survival and proliferation in culture. To identify anti-apoptotic pathways, we performed a retroviral-insertion mutagenesis on Baf-3 cells and selected mutants that have acquired a long term survival capacity. The phenotype of one mutant, which does not overexpress bcl-x and proliferates in the absence of IL-3, is described. We show that, in this mutant, Akt is constitutively activated leading to FKHRL1 phosphorylation and constitutive glycolytic activity. This pathway is necessary for the mutant to survive following IL-3 starvation but is not sufficient or necessary to protect cells from DNA damage-induced cell death. Indeed, inhibition of the phosphatidylinositol 3-kinase (PI3K)/Akt pathway in Baf-3 cells does not prevent the ability of IL-3 to protect cells against gamma-irradiation-induced DNA damage. This protective effect of IL-3 rather correlates with the expression of the anti-apoptotic Bcl-x protein. Taken together, these data demonstrate that the PI3K/Akt pathway is sufficient to protect cells from growth factor starvation-induced apoptosis but is not required for IL-3 inhibition of DNA damage-induced cell death.

A novel nuclear receptor corepressor complex, N-CoR, contains components of the mammalian SWI/SNF complex and the corepressor KAP-1.

Transcriptional silencing by many transcription factors is mediated by the nuclear receptor corepressor (N-CoR). The mechanism by which N-CoR represses basal transcription involves the direct or indirect recruitment of histone deacetylases (HDACs). We have isolated two multiprotein N-CoR complexes, designated N-CoR-1 and N-CoR-2, which possess histone deacetylase activity that is mediated by distinct HDACs. Based on Western blotting using antibodies against known subunits, the only HDAC found in the N-CoR-1 complex was HDAC3. In contrast, N-CoR-2 contained predominantly HDAC1 and HDAC2 as well as several other subunits that are found in the Sin3A.HDAC complex. Using mass spectrometry and Western blotting, we have identified several novel components of the N-CoR-1 complex including the SWI/SNF-related proteins BRG1, BAF 170, BAF 155, BAF 47/INI1, and the corepressor KAP-1 that is involved in silencing heterochromatin. Indirect immunofluorescence has revealed that both KAP-1 and N-CoR colocalize throughout the nucleus. These results suggest that N-CoR is found in distinct multiprotein complexes, which are involved in multiple pathways of transcriptional repression.

Organic and hydraulic shock loadings on a biological aerated filter.

The performance of a biological aerated filter (BAF) during short-term (60 min) organic and hydraulic shock loads was investigated. The BAF used for the present experiment was not capable of absorbing high peak organic shock loads when the organic loads were increased from a normal load of 1.2-1.4 kg soluble chemical oxygen demand (sCOD ) m(-3) d(-1) to shock loads between 5.1- 7.3 kg sCOD m(-3) d(-1), based on empty bed volumes. Nevertheless, the effect on the biomass was limited as normal performance resumed very quickly. Increases in the hydraulic velocity from 0.7-1.0 m h(-1) to 1.5-2.9 m h(-1) had little effect on BAF performance in terms of soluble COD removal. Yet, sCOD effluent concentrations of <55 mg l(-1) (approximately 22 mg l(-1) biochemical oxygen demand) were always achieved. The recovery of the reactor in both cases was fast, resuming normal performance within 60 min after the application of the shock load.

Caspase inhibitors block zinc-chelator induced death of retinal ganglion cells.

Zinc-chelating agents, including ethambutol and its metabolite 2,2'(ethylenediamino)-dibutyric acid (EDBA) are toxic to retinal ganglion cells through a glutamate dependent mechanism. We explored whether such cell death was mediated through the caspase family of cysteine proteases. Retinal cultures were treated with EDBA alone, or EDBA plus a variety of known caspase inhibitors, and ganglion cell viability was assayed. EDBA killed 20-30% of ganglion cells. A general caspase inhibitor, BAF, prevented EDBA induced ganglion cell death. Specific inhibitors of caspase-3 and caspase-6 showed a similar ability to BAF in preventing EDBA mediated ganglion cell loss, whereas inhibitors of caspase-8 and caspase-9 were not able to rescue EDBA treated ganglion cells. A caspase-1,4 inhibitor was less effective than BAF. These studies show that a caspase mediated mechanism of apoptosis accents for a portion of EDBA mediated retinal ganglion cell death. This toxicity was mediated by downstream effector caspases, 3 and 6. Caspase inhibitors may prevent ganglion cell death secondary to ethambutol treatment.

