Studies on transmissible gastroenteritis of swine. 3. The effect of selective inhibitors of viral replication on a cytopathogenic virus from transmissible gastroenteritis.

The effect on the plaque production of the Purdue strain of cytopathogenic virus from transmissible gastroenteritis of swine by 5-bromo-2'-deoxyuridine (BUDR), 5-iodo-2'-deoxyuridine (IUDR), actinomycin-D, puromycin, and amantadine-HCI (Symmetral) has been studied.Amantadine-HCI reduced the plaque-forming units of virus per ml by approximately 98%. Puromycin prevented almost all virus reproduction while actinomycin-D caused approximately a 22% reduction. Both IUDR and BUDR produced approximately a 20% increase in plaque-forming units of virus per ml. Swine testis cells stained with acridine orange early in the course of infection contained brick-red particles in the cytoplasm, indicative of a ribonucleic acid (RNA) type virus.

A live vaccine against tick-borne encephalitis: integrated studies. I. Basic properties and behaviour of the E5 "14" Clone (Langat virus).

The course of plaque segregation from the Langat E5 strain of the E5 "14" clone is described. The virus, displaying an ic+ sc s plus or minus t e u s character, reproduced at 35 and 37, but not at 39 degree C. In subcutaneously (sc) inoculated monkeys, viraemia lasted for 2--4 days without detectable central nervous system (CNS) involvement. In sc inoculated 8--10 g mice, the infection was manifested only by a transient trace viraemia, but caused a marked resistance against challenge with virulent tick-borne encephalitis (TE) viruses, protection indices being 5.7--6.7. Pathogenetic investigations in challenged, live virus-immunized mice showed no signs of a marked productive infection except of a booster effect.

Comparison of the effects of 3'-azidothymidine with those of neutralizing antibodies on simian immunodeficiency virus infection in macrophages.

We previously showed that simian immunodeficiency virus-infected macaque macrophages contacting uninfected CD4+ lymphocytes caused extensive cell fusion and synthesis of phlogistic cytokines. In this study, macaque macrophage cultures inoculated with SIVmac251 and treated simultaneously with 10 microM 3'-azidothymidine (AZT) became infected and produced small amounts of viral antigen (p27) but failed to fuse with CD4+ CEM174 cells. When AZT was added 1 to 3 days after virus inoculation, the infected cells caused fusion and the release of tumor necrosis factor and produced increasing amounts of p27. In contrast, neutralizing antibodies prevented infection when added at the time of virus inoculation, and they were much more effective than AZT in limiting virus replication, fusion cytopathic effect, and cytokine production when added up to 3 days postinoculation. Neutralizing antibodies may be more effective than AZT in reducing the virus load in the macrophage population and in preventing both cell fusion and the production of potentially pathogenic cytokines.

Characterization of the TO strains of Theiler's mouse encephalomyelitis viruses.

Theiler's mouse encephalomyelitis virus isolates from the central nervous systems of spontaneously paralyzed mice and stools of asymptomatic mice resemble Theiler's original virus isolates. In this study four such strains were adapted by blind subpassage to replicate and to produce cytopathic effect in cell culture. These viruses were then found to be closely related to each other and to GDVII virus by cross-neutralization and to form small plaques. Bovine serum was found to contain cross-reacting antibodies to these viruses.

Detection and serotyping of herpes simplex virus in MRC-5 cells by use of centrifugation and monoclonal antibodies 16 h postinoculation.

Monoclonal antibodies (Syva Co., Palo Alto, Calif.) were used for the detection and serotyping of herpes simplex virus (HSV) isolates by immunofluorescence 16 h after inoculation of MRC-5 monolayers in 3.7-ml shell vials and after low-speed centrifugation. A total of 119 specimens were inoculated into conventional tube cell cultures and shell vials. Of 98 specimens inoculated on the same day of receipt in the laboratory (fresh specimens), all 23 (23.5%) HSV-positive specimens were identified by serotype in 16 h in shell vials by immunofluorescence, whereas only 8 of 23 HSV-positive specimens (34.8%) produced cytopathic effects in conventional tube cell cultures in this time period. Similarly, of 21 original specimen extracts previously determined to be culture positive for HSV (stored specimens), all were detected and serotyped by the immunofluorescence test with monoclonal antibodies 16 h postinoculation compared with the recognition of only 8 of these isolates (38.1%) by cytopathic effect that soon. This technique of centrifugal inoculation of HSV in shell vials containing MRC-5 cells permitted detection of this virus in all positive specimens with serotype determination within 16 h postinoculation.

[Intratypical antigenic differentiation of poliovirus strains circulating in Mali by means of cross-adsorbed sera].

The intratypic antigenic differentiation of poliovirus strains isolated from sick or healthy children in Mali in 1975-1976 was performed. The method is based on the use of cross-adsorbed sera in the neutralization tests in which the virus is titrated by the plaque procedure or by the cytopathic effect. All the examined strains of serologic types I, II, and III were distinguished by the antigenic structure from the vaccine strains and were classified as "wild". Quantitative differences in the antigenic structure of type I strains isolated in different towns of Mali were found.
