Trabecular organization in mandibular osteodistraction in growing and maturing rats.

PURPOSE: Our goal was to investigate the trabecular organization of the distraction regenerate during various consolidation phases and as a result of various distraction rates. MATERIALS AND METHODS: One hundred ninety-two growing and maturing rats (96 each) received unilateral mandibular osteotomies and distraction device placement. They were randomly allocated into 4 distraction rate groups (0, 0.2, 0.4, and 0.6 mm/day for 5 days) after a 3-day latency. Eight rats from each rate group were sacrificed at early (10 days), mid (24 days) and late (38 days) consolidation time points. Harvested hemimandibles were embedded in micro-bed resin, sectioned sagittally at 10 mum thickness and stained using the Von Kossa method. The histologic images were captured and processed using Adobe Photoshop (Version 7.0; Adobe Systems Inc, San Jose, CA). Custom-made software (MacAzimuth; written by Prof. J.M. Rensberger, University of Washington) was further used to analyze the orientation (anisotropy and angle distribution) and mass (density and thickness) of trabecular structures in the regenerates. RESULTS: Trabecular orientation significantly differed at the mid-consolidation time point with less anisotropy ( P < or = .000 to .023) and narrower angular distribution ( P < or = .007 to .021). Trabecular mass increased significantly from early to mid-consolidation ( P < or = .000 to .002). Growing rats showed greater trabecular organization than maturing rats did at the same time points ( P < or = .000 to .009). The trabecular organization parameters were associated significantly with bone mineral density and microdensity ( P < or = .000 to .044). CONCLUSION: The peak change in orientation occurred around the mid-consolidation with a greater increase in trabecular mass. Growing rats showed a greater capacity for trabecular organization at earlier time points. However, distraction rate did not have an effect on trabecular organization. These results suggest that trabecular organization can be used as an important indicator to evaluate bone maturation and quality in the distraction regenerate.

Regeneration of the sagittal suture by GTR and its impact on growth of the cranial vault.

The aim of this study was to investigate the effect of bone grafting, suture transplantation, and guided tissue regeneration (GTR) treatment on healing of craniectomy defects involving the sagittal cranial suture and on the growth of the cranial vault. Fifty 4-week-old rats were included in the study. A 5.0 mm wide trephine defect was created with its midline corresponding to the sagittal cranial suture between the coronal and occipital cranial vault sutures. The animals were randomly allocated to five groups of 10 animals. Group A: The cranial defect was left untreated. Group B: An occipital bone graft was placed into the cranial defect. Group C: A cranial bone graft including a portion of the frontal suture was placed in the cranial defect. Group D: The cerebral and galeal aspect of the defect were covered with an e-PTFE membrane. Group E: The animals were sham-operated, no defect was created. In all animals, two gutta-percha points were placed demarcating the lateral borders of the parietal bones. Histological analysis at 4 months following surgery showed that the untreated cranial defects healed with fibrous connective tissue. The bone-grafted defects healed partially with bone and connective tissue in the periphery of the bone graft. The healing of suture-grafted defects resembled that of the bone-grafted defects, since the transplanted suture became completely obliterated with bone. The membrane-treated defects healed with bone and a suture-like tissue resembling the sagittal suture of the sham-operated controls. Cephalometric measurements demonstrated that membrane-treated and sham-operated control animals exhibited significantly more (P < 0.05) coronal growth (approximately 1.2 mm) than that of the remaining three groups of animals (approximately 0.7 mm). These findings were supported by the craniometry measurements demonstrating that sham-operated control and membrane-treated specimens presented significantly more cranial width than that of the remaining groups of animals (P < 0.05). It can be concluded that complete osseous healing, creation of a new sagittal suture, and increased cranial growth can be achieved by the treatment of craniectomy defects with the GTR technique.

Epidemiology of Oestrus ovis infection of sheep in Argentina's Western Pampas.