Effects of Bafilomycin A1 and Metabolic Inhibitors on the Maintenance of Vacuolar Acidity in Maize Root Hair Cells.

Proton pumps of tonoplast membranes have been studied extensively in vitro, but data concerning their regulation in vivo are lacking. Effects of either anoxia, or the addition of KCN, 2-deoxy-d-glucose (deoxy-glucose), or bafilomycin-A1 (BAF) on vacuolar pH of maize (Zea mays L.) root hair cells were followed by fluorescence microscopy after loading of 2[prime]7[prime]-bis-(2-carboxyethyl)-5-(and-6) carboxyfluorescein. Root hair cells were able to maintain vacuolar acidity for at least 2 h in the presence of either 10 mM KCN or 50 mM deoxy-glucose or during anoxia. Treatments with either deoxy-glucose or KCN reduced total tissue ATP more than anoxia. ADP accumulated during anoxia and treatment with KCN as detected by in vivo 31P-NMR spectroscopy, but not during deoxy-glucose treatment. With control roots and roots treated with deoxy-glucose, the presence of BAF, a specific inhibitor of the V-type ATPase, caused alkalization of the vacuolar pH. However, either in the presence of KCN or under anoxic conditions, BAF was relatively ineffective in dissipating vacuolar acidity. Therefore, under anoxia or in the presence of KCN, unlike the situation with air or deoxy-glucose, the V-type ATPase apparently is not required for maintenance of vacuolar acidity.

E4BP4 expression is regulated by the t(17;19)-associated oncoprotein E2A-HLF in pro-B cells.

The E4BP4 basic leucine zipper (bZIP) transcription factor is regulated by interleukin-3 (IL-3) in pro-B cells and has been reported to promote survival of the murine IL-3-dependent pro-B cell lines, FL5.12 and Baf-3. The E2A-HLF oncoprotein arises from a t(17;19) translocation in childhood pro-B cell acute lymphoblastic leukaemia and acts as an anti-apoptotic factor in FL5.12 and Baf-3 cells. To assess the functions of E2A-HLF and E4BP4 in cell survival, a tetracycline-inducible system was established in Baf-3 cells to express E4BP4 or E2A-HLF. Upon IL-3 withdrawal, expression of E2A-HLF conferred resistance to apoptosis whereas overexpression of E4BP4 did not. E4BP4 and E2A-HLF both recognized the same DNA sequence in reporter gene assays, but had opposite effects on transcription. E2A-HLF acts as a transcriptional activator and E4BP4 as a transcriptional repressor. Furthermore, E4BP4 is a downstream transcriptional target of E2A-HLF. Our data suggests that the overexpression of E4BP4 is unable to block apoptosis induced by IL-3 withdrawal and that the expression of E2A-HLF does not replace the function of E4BP4 in mediating survival.

Genetic variability in French dog breeds assessed by pedigree data.

Pedigree data of nine French dog breeds, namely Barbet (BAR), Basset fauve de Bretagne (BAF), Beauceron (BEN), Berger des Pyrénées (BRP), Bouledogue Français (BUF), Braque Saint-Germain (BQG), Dogue de Bordeaux (DOB), Epagneul Breton (EPB) and Montagne des Pyrénées (MOP), were analysed. The effective numbers of ancestors of dogs born from 1997 to 2001 were equal to 6.7 (BAR), 40.2 (BAF), 36.5 (BEN), 16.0 (BRP), 37.0 (BUF), 13.1 (BQG), 28.9 (DOB), 33.3 (EPB) and 34.0 (MOP). The expected contributions of the major ancestors were found to be highly unbalanced in the EPB and BRP. The average coefficient of inbreeding of dogs born from 1997 to 2001 with both parents known was equal to 12.4% (BAR), 3.9% (BAF), 5.4% (BEN), 7.2% (BRP), 3.3% (BUF), 6.0% (BQG), 4.1% (DOB), 4.5% (EPB) and 4.0% (MOP). These values were found to be significantly higher than the average coefficient of kinship between the male and the female parents of these animals, except in the BAR and BQG, revealing an usual practice of mating between related animals. The results are discussed in relation with the demographic situation and the use of each breed. The method used to class an endangered breed and the ways to preserve the genetic variability, when necessary, are evoked.

Comparison of the plaque-stimulating and thymocyte-stimulating activities derived from human monocytes.

