Reducing recurrence and costs in superficial bladder cancer: preclinical evaluation of osmotic cytolysis by distilled water vs. mitomycin.

Transurethral resection followed by instillation of chemotherapeutic agents such as mitomycin is considered as standard therapy in recurrent superficial bladder cancer. However, incidence of bladder cancer is increasing, and contrariwise resources to finance the health care systems are decreasing. Therefore, effective alternatives to expensive chemotherapeutics are necessary. Recurrence of bladder cancer after transurethral resection is mostly promoted by reimplantation of dissolved tumour cells which are therapeutical purpose of any intravesically instillated agent. Monolayer cultures of human RT112, RT4, T24 and TCC SUP bladder cancer cells were incubated and exposed to mitomycin or distilled water. Cell survival was determined by microculture tetrazolium assay. Distilled water led to significant cytolysis in all tumour cells. This effect was comparable to exposition to mitomycin. Distilled water and mitomycin have comparable in vitro effects in bladder cancer cells. These findings have to be substantiated by an animal model emphasising the aspect of larger tumour cell compounds or possible damage to healthy bladder tissue.

An outbreak of endophthalmitis after extracapsular cataract surgery probably caused by endotoxin contaminated distilled water used to dissolve acetylcholine.

AIM: To study possible causes of an outbreak of severe endophthalmitis after planned extracapsular cataract surgery in Medan, Indonesia. METHODS: In a 3 week period in November 2001, 17 of 43 patients developed signs of endophthalmitis after planned extracapsular cataract surgery. A search for possible causes was undertaken 4 months later. RESULTS: In autoclaved stored distilled water used to dissolve acetylcholine (used in 16 of 17 patients with endophthalmitis) a high amount of endotoxin was detected in a human blood essay, as well as a small number of non-typeable Pseudomonas spp. CONCLUSIONS: These findings suggest that distilled water used as solvent for acetylcholine was responsible for this outbreak of endophthalmitis. As a consequence, we now rely on solvents that are regularly checked for impurities such as an intravenous infusion fluid, rather than on vials with distilled water that is presumed to be sterile and kept for some time.

Local cellular and humoral responses to antigenic and distilled water challenge in subjects with allergic rhinitis.

We studied nasal responses to allergen, saline, and distilled water aerosol in 7 subjects with allergic rhinitis to determine whether they caused a similar release of mediators and influx of inflammatory cells into the nasal lumen. The subjects were challenged first with allergen and then with aerosols of distilled water and of normal saline in random order on separate days. We measured nasal airway resistance (Rn) by posterior rhinomanometry, measured the concentrations of protein, histamine, leukotrienes (LT) B4, C4, and D4, and platelet-activating factor (PAF-acether) and performed total and differential cell counts in nasal lavage fluid obtained before and after each challenge. Allergen challenge provoked a 2-fold or greater increase in nasal airway resistance in all subjects (mean increase = 12.2-fold). This response was associated with significant increases in protein, histamine, LTB4, and sulfidopeptide leukotrienes. 2-lyso-PAF-acether increased significantly, indicating activation of phospholipase A2, but PAF-acether was detected in only one subject. The total cell count increased from 55 +/- 44 x 10(3)/ml to 200 +/- 168 x 10(3)/ml; polymorphonuclear neutrophils increased from 11 +/- 22 x 10(3)/ml to 108 +/- 96 x 10(3)/ml, and eosinophils increased from 1.3 +/- 1.8 x 10(3)/ml to 10.6 +/- 15.3 x 10(3)/ml (p less than 0.05). Saline insufflation provoked insignificant changes in mean Rn, in the levels of protein and all inflammatory mediators, and in the number and types of cells in nasal lavage fluid. Distilled water insufflation also provoked an insignificant increase in mean Rn, but there was a 14-fold increase in one subject.(ABSTRACT TRUNCATED AT 250 WORDS)

Comparative acute toxicity of shale and petroleum derived distillates.