Seasonal population trends and effects of Oestrus ovis in naturally infected sheep were studied over 13 months, in the Western of the Pampeana region. At weaning, 140 growing lambs were randomly allocated to two groups: UG, untreated group and TG treated every 4 weeks with closantel (10 mg/kg). Successive Oestrus free tracer lambs (TL) by previous treatment (n = 65) were slaughtered after 20-30 exposition days for larval counts. Likewise, other group PL of 117 permanent untreated lambs was slaughtered from four to 17 months of age. Weighing and assessment of health signs of UG and TG and blood samples were monthly carried out. The prevalence of infection in permanent group varied from 33% to 100%. Mean number of larvae in PL was 6.1 with 3 L1, 1.4 L2 and 1.6 L3 during spring-summer and 17.9 with 16.9 L1, 0.5 L2 and 0.4 L3 during autumn-winter months. In PL the proportions of larvae in each of the different larval stages was similar during spring and summer, but a significant (P < 0.01) increase of L1 was detected during autumn and winter. The prevalence in tracer lambs was 100% during summer time and larvae were absent from 25-May to 25-October. Mean larval burdens of positive TL varied from 6.4 to one Oestrus and a significant peak (P < 0.05) of larvae was seen from December to March. Since March to November only L1 was recovered from TL. TG group showed a reduction in nasal discharge and in antibody ELISA levels, but no difference was observed in live weight gain between TG and UG. These results show a high prevalence during summer and that the perpetuation of Oestrus is ensured by an autumn period of arrested development and the over wintering larvae in the sheep heads.

Effects of nitric oxide synthase inhibition on angiotensin receptors and metabolism in the pregnant hypertensive rat.

Endothelial dysfunction and a consequent decrease in nitric oxide production have been implicated in the pathogenesis of pre-eclampsia. A prominent feature of the pre-eclamptic syndrome is a loss of the pregnancy-induced refractoriness to infused pressor agents, such as angiotensin. In this study, we sought to determine whether a decrease in nitric oxide production might be linked via changes in angiotensin II receptors and angiotensin II metabolism to changes in pressor sensitivity to infused angiotensin II. Pregnant and non-pregnant spontaneously hypertensive rats (SHRs) were randomly allocated to receive 5 mg x kg(-1) x day(-1) N(G)-nitro-L-arginine methyl ester (L-NAME) in the drinking water or drinking water alone from days 7 to 14 of gestation. Steady-state metabolic clearance studies of angiotensin II were then performed, or tissues were harvested for angiotensin II receptor studies. Treatment with L-NAME caused an increase in systolic pressure (P<0.001) in both pregnant and non-pregnant rats, while urinary protein excretion increased only in the pregnant SHRs (P<0.001). Plasma angiotensin II levels were significantly increased in the L-NAME-treated SHRs compared with controls (non-pregnant, P<0.0005; pregnant, P<0.01). The metabolic clearance rate of angiotensin II was decreased by L-NAME treatment in non-pregnant SHRs (P<0.05), but was increased by L-NAME treatment in the pregnant rats (P<0.01). In the aorta, the angiotensin II receptor number increased after treatment with L-NAME in both non-pregnant (P<0.0005) and pregnant (P<0.05) SHRs, and the dissociation constant increased in the non-pregnant SHRs (P<0.005). Thus treatment of SHRs with L-NAME increased blood pressure, as well as the circulating angiotensin II concentration and vascular angiotensin II receptor expression. However, treatment with L-NAME did not affect pressor sensitivity to infused angiotensin II. We conclude, therefore, that although a decrease in nitric oxide production is associated with changes in angiotensin II concentrations and receptor numbers, it does not induce changes in pressor sensitivity to infused angiotensin II in the SHR.

[Effects of thyroid hormone on cognitive function in rats with chronic cerebral ischemia].

OBJECTIVE: To study the characteristics of cognitive dysfunction in rats with chronic cerebral ischemia and the effects of thyroid hormone on the rats' cognitive function. METHODS: Thirty-one male SD rats were randomly allocated into normal control group (n=12), operation group (with bilateral carotid artery ligation and examined 5 weeks later, n=6), acute phase treatment group (APT, with bilateral carotid artery ligation and intragastric administration of thyroid hormone at 20 mg once daily for 5 weeks starting from the day of operation, n=7) and chronic phase treatment group (CPT, with the operation and thyroid hormone administration in an identical manner started from the sixth week following the operation, n=6). Morris water maze test was performed at the end of experiment. One-way ANOVA was used to estimate the differences in the learning and memory functions of the rats using SPSS10.0 for Windows. RESULTS: The average latent period of the operation group was significantly prolonged in comparison with that of the other groups (P<0.05) in spatial orientation test. The probe time (time spent in the quadrant where the platform was once situated) of normal control was much shorter than those of the operation, APT and CPT groups in spatial probe test (P<0.05), and the operation group had the poorest score. The average latent period of the operation and CPT was longer than that of the other groups (P<0.05) in working-memory task (P<0.05), and the operation group again had the poorest score. CONCLUSION: Spatial cognitive function is totally damaged in rats with chronic cerebral ischemia, and learning can not induce the formation long-term memory because of short-term memory damage. Thyroid hormone may lessen but can not fully repair the damage of the cognitive dysfunction resulting from chronic cerebral ischemia, and early intervention with thyroid hormone may be beneficial for chronic cerebral ischemia.