Human monocytes have been reported to release factors that can elicit distinct responses from a number of different target cells. In this report, it is shown that most of the thymocyte-stimulating activity in supernatants of endotoxin-stimulated monocytes can be separated from the plaque-stimulating factor (BAF) by gel filtration and isoelectric focusing; however, since these activities could not be entirely resolved, the question was addressed whether the plaque-stimulating activity of BAF depends upon the stimulation of T-cells. Several critical experiments are reported which fail to support this hypothesis. On the other hand, these experiments led to the observation that the response to BAF depends on both an IgM-positive B-cell and a G10-adherent, plastic nonadherent, IgM-negative, irradiation-insensitive cell found in nude splenocytes. Finally, the possibility is discussed that this factor may be responsible for many of the physiological sequelae of infection.

Improved computed radiography image quality from a BaFl:Eu photostimulable phosphor plate.

Recent advances in the design of photostimulable phosphor (PSP) plates for computed radiography (CR) systems have made it possible to manufacture plates made of BaFI:Eu phosphor material in the cassette form. The image quality of this plate, six BaF(Br,I):Eu plates, and one BaFBr:Eu plate were evaluated in terms of presampling modulation transfer function (MTF), normalized Wiener spectra (WSN), and detective quantum efficiency (DQE). Compared with the best-performing BaF(Br,I):Eu plate, the BaFI:Eu plate provided DQE that was higher, at spatial frequencies of 0.5, 1.0, and 2.0 cycles/mm, by 12% (21.8 versus 19.4), 13% (18.8 versus 16.7), and 11% (12.0 versus 10.8), respectively. Since presampling MTF values of the two plates were comparable, the BaFI:Eu plate's higher DQE is attributable to total WSN conversely being lower by 17% [8.65 x 10(-5) (mm2) versus 10.38 x 10(-5) (mm2)], 17% [5.85 x 10(-5) (mm2) versus 7.03 x 10(-5) (mm2)], and 12% [2.82 x 10(-5) (mm2) versus 3.19 x 10(-5) (mm2)] at the specified frequencies, respectively, primarily due to the contribution of x-ray quantum WSN. This jibes with the high x-ray absorption provided by the 27%-higher x-ray attenuation coefficients (7.54 versus 6.07, at 60 KeV) that BaFI offers over BaF(Br(0.85),I(0.15)), a result of the high atomic number of BaFI's exclusively iodine halide content. The results were consistent with earlier studies of several of these same plates and indicate that BaFI:Eu is a promising avenue to lower image noise and higher overall CR system image quality.

Effect of bafilomycin A1 and nocodazole on endocytic transport in HeLa cells: implications for viral uncoating and infection.

Bafilomycin A1 (baf), a specific inhibitor of vacuolar proton ATPases, is commonly employed to demonstrate the requirement of low endosomal pH for viral uncoating. However, in certain cell types baf also affects the transport of endocytosed material from early to late endocytic compartments. To characterize the endocytic route in HeLa cells that are frequently used to study early events in viral infection, we used 35S-labeled human rhinovirus serotype 2 (HRV2) together with various fluid-phase markers. These virions are taken up via receptor-mediated endocytosis and undergo a conformational change to C-antigenic particles at a pH of <5.6, resulting in release of the genomic RNA and ultimately in infection (E. Prchla, E. Kuechler, D. Blaas, and R. Fuchs, J. Virol. 68:3713-3723, 1994). As revealed by fluorescence microscopy and subcellular fractionation of microsomes by free-flow electrophoresis (FFE), baf arrests the transport of all markers in early endosomes. In contrast, the microtubule-disrupting agent nocodazole was found to inhibit transport by accumulating marker in endosomal carrier vesicles (ECV), a compartment intermediate between early and late endosomes. Accordingly, lysosomal degradation of HRV2 was suppressed, whereas its conformational change and infectivity remained unaffected by this drug. Analysis of the subcellular distribution of HRV2 and fluid-phase markers in the presence of nocodazole by FFE revealed no difference from the control incubation in the absence of nocodazole. ECV and late endosomes thus have identical electrophoretic mobilities, and intraluminal pHs of <5.6 and allow uncoating of HRV2. As bafilomycin not only dissipates the low endosomal pH but also blocks transport from early to late endosomes in HeLa cells, its inhibitory effect on viral infection could in part also be attributed to trapping of virus in early endosomes which might lack components essential for uncoating. Consequently, inhibition of viral uncoating by bafilomycin cannot be taken to indicate a low pH requirement only.

