Interactive effects of saffron with garlic and curcumin against cyclophosphamide induced genotoxicity in mice.

Saffron is a well-known spice and food colorant commonly consumed in different parts of the world. Recently, much attention has been focused on the biological and medicinal properties of saffron. In the present study the interactive effects of saffron with two commonly consumed dietary agents, garlic and curcumin was evaluated for anti-genotoxic effects against cyclophosphamide (CPH) in the mouse bone marrow micronucleus test. Experimental animals were orally pretreated with saffron (100 mg/kg body weight), garlic (250 mg/kg body weight) and curcumin (10 mg/kg body weight), either alone or in combination for five consecutive days, 2h prior to the administration of CPH. Maximum reduction in the frequencies of micronucleated polychromatic erythrocytes (Mn PCEs) induced by CPH was observed when all the three test compounds were administered together. Furthermore, the protective effects were more pronounced in the garlic-administered groups compared to curcumin and/or saffron administered groups.

[Comparison on ingredients between cultured cell and cultivated Saffron pistils].

OBJECTIVE: To compare the differences of active ingredients between tissue cultured cells and cultivated saffron pistils. METHOD: The experiment was carried out by the Fourier transform infrared spectroscopy (FTIR) spectra and high performance liquid chromatography (HPLC). RESULT: The data indicated that the species and contents active ingredients in saffron pistils from different places were different. The species of active ingredients in tissue cultured cells are less than those in cultivated saffron pistils. However, the quantity of crocin A, which showed good anticancer effect, is 2-3 times more than that in cultivated saffron pistils. CONCLUSION: The active ingredients of the tissue cultured cells are similar to those of saffron pistils, but their contents are different. Therefore, the tissue cultured cells can only be the part-substitutes of cultivated saffron pistils.

[Protective effect of crocetin on primary culture of cardiac myocyte treated with noradrenaline in vitro].

AIM: To investigate the cardio-protective effect of crocetin on primary culture of cardiac myocyte treated with noradrenaline. METHODS: After adding crocetin, the primary culture of cardiac myocyte was injured by 1.0 micromol x L(-1) noradrenaline. The activity of lactic dehydrogenase (LDH), mitochondrion succinic dehydrogenase (MSDH) and ATPase were assayed. The mitochondrion membrane potential was detected by Rh123. The percentage of cardiac myocyte apoptosis was observed by flow cytometry. RESULTS: Crocetin significantly decreased the activity of LDH in culture supernatant, increased the activity of MSDH, ATPase (Na+-K+ ATPase, Ca2+ ATPase) and mitochondrion membrane potential. CONCLUSION: Crocetin could alleviate the disturbance of energy metabolism and decrease the percentage of apoptosis of cardiac myocyte treated with noradrenaline.

[Determination of crocin-1 in rabbit plasma and the pharmacokinetics by RP-HPLC].

AIM: To develop a sensitive and specific HPLC method for the determination of crocin-1 in rabbit plasma, and study the pharmacokinetics in rabbit. METHODS: After i.v. administration of crocin-1 in rabbit, the sample could be well purified after precipitation of protein with methanol. Waters spherisorb C18 column (250 mm x 4.6 mm ID, 5 microm) was used and the mobile phase consisted of methanol-acetonitrile-1% aqueous acetic acid (15:10:50) at the flow rate of 1 mL x min(-1), and the UV detection wave length was 440 nm. The column temperature is 25 degrees C. RESULTS: The calibration curve was linear (r = 0.9997) in the range from 0.86 to 27.54 mg x L(-1) for crocin-1. The lowest detectable concentration of crocin-1 was 0.42 mg x L(-1). After given i.v. crocin-1 in rabbit, the concentration-time curves of crocin-1 was shown to fit two-compartment open model. CONCLUSION: This method is proved to be rapid, precise and reliable enough to be applied to the pharmacokinetics studies of crocin-1 in rabbit.

[Study of the effect of crocin on rat experimental hyperlipemia and the underlying mechanisms].

OBJECTIVE: To study the effect of crocin on rat experimental hyperlipemia and its mechanisms. METHOD: Hyperlipemia model was established by feeding heavy cholesterol for 2 months and the effect of crocin on blood lipid in experimental hyperlipemia rats was observed. Aortic smooth muscle cells were cultured in different culture media and proliferation was measured by MTT assay. Western blotting was used to detect the effect of crocin on phosphorylation of p38 MAPK. RESULT: Crocin not only decreased greatly the content of cholesterol, triglyceride and density lipoprotein in blood, but also increased the content of high density lipoprotein. In addition, the proliferation of smooth muscle cells and the activation of p38MAPK were inhibited by Crocin. CONCLUSION: Crocin prevents atherosclerosis in hyperlipemia, which may be mediated by the inhibition of both proliferation of smooth muscle cells and activation of p38MAPK.

[Adsorption behaviors of chitosan and the analysis of FTIR spectra].

This paper studied the adsorption behavior of lab-made chitosan for acid dyeing waste water and basic dyeing waste waters, based on the effects of adsorbing time, deacetylation degree of chitosan, and pH of waste water. The results showed that the adsorption of basic fuchsin and saffron by the chitosan almost reached equilibrium state in 20 min. The adsorption capacities of the chitosan for dyes was comparable with activated carbon. Moreover, its dosages were only 2/3 of the latter. The adsorption process for Acid Red presented a dynamics character of first order reaction. FTIR spectroscopy was used to analyze the adsorption mechanisms of chitosan for different kinds of dyes. It was found that a great deal of hydroxide radicals in the chitosan molecule participated in the adsorption of basic fuchsin and saffron.