In anticipation of the commercialization of its shale oil retorting and upgrading process, Unocal Corp. conducted a testing program aimed at better defining potential health impacts of a shale industry. Acute toxicity studies using rats and rabbits compared the effects of naphtha, Jet-A, JP-4, diesel and "residual" distillate fractions of both petroleum derived crude oils and hydrotreated shale oil. No differences in the acute oral (greater than 5 g/kg LD50) and dermal (greater than 2 g/kg LD50) toxicities were noted between the shale and petroleum derived distillates and none of the samples were more than mildly irritating to the eyes. Shale and petroleum products caused similar degrees of mild to moderate skin irritation. None of the materials produced sensitization reactions. The LC50 after acute inhalation exposure to Jet-A, shale naphtha, (greater than 5 mg/L) and JP-4 distillate fractions of petroleum and shale oils was greater than 5 mg/L. The LC50 of petroleum naphtha (greater than 4.8 mg/L) and raw shale oil (greater than 3.95 mg/L) also indicated low toxicity. Results demonstrate that shale oil products are of low acute toxicity, mild to moderately irritating and similar to their petroleum counterparts. The results further demonstrate that hydrotreatment reduces the irritancy of raw shale oil.

Possible remediation of dioxin-polluted soil by steam distillation.

2,7-Dichlorodibenzo-p-dioxin (DCDD) was found to evaporate easily with water vapor from a heated solution. Steam distillation was also effective for the removal of DCDD from DCDD-applied soil; its concentration (250 microg/50g soil) in the original soil decreased to less than 5% after steam distillation for only 20 min. Actual dioxin-polluted soil in Tokorozawa City was partially decontaminated using the same method. These results suggest that steam distillation could be a new remedial method for soils contaminated with persistent environmental pollutants, such as dioxins and polychlorinated biphenyls.

The coupling of supercritical fluid chromatography and field ionization time-of-flight high-resolution mass spectrometry for rapid and quantitative analysis of petroleum middle distillates.

We report the first coupling of supercritical fluid chromatography (SFC) with field ionization time-of-flight high-resolution mass spectrometry (FI-ToF HRMS), in parallel with ultraviolet (UV) detection and flame ionization detection (FID), for rapid and quantitative analysis of petroleum middle distillates. SFC separates petroleum middle distillates into saturates and 1- to 3-ring aromatics. FI generates molecular ions for hydrocarbon species eluted from the SFC. The high resolution and exact mass measurements by ToF mass spectrometry provide elemental compositions of the molecules in the petroleum product. The amounts of saturates and aromatic ring types were quantified using the parallel SFC-FID assisted by SFC-UV. With a proper carbon-number calibration, the detailed composition of the petroleum middle distillate was rapidly determined.

Determinants of airway response to challenge with distilled water in a population sample of children aged 7 to 10 years old.

We challenged 446 schoolchildren and measured the percent decrease in FEV1 following 10 min of tidal inhalation of UNDW. Assessment tools for respiratory symptoms and atopy were questionnaire and skin testing, respectively. A previous diagnosis of asthma was most strongly associated with a positive airway response (defined as a fall in FEV1 greater than or equal to 10 percent. A positive response was also associated with atopy, presence of cough, cough during night, or any respiratory symptom. A child's age and the prechallenge FEF75% also explained response to distilled water indicating less responsiveness for older children and those with relative greater airway diameter. For the previous diagnosis of asthma, a positive distilled water challenge test had a sensitivity of 36 percent and a specificity of 92 percent. We conclude that a significant relationship between airway response to distilled water, asthma and symptoms suggestive for asthma exists for a childhood population sample.

Measurement of rheology of distiller's grain slurries using a helical impeller viscometer.