Evidence for a critical role for the cytoplasmic region of the interleukin 2 (IL-2) receptor gamma chain in IL-2, IL-4, and IL-7 signalling.

The high-affinity interleukin 2 receptor (IL-2R) consists of at least three distinct subunits: the IL-2R alpha chain (IL-2R alpha), beta chain (IL-2R beta), and gamma chain (IL-2R gamma). It has been shown that the cytoplasmic region of IL-2R beta, but not of IL-2R alpha, is essential for IL-2 signalling to the cell interior. In the present study, we examined the functional role of the IL-2R gamma cytoplasmic region in the IL-3-dependent mouse hematopoietic cell line BAF-B03, which expresses the endogenous IL-2R alpha and IL-2R gamma, or its subline F7, which additionally expresses human IL-2R beta cDNA. We show that overexpression of a mutant IL-2R gamma, lacking all but 7 amino acids of its cytoplasmic region, results in the selective inhibition of IL-2-induced c-fos gene activation and cellular proliferation in F7 cells. When two chimeric receptor molecules in which the cytoplasmic regions of IL-2R beta and IL-2R gamma had been swapped with each other (IL-2R beta/gamma and IL-2R gamma/beta) were coexpressed in BAF-B03, the cells responded to IL-2. These results indicate the critical importance of the IL-2-induced functional cooperation of the two cytoplasmic regions. Finally, we provide evidence that the IL-2R gamma cytoplasmic region is also critical for the IL-4 and IL-7-induced growth signal transduction in BAF-B03.

Reconstitution of glucocorticoid receptor-dependent transcription in vivo.

We developed a model system to study glucocorticoid receptor (GR)-mediated chromatin remodeling by the BRG1 complex. Introduction of the BRG1 ATPase into the SW-13 cell line initiates the formation of a functional remodeling complex. This complex is able to induce transcriptional activation from a transiently transfected promoter with wild-type and chromatin-remodeling-deficient BRG1 mutants, suggesting that the complex possesses a coactivator function independent from remodeling. Transactivation from a chromatin template requires the BRG1 remodeling function, which induces regions of hypersensitivity and transcription factor loading onto the integrated MMTV promoter. We report that BRG1 remodeling activity is required for GR-mediated transactivation and that this activity cannot be replaced by other ATP-dependent remodeling proteins. Further characterization of the BRG1-associated factors (BAFs) present in these cells (for example, the expression of BAF250 but not BAF180) reveals that the BAF complex rather than the polybromo-associated BAF complex is the necessary and sufficient chromatin-remodeling component with which the receptor functions in vivo. These results in conjunction with previous findings demonstrate that the GR functions with multiple forms of the SWI/SNF complex in vivo.

Production of NAADP and its role in Ca2+ mobilization associated with lysosomes in coronary arterial myocytes.

The present study was designed to determine the production of nicotinic acid adenine dinucleotide phosphate (NAADP) and its role associated with lysosomes in mediating endothelin-1 (ET-1)-induced vasoconstriction in coronary arteries. HPLC assay showed that NAADP was produced in coronary arterial smooth muscle cells (CASMCs) via endogenous ADP-ribosyl cyclase. Fluorescence microscopic analysis of intracellular Ca2+ concentration ([Ca2+]i) in CASMCs revealed that exogenous 100 nM NAADP increased [Ca2+]i by 711 +/- 47 nM. Lipid bilayer experiments, however, demonstrated that NAADP did not directly activate ryanodine (Rya) receptor Ca2+ release channels on the sarcoplasmic reticulum. In CASMCs pretreated with 100 nM bafilomycin A1 (Baf), an inhibitor of lysosomal Ca2+ release and vacuolar proton pump function, NAADP-induced [Ca2+]i increase was significantly abolished. Moreover, ET-1 significantly increased NAADP formation in CASMCs and resulted in the rise of [Ca2+]i in these cells with a large increase in global Ca2+ level of 1,815 +/- 84 nM. Interestingly, before this large Ca2+ increase, a small Ca2+ spike with an increase in [Ca2+]i of 529 +/- 32 nM was observed. In the presence of Baf (100 nM), this ET-1-induced two-phase [Ca2+]i response was completely abolished, whereas Rya (50 microM) only markedly blocked the ET-1-induced large global Ca2+ increase. Functional studies showed that 100 nM Baf significantly attenuated ET-1-induced maximal constriction from 82.26 +/- 4.42% to 51.80 +/- 4.36%. Our results suggest that a lysosome-mediated Ca2+ regulatory mechanism via NAADP contributes to ET-1-induced Ca2+ mobilization in CASMCs and consequent vasoconstriction of coronary arteries.

