Functional interactions of the suprachiasmatic nuclei and the hippocampus during the acquisition and recovery of a conditioned reflex to time in rats.

Electrolytic destruction of the dorsal hippocampus prevented the acquisition of a conditioned reflex to time regardless of the volume of brain damage. A similar effect was obtained in rats with combined lesions consisting of damage to the suprachiasmatic nuclei of the hypothalamus and hippocampectomy (in the same animal). Denucleation (without hippocampectomy), conversely, accelerated the acquisition of this type of time-related behavior. Lesions to the hippocampus had no effect on the recovery of the conditioned reflex to time when this had been fixed before surgery in denucleated rats. It is proposed that the suprachiasmatic nuclei and hippocampus have simultaneous roles in organizing the endogenous counting of short time periods.

[Responses of neurons in the secondary somatosensory zone of the cat cortex during elaboration of a defensive conditioned reflex].

The neuronal responses of the somatosensory secondary cortex during the process of formation and extinction of a conditioned defence reflex to sound stimulus were studied in chronic experimentss on cats. In 21 out of 28 examined neurons a change in activity took place during the formation of a conditioned reflex as a response to the conditional signal. Two types of conditioned reflex responses were observed: excitatory and inhibitory ones. Most of the neurons excited by the conditioned stimulus responded during the first 50 ms, preceding the conditioned movement by 80-100 ms. Significant variability of neuronal responses was observed during formation of the conditioned reflex. After stabilization the neuronal responses to the conditioned stimulus acquired a stable from. During extinction of the neuronal conditioned activity a decrease of the discharge frequency in the excitatory neuronal response and disinhibition of the activity in the inhibitory responses were found.

[Integration of conditioned reflex systems in intact and callosotomized dogs].

Characteristics of integrative activity were studied by Kupalov's method of situational conditioned reflexes in 2 groups of dogs with complete section of the callosal body and in group of intact animals. The dogs of the first group were callosotomized before the formation of the conditioned reflex systems, the dogs of the second group--after their stabilization. The level of the reflex systems integration in the first group was considerably lower than in the second one. This testifies to a special role of the callosal body in the initial period of formation of complex conditioned systems. The callosotomized dogs developed inertia of nervous processes which was manifested in slowing down of formation and of adaptive realization of reactions during integration of various reflex systems. It was shown that the integrative activity is provided for by the interaction of cerebral hemispheres at different structural levels.

A comparative analysis of the conditional reflex discoveries of Pavlov and Twitmyer, and the birth of a paradigm.

Research on conditional reflex (CR) in Pavlov's Physiological Laboratory has preceded Twitmyer's work on conditioning at the University of Pennsylvania by 3 or 4 years. The events in Pavlov's laboratory lead toward the postulation of a new paradigm that rejected the Cartesians conceptualization of the reflex as a mechanistic response to stimuli by replacing it with the Darwinian notion of the organism's adaptation to the environmental conditions. The Pavlovian paradigm rejected the Wundtian method in favor of the objective, conditional reflex method.

[Conditioned reflex principle in the system of neurosciences: concepts and prospects].

The role played by Pavlov's principle of conditioned reflexes in neurosciences is discussed. The problem of interneuronal connections is shown to be key in the system of the sciences. The concept of formation of interneuronal connections underlying the conditioned reflex and long-term memory has been elaborated at the Department of Physiology of the Institute of Experimental Medicine named after Pavlov. Possible mechanisms of associations between the neural memory and cell genome are analysed and the problem of the engram localization and storage are discussed. Special attention is devoted to the role played by the subjective experience, mainly emotions, in the organization of the higher brain functions. The extensive experimental evidence gained by the authors during the recent 10-12 years is generalized.

[Description of conditioned reflex elaboration in cats in response to electric stimulation of the hippocampal formation].

In six cats with chronically implanted brain electrodes conditioned running to the feeding trough was elaborated in response to electrical stimulation of the ventral hippocampal formation (VHF), which at first produced inhibition of running. The stages of conditioning were as follows: 1) inhibition of conditioned activity; 2) replacement of inhibition by more frequent runnings--generalization of the conditioned reflex; 3) enhancement of signal significance of VHF electrical stimulation and subsequent decrease of intersignal reactions. Conditioned reflex to electrical stimulation of CA1 field was elaborated slower than that to electrical stimulation of other VHF points. At the beginning of conditioning a periodic decrease of probability of conditioned reactions manifestation was observed, which is estimated as an additional characteristic of the hippocampus activity.

Elaboration of a conditioned reflex in a single experiment with simultaneous recording of neural activity.

In 18 of 20 experiments with click CS, and in 5 of 10 experiments with flash CS, we have elaborated a conditioned EMG response in a single session (less than 60 pairings). The US was direct electrical stimulation of the rabbit's motor cortex that evoked a movement of the forelimb. Electrical stimulation of the lateral hypothalamus was used as reinforcement. Parameters for this reinforcement were chosen to evoke a feeding reaction or self-stimulation in the freely behaving animal. The elaborated EMG response satisfied most of the required characteristics of a conditioned reflex. These included spontaneous recovery after extinction, savings of long duration, specificity to stimulus pairing, and weak efferent and afferent generalization. In most experiments with click CS, the elaborated phasic response had an amplitude of 0.1-6 mV with a latency of 12-16 msec. In form and latency the conditioned response was similar to the unconditioned startle reaction of the same animal under chloralose anesthesia, or to its unanesthetized response to a loud sound. However, the conditioned response differed from the startle reaction in that it was localized. Extracellular recordings of 2-5 neurons were simultaneously made from sensory motor cortex near the point at which the US was applied. In 7 experiments 17 neurons were followed through the entire cycle of elaboration and extinction of the conditioned response. Seven neurons showed a statistically significant (P less than 0.05) increase of the response to CS during conditioning. Latencies were 20-140 msec. Interactions between neurons were studied by computing cross-correlograms and joint PST scatter diagrams. These measures were less informative than we had hoped because of the low level of spontaneous and evoked activity, and because of the small numbers of stimulus presentations that were needed for elaboration and extinction of the conditioned reflex. In isolated presentations of the CS after elaboration of the CR, we sometimes observed neural responses to click with a latency less than 6 msec. We propose that at least one of the pathways involved in the localized conditioned startle reflex reported here goes through the sensory motor cortex.

