Morphometry and autoradiography of early hypertrophic changes in the ventricular myocardium of adult rat. A light microscopic study.

Quantitative autoradiographic localization of newly synthesized proteins was studied in the ventricular myocardium of adult rats 2 hours after the intravenous injection of 3H-leucine and 20 hours after the induction of hypertrophy by constriction of the abdominal aorta. Equal area samples of tissue from experimental and sham-operated controls were examined for differential grain counts and morphometric measurements by light microscopy. In comparison with control levels, the hypertrophic hearts show a 20 per cent increase in the average cross-sectional area of myocytes and a 43 per cent increase in the incorporation of amino acid into proteins. The higher concentration of labeled proteins is confined mainly to cardiac muscle cells, which show a 90 per cent increase in grain counts per cell cross-section, whereas no significant change is observed over the interstitium. A test of the specificity of the autoradiographic technique shows the expected reduction in tissue labeling following cycloheximide inhibition of protein synthesis, indicating that there is no significant amount of nonspecific binding of free amino acid during the paraformaldehyde-glutaraldehyde fixation procedure.

Focal segmental glomerulosclerosis in childhood: histology, glomerular morphometry, electron microscopy and immunofluorescence findings in biopsies performed early in the course of the disease.

The histological evolution of Focal Segmental Glomerulosclerosis (FSGS) is poorly documented due to variation in the time at which the biopsy is taken. We looked at patients presenting with steroid resistant nephrotic syndrome (SRNS) in which the first biopsy was performed within 3 months of presentation. FSGS lesion was demonstrable in 68.5% of cases in the first biopsy. Glomerular size was increased in 86% of patients indicating that is an early event in the course of the disease. The group was heterogenous with respect of mesangial cellularity, mesangial matrix, position of FSGS lesion in the glomeruli, glomerular size, lamina densa thickness and immunofluorescence findings. No association of morphological features was seen permitting subclassification of this group on morphological grounds. Thus, both the FSGS lesion and glomerular enlargement occur early in the evolution of idiopathic FSGS presenting with SRNS.

The influence of the ovulation rate on ultrasonically determined ovine corpus luteum morphometry and progesterone concentrations in cyclic and early pregnant sheep.

The objective of the present study was to describe morphology and function of the Corpora lutea (CL) during the oestrous cycle and early pregnancy in sheep with different ovulation rates. In total 40 Booroola. Mutton Merino crosses [heterozygous carriers (FecBFec+) and non-carriers (Fec+Fec+) of the Booroola-fecundity gene (FecB)] with ovulation rates 1 to 4 were examined. During the oestrous cycle (n = 20) and the first month of pregnancy (n = 20) blood samples were taken daily (radioimmunoassay of progesterone) and an ultrasonic ovary diagnosis was conducted. The ewes were scanned transrectally with a 7.5 MHz linear probe lying in a dorsal position. During every examination the CL could be detected. The number and the diameter of the CL were documented and the total volume of luteal tissue per ewe was calculated. The effect of the ovulation rate on CL-morphology (diameter and total volume of luteal tissue per ewe) and peripheral progesterone concentrations were assessed by one-way ANOVA. On day 6 and 7 post ovulationem in cyclic and early pregnant sheep 42% of the diagnosed CL had a cavity. On day 11 (cyclic sheep) and day 10 post ovulationem (early pregnant sheep) this number decreased to 22% (p < 0.05). Both conditions of the CL (compact or with a central cavity) are similar in function and should be regarded as appearances of the same basic process. From the third day onwards the ovulation rate influenced significantly (p < 0.05) the development of the outside diameters of the CL. However, the ovulation rate had no effect on the total volume of the luteal tissue per sheep and on the progesterone concentrations. Yet, in sheep with the ovulation rate 1 significantly lower progesterone concentrations were determined than in sheep with the ovulation rates 2 to 4. In sheep with the ovulation rates 2 to 4 the peripheral progesterone concentrations did not differ significantly. In cyclic and pregnant sheep there is a positive correlation (r = 0.75, p < 0.05) between the progesterone concentration and the total volume of luteal tissue. Considering the smaller diameters of the preovulatory follicles it seems that the development of the CL continues until a threshold-value of progesterone and/or of the total luteal tissue is exceeded. Ewes with low ovulation rates reach this threshold-value with only a few but large CL. With increasing ovulation rate the CL tend to have smaller diameters.

[Relation between placental morphometry and fetal growth].