Current research is focused on developing a process to convert the cellulose and hemicellulose in distiller's grains into fermentable sugars, increasing both ethanol yield and the amount of protein in the remaining solid product. The rheologic properties of distiller's grain slurries were determined for concentrations of 21, 23, and 25%. Distiller's grain slurries are non-Newtonian, heterogeneous fluids subject to particle settling. Traditional methods of viscosity measurement, such as cone-and-plate and concentric cylinder viscometers, are not adequate for these fluids. A helical impeller viscometer was employed to measure impeller torque over a range of rotational speeds. Newtonian and non-Newtonian calibration fluids were utilized to obtain constants that relate shear stresses and shear rates to the experimental data. The Newtonian impeller constant, c, was 151; the non-Newtonian shear rate constant, k, was 10.30. Regression analysis of experimental data was utilized for comparison to power law, Herschel-Bulkley, and Casson viscosity models with regression coefficients exceeding 0.99 in all cases.

Molecular distillation: a powerful technology for obtaining tocopherols from soya sludge.

Molecular distillation was studied for the separation of tocopherols from soya sludge, both experimentally and by simulation, under different operating conditions, with good agreement. Evaporator temperatures varied from 100 degrees C to 160 degrees C and feed flow rates ranged from 0.1 to 0.8 kg/h. The process pressure was maintained at 10(-6) bar, the feed temperature at 50 degrees C, the condenser temperature at 60 degrees C, and the stirring at 350 rpm. For each process condition, samples of both streams (distillate and residue) were collected and stored at -18 degrees C before tocopherols analyses. Owing to the differences between molecular weights and vapor pressures of free fatty acids and tocopherols, tocopherols preferentially remained in the residue at evaporator temperatures of 100 degrees C and 120 degrees C, whereas for higher temperatures (140 degrees C and 160 degrees C) and lower feed flow rate, tocopherols tended to migrate to the distillate stream.

Continuous wine making by gamma-alumina-supported biocatalyst: quality of the wine and distillates.

The main objective of the present work was the removal of aluminum from wines produced by gamma-alumina-supported yeast cells. Reagents such as Na2CO3, NH4OH, albumin, and Ca(OH)2 were used. Calcium in the presence of albumin was effective, whereas other reagents were not so effective. Because of the improved aroma and taste of distillates produced by gamma-alumina-supported biocatalyst, volatile byproducts of distillates were analyzed. They were also assessed by sensory tests. Methanol, acetaldehyde, ethyl acetate, propanol-1, isobutyl alcohol, and amyl alcohols were determined in distillates. It was noted that the amounts of higher alcohols and amyl alcohols decreased as the temperature of fermentation dropped, leading to a product of improved quality and reduced toxicity.

Comparison of the volatile composition of Stachys persica Gmel. and Stachys byzantina C. Koch. oils obtained by hydrodistillation and steam distillation.

The oils obtained by hydrodistillation and steam distillation of the aerial parts of Stachys persica Gmel. and Stachys byzantina C. Koch grown in Iran were analyzed by GC/MS. The essential oil obtained by hydrodistillation of the aerial parts of S. persica was characterized by a high amount of non-terpenoid components of which methyllinoleate (27.7%), hexadecanoic acid (9.8%) and 6,10,14-trimethyl-2-pentadecanone (9.2%) were the major constituents, whereas the steam distilled oil of the plant contained hexadecanoic acid (27.2%), carvacrol (9.4%) and eugenol (5.2%). Both hydrodistilled and steam distilled essential oils of the aerial parts of S. byzantina were rich in sesquiterpenes such as a-copaene (16.6% and 10.4%), spathulenol (16.1% and 18.5%) and beta-caryophyllene (14.3% and 13.5%), respectively.

Effects of season and inclusion of corn distillers dried grains with solubles in creep feed on intake, microbial protein synthesis and efficiency, ruminal fermentation, digestion, and performance of nursing calves grazing native range in southeastern North Dakota.