Functional role of Ca2+ currents in graded and spike-mediated synaptic transmission between leech heart interneurons.

We used intracellular recording and single electrode voltage-clamp techniques to explore Ca2+ currents and their relation to graded and spike-mediated synaptic transmissions in leech heart interneurons. Low-threshold Ca2+ currents (activation begins below -50 mV) consist of a rapidly inactivating component (I(CaF)) and a slowly inactivating component (I(CaS)). The apparent inactivation kinetics of I(CaF) appears to be influenced by Ca2+; both the substitution of Ca2+ (5 mM) with Ba2+ (5 mM) in the saline and the intracellular injection of the rapid Ca2+ chelator, bis-(o-aminophenoxy)-N,N,N',N'-tetraacetic acid (BAPTA), from the recording microelectrode, significantly increase its apparent inactivation time constant. The use of saline with a high concentration of Ba2+ (37.5 mM) permitted exploration of divalent ion currents over a broader activation range, by acting as an effective charge carrier and significantly blocking outward currents. Ramp and pulse voltage-clamp protocols both reveal a rapidly activating and inactivating Ba2+ current (I(BaF)) and a less rapidly activating and slowly inactivating Ba2+ current with a broad activation range (I(BaS)). Low concentrations of Cd2+ (100-150 microM) selectively block I(BaS), without significantly diminishing I(BaF). The current that remains in Cd2+ lacks the characteristic delayed activation peak of I(BaS) and inactivates with two distinct time constants. I(BaF) appears to correspond to a combination of I(CaF) and I(CaS), i.e., to low-threshold Ca2+ currents, that can be described as T-like. I(BaS) appears to correspond to a Ca2+ current with a broad activation range, which can be described as L-like. Cd2+ (100 microM) selectively blocks spike-mediated synaptic transmission between heart interneurons without significantly interfering with low-threshold Ca2+ currents and plateau formation in or graded synaptic transmission between heart interneurons. Blockade of spike-mediated synaptic transmission between reciprocally inhibitory heart interneurons with Cd2+ (150 microM), in otherwise normal saline, prevents the expression of normal oscillations (during which activity in the two neurons consists of alternating bursts), so that the neurons fire tonically. We conclude that graded and spike-mediated synaptic transmission may be relatively independent processes in heart interneurons that are controlled predominantly by different Ca2+ currents. Moreover, spike-mediated synaptic inhibition appears to be required for normal oscillation in these neurons.

BRCA1 negatively regulates the cancer-associated aromatase promoters I.3 and II in breast adipose fibroblasts and malignant epithelial cells.

CONTEXT: Heterozygous mutations of the breast cancer susceptibility gene 1 (BRCA1) gene that lead to haploinsufficiency increase the risk of breast cancer. The underlying mechanism is unknown. OBJECTIVE: Because excessive estrogen production increases the risk of breast cancer, we determined whether BRCA1 suppresses aromatase expression and thus local estrogen production in breast adipose fibroblasts (BAFs) and breast malignant epithelial cells by interacting with the cancer-associated promoter I.3/II region of the aromatase gene. RESULTS: Treatment of BAFs with prostaglandin E2 or a surrogate hormonal cocktail of dibutyryl (Bt2) cAMP plus phorbol 12, 13-diacetate (PDA) significantly reduced BRCA1 levels and induced aromatase mRNA levels. Reduction of BRCA1 in BAFs and in MCF7 and SKBR3 malignant breast epithelial cells using small interfering RNA (siRNA) or small hairpin RNA significantly increased aromatase mRNA levels and enzyme activity. This effect of BRCA1 was mediated via selective inhibition of aromatase promoters I.3 and II that are up-regulated by prostaglandin E2 or Bt2cAMP + PDA treatment. Chromatin immunoprecipitation assays revealed that BRCA1 binds directly to the aromatase promoter I.3/II region and that BRCA1 binding is abolished by treatment with Bt2cAMP + PDA. CONCLUSIONS: Selective inhibition of aromatase expression by BRCA1 binding to the I.3/II tumorigenic promoter region may be an important protective mechanism against breast cancer development.

Trabecular alveolar bone microarchitecture in the human mandible using high resolution magnetic resonance imaging.