Forty-eight placentae of full term infants, 21 placentae from appropriate for gestational age infants (AGA) and 27 placentae from small for gestational age infants (SGA) were measured by morphometric technic using the automatic image analyzer, in order to find out the extent of fetomaternal exchange which determines the transfer of oxygen and nutrition from mother to fetus and fetal growth. The results of measurement correlated well both with infant birth weight and placental weight. They demonstrated striking quantitative differences when the placentae of SGA were compared with those of AGA. The placenta weights in the group of SGA were notably less than those in the group of AGA. It seems that low birth weight relates to low functional tissue mass of placenta. This reduction of functional tissue is accompanied by diminution of the area for exchange between mother and fetus, both at the villous surface area and at fetal capillary surface area. Thus, the ability of transferring oxygen and nutrition from mother to fetus is curtailed. The results show that the rate of fetal growth is limited by placental function as well as its weight.

[Morphometry of the normal cadaveric cervical spinal cord].

We measured the cross-sectional area and diameters of the C7 segment in 80 cadaveric specimens obtained from patients without spinal cord lesions died at 60 to 79 years of age. We found considerable individual variation in the spinal cord size. The transverse area of the C7 segment varied from 33.3 mm2 to 67.7 mm2 (mean 48.4 +/- 6.9 mm2). The size of the cord had no correlation with body weight, and had some correlation with body height. But the individual variation in the cord size was considerably large even among the cases of the same height. This large variation in the cord size should be under consideration in morphometric analysis of the spinal cord. We showed the morphometric data based on the measurements of the cross-sectional area and diameters in each segment from C2 to Th1 in 14 cases. Although the size of the spinal cord varied markedly from case to case, the relative ratio of cross-sectional area in each segment to that of C3 was similar among the cases (C2; 1.01 +/- 0.04, C4; 1.02 +/- 0.03, C5; 1.09 +/- 0.04, C6; 1.09 +/- 0.06, C7; 1.07 +/- 0.06, C8; 0.98 +/- 0.05. Th1; 0.80 +/- 0.04). Based on our data, the proper cross-sectional area of each segment and of each individual may be calculable from measurement of a given single segment.

Morphology and morphometry of the cervix uteri of female albino rats in the estrus and diestrus phases.

The morphological aspects of the cervix uteri of twelve albino rats, six in estrus and six in diestrus were studied. Micrometry of the epithelium thickness was performed and it was observed that the epithelium is significantly thicker during the estrus phase of the cycle than in diestrus. The major causes for this are discussed and the findings are correlated with the estrogen and progesterone levels present in each of the mentioned phases.

The proliferative fraction in lymph nodes: a comparison of proliferating cell nuclear antigen morphometry to flow cytometry.

A monoclonal antibody to proliferating cell nuclear antigen (PCNA/cyclin) has recently become available. This antibody, as opposed to Ki-67, can be used on formalin-fixed, paraffin embedded tissue sections and allows retrospective comparison of PCNA positivity to percent S + G2 + M of the cell cycle. To compare fresh lymphoma deoxyribonucleic acid (DNA) analysis with PCNA activity on fixed, paraffin-embedded sections, prospective flow cytometric studies of cell cycle analysis were performed on lymph nodes removed from 10 patients for diagnosis. Six patients had T-cell lymphoma, two had B-cell lymphoma, and two were benign. Using the peroxidase-antiperoxidase method, the nuclear positivity of archival lymphoma cases was also examined. To quantify PCNA positivity, a unique morphometric method was employed that utilized digital imaging by high definition television and ELAS (Earth Land Application Software), a geoscience software used extensively for color quantitation of remote sensed data. The immunologic percent PCNA positivity was 26.1 +/- 20 vs. percent S + G2 + M by flow cytometry of 22.4 +/- 10 with a correlation coefficient (r) of 0.55. This r-value compared favorably to data generated for Ki-67 in solid malignant neoplasms. The six more concordant cases had a percent PCNA positivity of 26.5 +/- 10.0 and a percent S + G2 + M of 27.3 +/- 8.6, r = 0.96. Our study is unique in that it compared fresh lymphoma DNA analysis data with paraffin PCNA data. It is our conclusion that immunologic PCNA positivity in paraffin sections correlates with fresh flow cytometric S + G2 + M in lymph nodes, although careful attention must be paid to the area of the node quantified for PCNA.

Morphometry and autoradiography of early hypertrophic changes in the ventricular myocardium of adult rat: an electron microscopic study.