[Peculiarities of unconditioned and conditioned reflex stimuli effects on secretory function of gastric glands in elderly and senile people (author's transl)].

For understanding age peculiarities of gastric secretory function in aging.tory function in aging.tory function regulation, the investigation of nervous influences on gastric secretion is of importance. With this end in view the unconditioned reflex stimulus- the tube with rubber balloon, containing 10 ml of water and conditioned reflex stimulus - food stimulus were employed. The fractional gastric intubation was carried out. The gastric secretion in response to mechanical stimulus was investigated in 125 persons aged 60-89 and in 40 persons aged 20-25 (control group). In response to conditioned reflex stimulus it was studied in 30 persons aged 60-89 and 10 persons aged 20-35 (control group). With aging the response of gastric glands to unconditioned and conditioned stimuli was found to decrease. The amount of secretion, hydrochloric acid, pepsin, and gastromuco-protein were decreased, the secretory effect being more slowly developed, the torpid secretion type being observed. [Food getting conditioned reflexes in hedgehogs].

The object of the study were the characteristics of formation of complex food-procuring conditioned reflexes in hedgehogs to visual stimuli of different complexity, to a simultaneous complex, and the interaction of visual and auditory sensory modalities. It has been shown that hedgehogs are capable of learning to choose one of the three simultaneously presented visual stimuli and perform a sequence of complex motor behavioral acts in stage-by-stage learning. Conditioned reflexes to stimuli of different modality were elaborated with difficulty only at their interaction. Higher nervous activity of hedgehogs (insectivorous) is characterized by primitive organization, weakness of inertness of nervous processes and limited integrative possibilities.

The role of emotions in the formation of instrumental conditioned reflex.

The recording of vegetative parameters of emotional stress in humane and in dogs during elaboration of instrumental conditioned reflexes showed that such stress is a function of two major factors: (i) the value of the actual need (drive) and (ii) the estimation by the brain of the probability (possibility) of its satisfaction. Comparison of the need with the possibility of its satisfaction activates the mechanisms of positive (maximized by the subject) or negative (minimized) emotions; which serve as the direct reinforcement of instrumental conditioned reflexes. Experiments show that needs and emotions have an independent morphological substratum. The hypothalamus and amygdala must be intact in order to single out the need that is dominant at a given moment and is first to be satisfied. Estimation of the probability of need's satisfaction (probability of reinforcement)is the function of the frontal areas in the neocortex and hippocampus. These four brain structures play a decisive role in the genesis of emotional states and in the organization of behavior.

[The character of the effect of triftazin, carbidine and aminazine on defensive conditioned reflexes].

The author studied in chronical experiment on the rats the influence of triphtazine, karbidine and aminazine on the conditioned reflexes with the irritation of the mesencephalitio part of the reticular formation. The obtained data confirm the inhibitive influence of aminazine on the mesencephalitic part of the reticular formation. Triphtazine and karbidine, alike aminazine, prevent the inhibition of the defensive conditioned reflexes due to the irritation of the mesencephalitis part of the reticular formation testifies to the fact of their inhibitive influence on these structures.

[The possibility of elaborating conditioned reflexes on the basis of emotional reactions evoked by electrical stimulation of limbic structures].

In experiments on cats with chronically implanted electrodes a conditioned stimulus (200 c/s tone) was paired with electrical stimulation of limbic structures, which produces pronounced emotional behaviour: defensive, aggressive, alimentary or drinking reactions. It has been shown that conditioned reactions are not elaborated on the basis of electrical stimulation, which activates alimentary, drinking or aggressive behaviour. Neither is a conditioned reflex formed to the combination of sound and stimulation of the brain structures, which inhibits alimentary behaviour. A distinct conditioned fear reaction sets in as a result of combination of sound and electrical stimulation of brain structures, which evoke defensive behaviour. The data obtained are analyzed in the light of the significance of natural requirements of the organism for the elaboration of conditioned reflexes.

[Fromation of forward and reverse connections during elaboration of a classical and instrumental acid-defense conditioned reflex].

During elaboration of a classical acid defensive conditioned reflex to passive lifting of a foreleg in dogs, the reinforcing stimulus--acid elicited an extension of the leg via backward connection. At the initial stage of elaboration of an instrumental avoidance reflex, the reinforcing stimulus called forth an extension of the leg, whereas at the stage of a stable reflex it produced a flexion.

[Effect of the type of reinforcement on conditioned reflex activity in male and female white rats].

On 24 males and 26 females of white rats of a mixed line, a study was carried out of the speed of formation and consolidation of instrumental drinking and defensive conditioned reflexes, elaboration of differentiation, changes in conditioned activity by the use of stochastic stereotype and by changing the meanings of the conditioned signals to opposite. Conditioned reflexes were elaborated more rapidly in female rats at drinking reinforcement and in male rats--at the defensive one. Differentiations were elaborated more rapidly and were better expressed in females than in males at both types of reinforcement.