Systemic and colonic venous hemostatic alterations in horses during low-flow ischemia and reperfusion of the large colon.

Twenty-four horses were randomly allocated to 3 groups. All horses underwent a ventral midline celiotomy, and the large colon was exteriorized and instrumented. Group-1 horses served as sham-operated controls, group-2 horses underwent 6 hours of colonic ischemia, and group-3 horses were subjected to 3 hours of ischemia and 3 hours of reperfusion. Baseline blood samples were collected, then low-flow colonic ischemia was induced in horses of groups 2 and 3 by reducing colonic arterial blood flow to 20% of baseline. All horses were monitored for 6 hours. Citrated systemic venous (SV) blood samples were collected from the main pulmonary artery, and colonic venous (CV) samples were collected from the colonic vein draining the ventral colon. Samples were collected at 0, and 2, 3, 3.25, 4, and 6 hours for determination of one-stage prothrombin time, activated partial thromboplastin time, antithrombin III activity, and fibrinogen concentration. Data were analyzed statistically, using two-way ANOVA for repeated measures, and post-hoc comparisons were made by use of Student Newman Keul's test. Statistical significance was set at P < 0.05. There were significant decreases in all hemostatic variables by 2 hours in SV and CV samples from horses of all 3 groups, but there were no differences among the 3 groups for any of these variables. These hemostatic alterations could have been secondary to a hypercoagulable state or to fluid therapy-induced hemodilution. Colonic ischemia-reperfusion was not the cause of these alterations because these alterations also were observed in the sham-operated control horses. Significant temporal alterations existed even after accounting for the hemodilution.(ABSTRACT TRUNCATED AT 250 WORDS)

Chronic vanadium poisoning in calves and its treatment with calcium disodium ethylenediaminetetraacetate.

Sixteen Friesland heifer calves aged between 96 and 157 days were removed from a dairy farm that had been polluted with vanadium and randomly allocated into two equal groups (n = 8). The objective of the trial was to determine whether calcium disodium ethylenediaminetetraacetate (CaNa(2)EDTA) could be used as a treatment for cattle running in environments high in background vanadium. The treatment group received 80 mg CaNa(2)EDTA per kg body weight intraperitonealy (i.p.) twice a week over a 10-week period. The control group received normal saline i.p. over the same period. During the trial calves were exposed to a daily intake of vanadium in the form of contaminated tef hay derived from the farm of origin. In addition, the total mixed ration was spiked with a further 20 mg V(2)O(5)/kg feed to compensate for possible on-farm inhalation exposure. A stochastic model was used to estimate daily intake of vanadium as a distribution function. The model estimated that the daily intake of vanadium varied between an absolute minimum of 33 mg/day to an absolute maximum of 124 mg/day. The average intake of vanadium was 71.8 mg per day per calf. Various chemical pathology parameters were measured throughout the trial as well as urine excretion rates of vanadium and lymphocyte stimulation counts. All calves were slaughtered and necropsied in cohorts of 4-6 animals at monthly intervals after completion of the trial and withdrawal of vanadium from the ration. Tissue concentrations of vanadium were determined and necropsy findings were noted. The study found that CaNa(2)EDTA appears to enhance the excretion of vanadium in calves, but could not prove that the treatment had a protective effect against vanadium exposure. Calves were able to tolerate the prolonged treatment with CaNa(2)EDTA without side-effects.

Evaluation of physiologic indices in mice vaccinated with protein-ergotamine conjugates and fed an endophyte-infected fescue diet.