Nine ruminally and duodenally cannulated (145 +/- 21 kg of initial BW; Exp. 1) and sixteen intact (181 +/- 36 kg of initial BW; Exp. 2), commercial, Angus, nursing, steer calves were used to evaluate the effects of advancing season and corn distillers dried grains with solubles in creep feed on intake, digestion, microbial efficiency, ruminal fermentation, and performance while grazing native rangeland. Calves were assigned to 1 of 2 treatments: a supplement containing 41% soybean meal, 26.25% wheat middlings, 26.25% soybean hulls, 5% molasses, and 1.5% limestone (control) or a supplement containing 50% corn distillers dried grains with solubles, 14.25% wheat middlings, 14.25% soybean hulls, 14% soybean meal, 5% molasses, and 1.5% limestone (CDDGS). Calves were offered supplement individually (0.45% of BW) once daily. Three 15-d collection periods occurred in June, July, and August. In Exp. 1, there were no differences in OM intake, or OM, N, NDF, or ADF digestion between control calves and those fed CDDGS. Forage and total OM intake increased (P < 0.03), whereas OM digestion decreased (P < 0.01), with advancing season. Duodenal microbial N flow (g/d) was not affected (P = 0.50) by treatment and increased linearly (P = 0.003) as season progressed. Calves consuming CDDGS had decreased (P < 0.01) ruminal acetate:propionate ratio, increased (P < 0.01) molar proportion of butyrate, and decreased (P < 0.001) molar proportions of isobutyrate and isovalerate. In Exp. 2, supplement OM intake (% of BW) was less for CDDGS compared with control calves, but there were no differences in performance or subsequent carcass composition between treatments. Inclusion of 50% corn distillers dried grains with solubles in a creep supplement for nursing calves produced similar results compared with a control creep feed based on soybean meal, soybean hulls, and wheat middlings.

Individual characteristics of aggressive beer and distilled beverage drinkers.

Sixty male social drinkers completed the MMPI and a drinking history and attitude questionnaire. These individuals were then randomly divided into four groups, receiving either a distilled or brewed beverage, or two comparable placebos. Subjects in the alcohol groups became equally intoxicated (BAC = .71) and rated themselves as significantly more intoxicated than placebo subjects. All subjects then participated in a Buss aggression task. Aggression scales were related to the pretask personality, history, and attitude measures. Differences were found between factors descriptive of subjects who became aggressive after consuming or believing they had consumed a distilled versus a brewed beverage. In particular, a history of lack of positive alcohol effects and social maladjustment were typical of high-aggressive distilled alcohol and alcohol-placebo subjects, while the presence of family problems and anxiety typified high-aggressive beer and placebo-beer subjects.

Responses of dairy cows to additions of distillers dried grains with solubles in alfalfa-based diets.

Twenty-four Holstein cows in wk 3 through 12 of lactation were used to evaluate distillers dried grains with solubles as a protein source for alfalfa-based diets. Diets were formulated containing 0, 10.1, 20.8, or 31.5% distillers dried grains with solubles. Crude protein in these diets was 13.9, 16.0, 18.1, and 20.3%. Milk yield increased linearly with increasing dietary CP, whereas DMI was not affected. Dry matter intake averaged over 4% of BW for all treatments. Intake of undegraded intake protein was .93, 1.27, 1.63, and 1.97 kg/d. Yields of milk protein, casein, and lactose all increased linearly with increasing dietary CP. The proportion of milk N as whey N decreased, whereas N as NPN increased, with increasing dietary CP. Plasma urea N and essential and branched-chain AA increased linearly, and nonessential AA decreased linearly, with increasing dietary CP. Increasing the concentration of CP in the diet from 13.9 to 18.1% by the addition of distillers dried grains with solubles was beneficial to cows fed alfalfa-based diets in early lactation. Little additional benefit was observed by feeding greater than 18.1% dietary CP.

Corneoscleral cyst treated with distilled water injection.

To describe the first case of the treatment of a corneoscleral cyst by distilled water injection into a corneal cyst. The anterior wall of a cyst of the limbal communication was punctured with a surgical blade. Aspiration and irrigation of the contents of the cyst with a 27-gauge anterior chamber cannula were performed repeatedly, three times. Distilled water, instead of balanced salt solution, was injected into the collapsed cyst, and was then aspirated completely after 5 minutes. The injection and aspiration of distilled water was repeated once more. The scleral cyst was surgically excised. Twelve months after surgery, several small white granular opacities, presumably epithelial cell nests, were observed on the interface of the collapsed cyst cavity, but there was no recurrence of the cyst. The best spectacle-corrected visual acuity (BSCVA) was 1.0 with a correction of +1.25-2.00 X 45. No significant change in central corneal endothelial cell density was noted. We suggest that this simple technique may represent an alternative method for the management of corneal cysts, and may have less risk of developing a corneal opacity or causing other serious damage to surrounding tissues.