The ultrastructural composition of left ventricular myocardium and myocytes has been investigated at 20 hours after the surgical production of a subdiaphragmatic aortic stenosis in the adult rat. Morphometric analysis shows a 20% increase in the size of myocytes resulting principally from a 36% increase in mitochondrial volume and a 78% increase in the volumes occupied by smooth endoplasmic reticulum and matrix. Myofibrillar volume is increased only 4%. Measurements of the size and shape of individual mitochondria indicate that the augmentation of this compartment is wholly accountable by the enlargement of preexisting mitochondria. Quantitative autoradiographic analysis following a 2-hour pulse labeling with 3H-leucine shows that the rate of incorporation of newly synthesized proteins is practically uniform throughout all structural components of the sarcoplasm 18 to 20 hours after aortic banding. The conclusion is reached that this interval of hypertrophic response is a transition period characterized by an increasing rate of synthesis of contractile proteins overtaking a previously greater rate of mitochondrial synthesis.

Osteon morphometry in females with femoral neck fractures.

The authors measured osteon dimensions and number in specimens of cortical bone obtained from the medial femoral neck (calcar) from nine female patients treated with hemiarthroplasty for femoral neck fractures, and from 12 (seven women, five men) age-matched cadavers without fractures. Specimens from the same location in 14 (seven women, seven men) younger patients treated with total hip arthroplasty (THA) for osteoarthrosis were also studied. There were fewer osteons per unit area and the osteons and their Haversian canals were bigger in the fracture group than in the nonfracture or osteoarthritic groups. Patients with femoral neck fractures also exhibited a decreased Singh index and an increased intracortical porosity compared with age-matched normal controls. By contrast, the osteoarthritic group showed no difference in osteon or Haversian dimensions, osteon number per unit area, Singh index, or in porosity compared with the nonfracture group. These morphometric differences found in the cortical bone of the medial femoral neck may play a role in the incidence of fractures.

Morphometry of nuclei of the normal and malignant prostate in relation to DNA ploidy.

Fine needle biopsies from 70 patients with prostate carcinoma and 10 patients with benign hyperplasia were used to study area, variation in size and form factors of the nuclei by image analysis. The results were related to DNA ploidy of the cell populations as measured by flow cytometry, cytologic grade and patients' survival. Nuclear area differed significantly between benign lesions and tumors. It increased in diploid low-grade tumors from a normal value of 54.2 +/- 3.1 microns2 to 75.6 +/- 5.3 microns2. In aneuploid tumors with an increase in the chromosome number, the nuclear size further increased to about twice that of benign nuclei. Variation in size also differed between benign and malignant epithelium, with a further increase between diploid and gross aneuploid tumors. While nuclear size and variation in nuclear size made it possible to discriminate malignant from benign lesions, form factor did not differ between benign and malignant lesions. In follow-up, however, none of these factors reached significance for predicting survival.

Morphometry of cupromeronic blue-stained proteoglycan molecules in animal corneas, versus that of purified proteoglycans stained in vitro, implies that tertiary structures contribute to corneal ultrastructure.

Isolated, purified small chondroitin (dermatan) sulphate proteoglycans from corneas of cow and rabbit and cow sclera were stained with Cupromeronic blue in 'model' experiments. The lengths and thicknesses of the images were compared with those of the same proteoglycans stained in the tissue, using the critical electrolyte concentration principle to give specificity for sulphated proteoglycans, and keratanase 1 or chondroitinase ABC digestion to distinguish between chondroitin and keratan sulphate. Corrections for orientation of the stained glycan filaments within the section plane were made to convert the observed lengths to true average lengths. Observed lengths of stained chondroitin (dermatan) sulphate were greater than those of keratan sulphate, both in models and tissues, in agreement with published data from biochemical and rotary-shadowing studies, in both species. Corrected (true) average lengths of stained isolated chondroitin (dermatan) sulphate proteoglycans were slightly, but not significantly, longer than expected from rotary shadowing or biochemical measurements. Keratan sulphate lengths were similarly somewhat longer. The data support the idea that Cupromeronic blue acts as a scaffold that helps maintain polyanion shape against distortion on staining. Stained filaments in tissues were sometimes over twice the length of isolated stained proteoglycans, suggesting that 2 glycan chains were aligned end-to-end. Thicknesses of proteoglycan filaments suggested that at least 2 glycan chains were aligned side-by-side, both in models and in tissues. A scheme for proteoglycan tertiary structure in cornea is proposed, in which glycan chains may bridge collagen fibrils in duplexed forms similar to those observed in rotary shadowed preparations.