OBJECTIVES: To assess effects of vaccination against fescue toxicosis on weight gain, serum prolactin and cholesterol concentrations, and alkaline phosphatase (ALP) activity in mice fed an endophyte-infected (EI) or endophyte-free (EF) fescue diet. ANIMALS: 50 six-week-old male BALB/c mice. PROCEDURE: Mice were randomly allocated to the following 5 groups: 1, vaccinated intraperitoneally with a bovine serum albumin-ergotamine (EG) conjugate and fed an EI fescue diet; 2, orally vaccinated with cholera toxin (CT) subunit B-EG conjugate mixed with free CT and fed an EI fescue diet; 3, not vaccinated and fed an EI fescue diet; 4, passively vaccinated with monoclonal antibodies specific for ergovaline (EV) and fed an EI fescue diet; and 5, not vaccinated and fed an EF fescue diet. RESULTS: Antibodies against EG and EV were in serum of mice of groups 1 and 4, respectively. Secretory IgA and IgG coproantibodies against EG were induced in mice of group 2. Weight increased in groups 1 and 2 and tended to be increased in group 4 versus group 3. Prolactin concentration was similar in all groups; cholesterol concentration was decreased in groups 1, 3, and 4, compared with group 5. Compared with that in group 5, serum ALP activity decreased in groups 1 and 4 and was further decreased in group 1, compared with that in groups 2 and 3; it was negatively correlated with anti-EG titer. CONCLUSIONS AND CLINICAL RELEVANCE: Induction of anti-EG antibodies and administration of EV monoclonal antibodies tended to increase short-term weight gain in this murine model of fescue toxicosis. However, systemic IgG antibodies against EG or EV antibodies were not protective against decreases in serum ALP activity and cholesterol concentrations. Clinical significance of decreased ALP activity associated with vaccination is unknown, but represents a worsening of a response often associated with fescue toxicosis in cattle.

The pulmonary vasculature in a neonatal porcine model with increased pulmonary blood flow and pressure.

INTRODUCTION: Hypertension and hyperperfusion of the pulmonary vascular bed in the setting of congenital cardiac malformations may lead to progressive pulmonary vascular disease. To improve the understanding of the basic mechanisms of this disease, there is a need for clinically relevant animal models which reflect the disease process. MATERIAL AND RESULTS: We randomly allocated 45 newborn pigs, at the age of 48 hrs, to groups in which there was either construction of a 3 mm central aorto-pulmonary shunt, undertaken in 9, or ligation of the left pulmonary artery, achieved in 13. Controls included sham operations in 13, or no operations in 10 pigs. Follow-up was continued for three months. The interventions were compatible with survival in most pigs. The shunts resulted in an acute 85% increase in systolic pulmonary arterial pressure, and a more than twofold increase in pulmonary blood flow. By three months of age, nearly all shunts had closed spontaneously, and haemodynamics were normal. Ligation of the left pulmonary artery resulted in a normal total pulmonary blood flow, despite only the right lung being perfused, and a 33% increase in systolic pulmonary arterial pressure. These haemodynamic changes were maintained throughout the period of study. In both groups, histomorphometry revealed markedly increased muscularity of the intra-acinar pulmonary arteries. Circulating levels of endothelin were normal in the shunted animals, and elevated in those with ligation of the left pulmonary artery. CONCLUSION: In neonatal porcine models of pulmonary vascular disease, created by construction of 3 mm central aorto-pulmonary shunts and ligation of one pulmonary artery, we observed histopathological changes of the pulmonary vasculature similar to early hypertensive pulmonary vascular disease in humans. Elevated circulating levels of endothelin were associated with abnormal haemodynamics rather than abnormal pathology. These findings could be valuable for future studies on the pathogenesis of hypertensive pulmonary vascular disease associated with congenital cardiac malformations.

Effect of sucrose thermal oligosaccharide caramel, dietary vitamin-mineral level, and brooding temperature on growth and intestinal bacterial populations of broiler chickens.

