An enzyme in extracts of the protistan Euglena gracilis splits the polysaccharide laminarin (beta-1:3-glucosan). Its optimal pH is 5.0, and it is activated by Mn(++) ions.
Mass cultures of Chlamydomonas moewusii have been synchronized by means of light-dark shifts. Division of at least 91 percent of the population was made to occur in 1/24 of the life cycle of the cells. The advantages of working with synchronized cultures of obligate autotrophs are discussed.
An enzyme in extracts of Euglena gracilis splits both purine and pyrimidine internucleotide bonds of ribonucleic acid. Its pH optimum is at 4.5; it is very heat-labile and is rather insensitive to inhibition by metal ions or sulfhydryl group reagents. A partial purification of the enzyme is described.
Thalidomide inhibits growth of some protozoa. This inhibition was counteracted by nicotinic acid, nicotinamide, nicotinamide adenine dinucleotide, and vitamin K(1). The mechanism of toxicity may be an interference of cellular oxidation. A protozoan test system is useful for studying the potential "side actions" of drugs in higher animals and man.

The ichthyotoxin of Prymnesium parvum is inactivated by visible (400 to 510 mmu) as well as by ultraviolet light (255 mmu). The changes in the absorption spectrum of purified (pigment-free) ichthyotoxin during this process indicate that different mechanisms may be involved in the inactivation by visible or ultraviolet light. Photoinactivation is not affected by the presence of cells, cell pigments, oxygen, or glutathione.
Six synthetic chelators were investigated in culture media for Chlamydomonas mundana. The effects on growth of chelator-iron ratios in the range 25 to 400 muM were studied in relation to Ca, heavy metal concentrations, and incubation time. Effects ranged from none through stimulation to inhibition. The most delicate balance of trace metal requirements with any given chelator was established by growth determinations. The behavior of chelators as they affect trace metals in culture media was best understood by consideration of absolute as well as relative stabilities of their metal chelates. Although all the chelators tested, when properly supplemented with microelements, allowed excellent growth, diethylenetriaminepentaacetic acid and diaminocyclohexanetetraacetic acid were least affected by additions of heavy metals and are recommended for use at 50 to 100 muM.