[Experimental studies on hyperthermic distilled water combining with saline and dextran for preventing intraperitoneal cancer recurrence due to neoplasm seeding].

In order to know the efficacy of hyperthermic distilled water combining with saline and dextran (DSD) to prevent intraperitoneal cancer from recurrence due to neoplasm seeding, we carried out the experimental studies, including an observation on inhibiting cancer cells from implantation in vivo by abdominal cavity lavation, cytotoxic test and attachment test in vitro. The results of the tests in vitro showed no viable tumor cell exposed to 45 degrees C distilled water for 20 minutes in the tissue culture wells, meanwhile, dextran-40 partially blocked attachment of B16-F10 cells to gelatinized wells. In vivo test, DSD effectively prevented neoplasm seeding of B16-F10 viable melanoma cells in C57BL/6j mouse abdominal cavity and had no obvious toxicity. The efficiency was 70%; when cisplatin was added, the efficiency was enhanced to 88.9% with slight toxicity. These data indicate that the local application of hyperthermic distilled water combined with saline and DSD may prevent intraperitoneal neoplasm from recurring.

Analysis with gas chromatography and mass spectrography of volatile components of Ligusticum wallichii Franch extracted by CO2 supercritical fluid extraction in combination with molecular distillation.

OBJECTIVE: To evaluate the possibility of using the technique of CO2 supercritical fluid extraction (SFE) combined with molecular distillation (MD) to extract, separate and purify the volatile components of Ligusticum wallichii Franch, a Chinese herbal medicine. METHODS: SFE was employed to extract the volatile components of Ligusticum wallichii Franch, followed by MD of the product. Analysis of the chemical constituents of the extract both before and after MD was performed with gas chromatography (GC) in conjunction with mass spectrography (MS). RESULTS: There were 45 kinds of chemical constituents in the extract of Ligusticum wallichii Franch yielded from SFE, among which 39 were left after distillation by MD. Changes also took place in the content of the constituents in the extract after distillation. CONCLUSIONS: SFE combined with MD, a technique better than simple SFE, can be used to extract, separate and purify the volatile components of Chinese herbs.

Determination of semivolatile organic compounds in environmental samples by gas chromatography/mass spectrometry after extraction by cyclic steam distillation.

A method was developed for the multiple determination of semivolatile organic compounds found in groundwater, river water, seawater, sediment, and soil. Forty standard compounds were determined: n-alkenes, cycloalkanes, aromatic hydrocarbons, and polycyclic aromatic hydrocarbons. The compounds were isolated from water and soil samples by using an essential oil distillator (cyclic steam distillator) with hexane as a solvent. The extract was cleaned by using a silica gel cartridge with an acetone-hexane solution. The compounds were determined by using a gas chromatograph/mass spectrometer with 12 stable isotope-labeled compounds (surrogate compounds). The efficiencies of recoveries from water samples were 80.0-106% for groundwater, 80.1-106% for river water, and 81.2-103% for seawater. The relative standard deviation (RSD) values were 2.05-16.0% for groundwater, 3.22-16.6% for river water, and 4.45-16.0% for seawater. The efficiencies of recoveries from sediment and soil were 71.5-96.4% and 70.1-99.8%, respectively. RSD values ranged from 2.27 to 16.0% for sediment and from 2.12 to 15.1% for soil. Adjustment of recovery efficiencies of standard compounds by using surrogate compounds gave more accurate values. The present study proved that an essential oil distillator provides satisfactory results for multiple determinations of the semivolatile compounds in environmental waters, sediment, and soil.