Two experiments were conducted to determine the effect of sucrose thermal oligosaccharide caramel (STOC) and dietary vitamin-mineral (V/M) level on growth performance and intestinal microflora of broiler chickens. In Experiment 1, Peterson x Arbor Acres male broilers (n = 384) were randomly allocated into four groups that were fed either the control diet or diets containing the antibiotic virginiamycin (11 mg/kg), 3.7% STOC or 7.5% STOC for 4 wk at brooding temperatures of 32 to 29.7 C. Weight gains for broilers in Experiment 1 were greater (P < 0.001) for birds fed STOC diets, with weight gains of 763, 822, 1,124, and 1,080 g for birds on the control, antibiotic, 3.7% STOC, and 7.5% STOC diets, respectively. Feed intake and feed conversion by birds fed STOC diets were also significantly improved. Cecal bifidobacterial numbers were increased (P < 0.03) over the control diet with numbers being 5.98, 6.99, 7.47, and 7.39 log10 cfu/g cecal DM, respectively. In Experiment 2, Peterson x Hubbard male broilers (n = 384) were used in a 2 x 2 x 2 factorial arrangement with two levels of V/M premix (0.5 or 1% of the diet), two levels of STOC (0 or 3.5% of the diet), and two brooding temperatures, normal (32 to 23.6 C) or high (32 to 29.7 C) for 4 wk. Feeding the STOC diet improved (P < 0.05) weight gain, feed intake, and feed conversion of broilers. The effect of STOC on animal performance was less evident when broilers were fed twice the NRC recommended levels of V/M. Feeding the STOC diets resulted in a significantly greater increase in weight gain at high brooding temperatures than at normal brooding temperatures. There was also a reduction (P < 0.05) in numbers of total aerobes and coliforms in the ceca of birds fed diets containing STOC. Feeding STOC has potential to improve growth performance of broiler chickens.

Dietary-sodium-induced cardiac remodeling in spontaneously hypertensive rat versus Wistar-Kyoto rat.

OBJECTIVE: To study the effects of short-term and long-term high sodium intake on cardiac mass and design in sodium-sensitive spontaneously hypertensive rats versus sodium-resistant Wistar-Kyoto rats. METHODS: Young spontaneously hypertensive rats and Wistar-Kyoto rats were randomly allocated to control diet, 2 or 8% dietary sodium for 2-12 weeks and changes in resting hemodynamics, cardiac angiotensin II level, sympathetic activity and cardiac structure evaluated. Sympathetic activity was assessed by measuring levels of plasma catecholamines, responses of blood pressure to ganglionic blockade, and rates of cardiac turnover of norepinephrine. RESULTS: High sodium intake for 4 weeks increased left ventricle weight of Wistar-Kyoto rats aged 4 weeks (by 11 and 25% for 2 and 8% NaCl diets, respectively). This hypertrophic response was temporary, however, had already diminished after 6 weeks, and was absent after 12 weeks of a high sodium intake. However, after prolonged exposure concentric remodeling occurred (i.e. left ventricle wall thickness : radius ratio increased with no change in left ventricle mass). High sodium intake did not affect resting blood pressure, cardiac index, cardiac angiotensin II level, and general sympathetic activity of Wistar-Kyoto rats. Short-term high sodium intake did not increase left ventricle mass of young spontaneously hypertensive rats, unless sodium intake was so high (8% NaCl) that blood pressure and general sympathetic activity increased, too. However, a prolonged moderate (2%) increase in sodium intake also caused concentric remodeling in spontaneously hypertensive rats without increasing left ventricle mass, blood pressure, cardiac index, and general and cardiac sympathetic activities. CONCLUSIONS: The blood pressure in young Wistar-Kyoto rats is sodium-insensitive but the heart structure is sodium-sensitive and high dietary sodium intake causes an early hypertrophic response, and then concentric remodeling. In contrast, hypertrophic response appears to occur after the response of blood pressure in spontaneously hypertensive rats, whereas the remodeling is similar to that in Wistar-Kyoto rats.

Differential effects of the endocrine-disrupting compounds bisphenol-A and octylphenol on gonadotropin secretion, in prepubertal ewe lambs.