OBJECTIVES: Cancellous bone microarchitecture in the mandible can influence the success of dental implant osseointegration. The aims of this study were to explore the feasibility of two-dimensional (2D) high resolution magnetic resonance imaging (MRI) for the evaluation of trabecular bone architecture and to compare architecture parameters derived from MR images between different areas in the mandible, and between sex and dental status. METHODS: 45 mandibular bone specimens (8 mm thickness) were cut from 15 fresh cadavers. MR images were obtained at 2 T using a multislice 2D spin-echo sequence. After MR image binarization, histomorphometry parameters, i.e. bone area fraction (BAF), specific perimeter (La), trabecular bone width (Tb.Wi), trabecular bone separation (Tb.Sp) and trabecular network anisotropy (R), were computed from the mean intercept length. The angle between trabeculae and tooth axis and the Euler-Poincaré characteristics (EPCs) were also computed. RESULTS: BAF, Tb.Wi and R were significantly higher in male specimens compared with female specimens, whereas Tb.Sp was significantly lower. The apparent Tb.Wi was found to be significantly higher in dentate specimens compared with edentulous ones. The highest anisotropy, corresponding to the smallest R value, was seen in the incisal specimens. The preferential orientation of the trabeculae was close to perpendicular to the tooth axis, especially in the molar specimens. CONCLUSION: BAF, La, Tb.Wi, Tb.Sp and R, the most variable parameters, may potentially have a relationship with the biomechanical competence of trabecular bone and play a role both in primary stabilization of dental implant and the time needed before loading.

The role of desorption for describing the bioavailability of select polycyclic aromatic hydrocarbon and polychlorinated biphenyl congeners for seven laboratory-spiked sediments.

Lumbriculus variegatus and Diporeia spp. were exposed to two contaminant pairs 3H-benzo[a]pyrene (BaP) and 14C-2,4,5,2'4',5'-hexachlorobiphenyl (HCBP), and 3H-pyrene (PY) and 14C-3,4,3',4'-tetrachlorobiphenyl (TCBP) sorbed to each of seven field-collected sediments of varying composition. Toxicokinetic coefficients, bioaccumulation factors (BAF), and biota-sediment accumulation factors ([BSAF], BAF normalized to the organism lipid content and sediment organic carbon content) were determined. The contaminant desorption rates from sediments were measured with a Tenax resin extraction technique. The desorption rate constants for rapid, slow, and very slow contaminant compartments and the fractions of contaminant in each compartment were compared with the toxicokinetic coefficients, BAF, and BSAF to describe contaminant behavior across sediments, among compounds, and between species. The best description of the bioavailability was the log BSAF regressed against the fraction rapidly desorbed (F(rapid)) across all sediments and compounds for both species, r2 = 0.67 and 0.66 for L. variegatus and Diporeia, respectively. Use of a calculated fraction desorbed in 6 h or 24 h, which has been suggested as a surrogate for F(rapid,) did not produce as predictive a regression because of uneven desorption in a fixed duration for each compound among the sediments. Thus, F(rapid)provided a good surrogate for the bioavailability of the sediment-sorbed contaminant as represented by BSAF across seven sediments and four compounds with predictions within a factor of approximately two of the measured value.

Tests of bioaccumulation models for polychlorinated biphenyl compounds: a study of young-of-the-year bluefish in the Hudson River estuary, USA.

A field-based study regarding uptake of polychlorinated biphenyl compounds (PCBs) by young-of-the-year (YOY) bluefish (Pomatomus saltatrix) was initiated to test a steady-state model of bioaccumulation and trophic transfer in a rapidly growing fish. Determination of prey composition as well as size-dependent growth and specific consumption rates for YOY bluefish from separate field and laboratory studies enabled the input of these species-specific parameters into the model. Furthermore, the time and duration of the exposure of YOY bluefish to dissolved PCBs from a well-characterized system (Hudson River, USA) was well known. Patterns of accumulation of individual PCB congeners differed relative to the accumulation of total PCBs, with the greatest net accumulation occurring for the higher-molecular-weight congeners. Comparison of lipid-normalized bioaccumulation factors (BAFs) with the octanol-water partition coefficients of individual PCB congeners revealed bluefish to be above the BAFs predicted by lipid-based equilibrium partitioning, suggesting that uptake from food is an important source of PCBs in YOY bluefish. Comparison of measured BAFs with values predicted by a steady-state, food-chain model showed good first-order agreement.