This study examined the effects of long-term exposure to the endocrine-disrupting compounds (EDCs) Bisphenol-A (BPA) and Octylphenol (OP) on gonadotrophin secretion in pre-pubertal female sheep. Four-week-old, female lambs were randomly allocated to four groups (n=6), and twice each week treated with i.m. injections of either corn oil (vehicle controls), diethylstilbestrol (DES; 0.175mg/kg), BPA (3.5mg/kg) or OP (3.5mg/kg). After 5 weeks of treatment, animals were ovariectomized (ovx) and ovary weights recorded. Two weeks later, blood samples were collected from lambs every 15min for 6h, for LH pulse analysis. Animals were then euthanased and adrenal and kidney weight recorded. An age-related increase in tonic LH secretion was noted in Control, BPA- and OP-treated lambs, but was absent in DES-treated lambs. Following ovx, LH secretion increased in all except DES-treated lambs; FSH concentrations increased in all groups. BPA and DES significantly suppressed LH pulse frequency (C: 6.7+/-0.3pulses/6h, DES: 1.5+/-0.8pulses/6h, BPA: 2.3+/-0.8pulses/6h) and amplitude (C: 7.1+/-1.0ng/ml, DES: 1.9+/-0.6ng/ml, BPA: 1.6+/-0.4ng/ml). OP had no effect on LH secretion (Frequency: 5.8+/-0.5pulses/6h, amplitude: 8.0+/-2.0ng/ml). Ovary weight was similar among all groups. Results show that chronic in vivo exposure of prepubertal female lambs to BPA, at levels lower than those reported previously, can have significant effects on LH secretion that are comparable to those seen following exposure to the known xenoestrogen, DES. Exposure to an equal dose of the EDC OP, over the equivalent period of time was without effect on gonadotropin secretion in the prepubertal ewe lamb. These results indicate that exposure of prepubertal female lambs to the EDC BPA can induce significant effects on gonadotropin secretion, the potential long-term effects of exposure and the effects of these changes on reproductive performance and efficacy, therefore, merit further study.

Superovulation in llamas (Lama glama) with pFSH and equine chorionic gonadotrophin used individually or in combination.

The purpose of this experiment was to evaluate the efficacy of pFSH and/or equine chorionic gonadotrophin (eCG) for inducing superovulation in llamas. Sixteen adult llamas weighing on average 130 kg (range 110-140) and which had been showing signs of oestrus for 5 days were randomly allocated to three treatment groups and one control group (n = 4). Llamas in Group A received eCG (500 IU, i.m.) once daily for 3 days, those in Group B received pFSH i.m., in decreasing doses every 12 h for 5 days for a total of 220 mg, while those in Group C received eCG (500 IU, i.m.) once, and pFSH (total of 156 mg, i.m.) in decreasing doses for the next 4 days. Llamas in Group D (control) received saline (5 ml, i.m.) every 12 h for 5 days. All llamas were allowed to be mated on the evening of Day 5 and were given hCG (750 IU, i.m.) at that time; a second mating was carried out 12 h later. A non-surgical ova/embryo collection technique was performed 7 days after the first mating and then the ovarian response was evaluated by way of laparoscopy. All 16 llamas were mated successfully. The mean (SEM) number of ovulations (7.3 +/- 3.1) in Group B was significantly higher (P < 0.05) than in the other groups (1.5 +/- 0.5, 2.0 +/- 0.7, and 0.3 +/- 0.3 for groups A, C and D, respectively). The number of follicles > 10 mm at the time of ova/embryo collection was significantly higher (P < 0.05) in the groups treated with eCG. A total of 21 ova/embryos was recovered from the all flushed llamas, corresponding to 47.7% of corpora lutea observed. Of the 21 ova, 15 were fertilised; 13 of those were classified as excellent blastocysts and the remaining 2 were classified as dead or degenerating. Results demonstrate that llamas can be successfully ovarian superstimulated while expressing behavioural oestrus and suggest that pFSH is more effective than eCG to induce superovulation.

Length of docked tail and the incidence of rectal prolapse in lambs.

A multistate cooperative study was conducted to study the current issue of tail length in docked lambs and its relationship to incidence of rectal prolapse. A total of 1,227 lambs at six locations were randomly allocated to two or three tail dock treatments: 1) short--tail was removed as close to the body as possible, 2) medium--tail was removed at a location midway between the attachment of the tail to the body and the attachment of the caudal folds to the tail, and 3) long--tail was removed at the attachment of the caudal folds to the tail. Short-docked lambs had a greater (P < 0.05) incidence of rectal prolapse (7.8%) than lambs with a medium (4.0%) or a long (1.8%) dock. Female lambs had a higher (P < 0.05) incidence of rectal prolapse than male lambs. At two stations, lambs were finished either in a feedlot on a high-concentrate diet or on pasture with no grain supplementation. At one station, with a very low incidence of rectal prolapse, there was no difference in incidence between lambs finished in the feedlot or on pasture; however, at the station with a relatively high incidence of rectal prolapse, lambs in the feedlot had a higher (P < 0.05) incidence than lambs on pasture. The half-sib estimate of heritability for the incidence of rectal prolapse was low (0.14). The results of this study strongly implicate short dock length as a cause of rectal prolapse in lambs finished on high-concentrate diets. Furthermore, the results of this study and the only other study known conducted on this issue strongly suggest that docking lambs at the site of the attachment of the caudal folds to the tail will result in a negligible incidence of rectal prolapse.

Effect of light-tip distance on the shear bond strengths of resin-modified glass ionomer cured with high-intensity halogen, light-emitting diode, and plasma arc lights.

INTRODUCTION: The purpose of this study was to assess the effect of light-tip distance on the shear bond strength and the failure site of brackets cured with 3 light-curing units (high-intensity halogen, light-emitting diode, and plasma arc). METHODS: One hundred thirty-five bovine mandibular permanent incisors were randomly allocated to 9 groups of 15 specimens each. Stainless steel brackets (Victory Series, Unitek/3M, Monrovia, Calif) were bonded with a resin-modified glass-ionomer (Fuji Ortho LC, GC Europe, Leuven, Belgium) to the teeth, and each curing light was tested at 3 distances from the bracket: 0, 3, and 6 mm. After bonding, all samples were stored in distilled water at room temperature for 24 hours and subsequently tested for shear bond strength. RESULTS: When the 3 light-curing units were compared at a light-tip distance of 0mm, they showed no significantly different shear bond strengths. At a light-tip distance of 3 mm, no significant differences were found between the halogen and plasma arc lights, but both lights showed significantly higher shear bond strengths than the light-emitting diode light. At a light-tip distance of 6 mm, no significant differences were found between the halogen and light-emitting diode lights, but both showed significantly lower bond strengths than the plasma arc light. When the effect of the light-tip distance on each light-curing unit was evaluated, the halogen and light-emitting diode lights showed no significant differences among the 3 distances. However, the plasma arc light produced significantly higher shear bond strengths at a greater light-tip distance. No significant differences were found among the adhesive remnant index scores of the various groups, except with the LED light at a distance of 3 mm. CONCLUSIONS: In hard-to-reach areas, the plasma arc curing light is suggested for optimal curing efficiency.

Effect of customization of master gutta-percha cone on apical control of root filling using different techniques: an ex vivo study.

AIMS: (i) To compare the prevalence of extrusion of root filling material when placed using different root filling techniques, with or without customization of the master gutta-percha (GP) cone; and (ii) to investigate the effects of some factors influencing root filling extrusion and presence of voids. METHODOLOGY: A total of 180 roots were selected, prepared and randomly allocated to three groups. Five general dental practitioners performed the root fillings; each filled one group of roots (n = 60) using each of three techniques; 'cold lateral compaction' (n = 20), 'warm vertical compaction' (n = 20) and 'continuous-wave' (n = 20) techniques. For each obturation technique, the master GP cone was customized using chloroform in 10 samples. Two groups of the roots were recycled to allow all five operators to fulfill their remit. Two observers, blind to operator and obturation technique, examined the radiographs (master apical file, post-obturation) to determine the presence of root filling extrusion and voids within the apical 5 mm, independently. Root filling extrusion was also confirmed by direct inspection of the root apex after obturation. The data were analysed using logistic regression models. RESULTS: A total of 300 root fillings were performed; nine were excluded from the analysis. Most of the root fillings (80%, n = 233) were placed within 0.5 mm of the working length; only 20% (n = 58) were placed >0.5 mm beyond the working length. The odds of prevalence of extrusion (>0.5 mm) were significantly reduced by about 50% when cold lateral compaction or customization of GP were used. One operator produced 2.5 times more extruded root fillings than others. Curvature & length of root canal, apical size of prepared canal, as well as operator's preferred obturation technique had no significant influence on the prevalence of extrusion. Customization of GP was the sole factor to significantly reduce the prevalence of voids within the apical 5 mm of working length. CONCLUSIONS: Root filling extrusion was significantly influenced by 'operator' and was reduced by cold lateral compaction and customization of the master cone. Customization of master cone was the only factor that reduced voids apically.

Effect of pasture vs. concentrate diet on CLA isomer distribution in different tissue lipids of beef cattle.

This study examined the effects of feeding pasture vs. concentrate on the distribution of CLA isomers in the lipids of longissimus and semitendinosus muscle, liver and heart muscle, and subcutaneous fat in beef bulls. Sixty-four German Holstein and German Simmental bulls were randomly allocated to either an indoor concentrate system or periods of pasture feeding followed by a finishing period on a concentrate containing linseed to enhance their beef content of n-3 PUFA and CLA. The concentrations of CLA isomers in the different tissues were determined by GC and silver ion HPLC. The diet affected the distribution of individual CLA isomers in the lipids of the different tissues. The concentration (mg/100 g fresh tissue) of the most prominent isomer, cis-9,trans-11 18:2, was increased up to 1.5 times in liver and heart tissue of bulls fed on pasture as compared with concentrate. However, no diet effect was observed for cis-9,trans-11 18:2 in the lipids of longissimus muscle and subcutaneous fat. In all tissues, the second-most abundant CLA isomer in concentrate-fed bulls was trans-7,cis-9 18:2. In contrast, trans-11,cis-13 18:2 was the second-most abundant CLA isomer in all investigated tissue lipids of pasture-fed bulls. The concentration of the trans-11,cis-13 18:2 isomer was up to 15 times higher in tissues of pasture-fed bulls as compared with concentrate-fed animals. Furthermore, diet affected the concentrations of the CLA trans, trans 18:2 isomers. Pasture feeding significantly increased the concentrations of some trans, trans 18:2 isomers as compared with concentrate, predominantly trans-12,trans-14 18:2 and trans-11,trans-13 18:2. Overall, pasture feeding resulted in significantly increased concentrations of the sum of CLA isomers in the lipids of longissimus muscle, subcutaneous fat, heart and liver muscle of German Holstein and German Simmental bulls, but not in semitendinosus muscle.

Periodontal regeneration with peptide-enhanced anorganic bone matrix in particulate and putty form in dogs.

BACKGROUND: Peptide-enhanced (using the peptide 15 [P-15] synthetic peptide) anorganic bone matrix (ABM) particulate (PPart) grafts have demonstrated clinical and histologic success in human periodontal defects. Dispersion of ABM/P-15 in sodium hyaluronate carrier (PPutty) improves the handling properties of the graft material. The healing of ABM/P-15 particulate and ABM/P-15 putty was compared in critical-sized fenestration defects in 16 mongrel dogs. METHODS: After full thickness flap reflection, 7 mm diameter fenestrations were made with a trephine in the mid-root of both maxillary canines in each dog. Bone, periodontal ligament, and cementum were removed as completely as possible with hand root planing within the trephine-produced notches. By random allocation, each defect was filled with PPart or PPutty, and the flaps were closed with sutures. One dog contributing two defects served as a negative control. Block sections were retrieved at 3 and 8 weeks for histologic processing. Three 6-mu step serial sections in the center of the defects were used for analysis. Parameters measured included the original length of the wound, linear amount of periodontal regeneration including new cementum, bone and connective tissue, and area measurements of new bone and remaining particles. PPutty and PPart results were compared for significant differences using the Wilcoxon rank sum test. RESULTS: Clinical healing was uneventful in all cases. There was no evidence of inflammation or adverse tissue reactions with either material. The controls showed minimal regeneration at the periphery of the defect. Histomorphometric evaluation of the grafted defects revealed the following: at 3 weeks, there was minimal new bone formation (occupying 4.2% of the grafted area for the PPutty and 1.2% for the PPart). The grafted particles occupied 21.2% and 35.6% of the area for the PPutty and PPart, respectively (P = 0.039). At 8 weeks, there was a tendency for greater new bone formation compared to 3 weeks with both materials. There was significantly more new bone with the PPutty (49.3%) compared to the PPart (14.8%) (P = 0.045). The grafted particles occupied 7.9% and 17% of the grafted area for the PPutty and PPart, respectively (no significant difference). There were no significant differences for any of the linear measurements. ABM/P-15 PPutty had superior handling characteristics. CONCLUSIONS: Both ABM/P-15 materials yielded satisfactory healing and resulted in the greater regeneration of fenestration defects in dogs at 8 weeks compared to controls. In addition, AMB/P-15 putty resulted in more bone formation compared to ABM/P-15 particulate.
