Interactions between monogenean parasites and their fish hosts.

Parasite factors associated with recognition and selection of the host and the mechanisms in the host responsible for acceptance or rejection of the invading organism were evaluated. Sensory structures in parasites are able to detect differences between different fish species and this ability to discern between fishes may be based on both chemical and mechanical stimuli on the host surface. Complex glycoproteins, proteins, carbohydrates and simple molecules attract parasites or modify their behaviour. Furthermore, attachment of the monogenean parasite to a host is dependent on both mechanical structures and chemical factors in the parasite. These systems comprise anterior pads, posterior haptors, gland secretions, and muscular elements. The parasite needs access to appropriate nutrients which can be absorbed and used for reproduction and in this context signals from the host are needed for an optimal physiological response of the parasite. The innate and adaptive immune systems of the host are important elements in this question. Investigations have indicated that innate host factors (complement, lectins, acute phase reactants, macrophages) can bind to monogeneans and elicit severe damage to the parasites. The targets for these hostile products are not only the monogenean tegument, but may involve the gastrodermis and glands. However, the parasite's ability to avoid and even exploit the wide array of immunological elements of the host may be an important player in the dynamic interactions between host and monogenean determining host specificity. Even fish hosts susceptible to a certain parasite show an ability to mount a protective response at post-infection periods. Elevation of the host's production of adaptive and non-adaptive factors following monogenean infections of a certain duration may explain the acquired response.

[Acid-fast bacilli isolated from foot pads of nude mice infected with leprosy bacilli].

When leprosy bacilli grown in nude mouse foot pad were used for culture experiments, cultivable acid-fast bacillus was sometimes isolated as a contaminant. Whenever bacilli were inoculated to nude mice, the same leprosy bacilli were killed by autoclaving and were inoculated in to foot pads of 5 nude mice for examination of this cause of the contamination. Acid-fast bacillus was cultivated on 3% Ogawa egg medium at 33 degrees C from homogenates of foot pads of nude mice infected with M. leprae after one year and a while of infection. Foot pad of nude mouse injected with leprosy bacilli was cut off, ground in mortar and passed through sterile absorbent cotton and the filtrate was centrifuged at 10,000 rpm for 30 minutes. The sediment was inoculated on 3% Ogawa egg medium after treating with a small amount of sterile 1 N sodium hydroxide. Acid-fast bacilli were isolated from 3 out of 41 mice inoculoted with heat killed bacilli. The isolated acid-fast bacillus did not be observed in the same experimental group inocudated with live bacilli, positive cases were scattered in another groups. Four out of 16 tubes were positive for acid-fast bacilli in mice infected with Kurume-naha and 5 out of 7 tubes in the Amami-KM infected mouse group. The two negative tubes were discarded due to contamination. Kurume-Oki strain which has yellow colonial morphology was isolated from one out of 6 culture tubes. Strains Kurume-naha and Amami-KM have the same characteristics as follows: slow grower with pale yellow smooth colonial morphology, strongly positive for niacin production and ureas; positive for nicotinamidase, pyradinamidase and 68 degrees C catalase; no growth at 45 degrees C, negative for nitrate reduction, hydrolysis of Tween 80, diamine oxidase, heat stable acid-phosphatase and arylsulphatase; resistant to streptomycin, isoniazid, rifampicin and B 663. Two isolates were identified as Mycobacterium simiae from these characteristics. Characteristics of a Kurume-Oki isolate was as follows: slow grower with yellow smooth colonial morphology, positive for urease, 68 degrees C catalase, hydrolysis of Tween 80 and arylsulfatase; no growth at 45 degrees C, negative for niacin production, nicotinamidase, pyradinamidase, nitrate reduction, daimine oxidase and heat stable acid-phosphatase; resistant to streptomycin, isoniazid, rifampicin and B. 663. This bacillus was identified as Mycobacterium gordonae from these characteristics.(ABSTRACT TRUNCATED AT 400 WORDS)

Estimating vaginal blood loss.

In order to estimate amounts of vaginal bleeding, obstetrician-gynecologists have traditionally asked women the number of napkins, pads or tampons they use. This method of estimation rests on the assumptions that the volume of blood absorbed by various sanitary products is (1) known and (2) equivalent. To test these underlying assumptions the blood absorption capacity of 15 commercially available sanitary products was measured. Mean absorptions ranged from 1.32 to 94.86 ml for different products; a one-way analysis of variance showed that these mean absorptions were not equal (p less than .001). Moreover, products within a single package revealed substantial variations in absorbency. Thus, asking women the number of sanitary products they use is unlikely to yield clinically useful information; such questions should probably be deleted from the gynecologic history.

Shock attenuation of various protective devices for prevention of fall-related injuries of the forearm/hand complex.

BACKGROUND: Attenuation of the peak impact force is essential in any protective devices for prevention of fall-related injuries. HYPOTHESIS: Common wrist guards have limited effectiveness because of the multifaceted nature of wrist injury mechanisms, and other modalities may provide enhanced shock-absorbing functions. STUDY DESIGN: Controlled laboratory study. METHODS: A free-fall device was constructed using a mechanical surrogate to simulate falling impact. At 4 different falling heights, 5 different hand conditions were tested: bare hand, a generic-brand wrist guard, a Sorbothane glove, an air cell, and an air bladder condition. The impact force from the ground and the transmitted impact force to the forearm/hand complex were simultaneously measured. RESULTS: The falling height and hand condition significantly modulated the impact responses. The padded conditions always had significantly smaller peak impact forces compared with the bare-hand condition. The wrist guard became ineffective in impact force attenuation beyond the falling height of 51 cm. On the other hand, the air bladder condition maintained less than 45% of the peak impact force of the bare-hand condition and remained below the critical value, whereas other conditions were all ineffective. CONCLUSION: It was reconfirmed that common wrist guard design could provide limited impact force attenuation, whereas damped pneumatic springs would provide substantially enhanced shock-absorbing functions. CLINICAL RELEVANCE: A wrist guard incorporating volar padding with the pneumatic spring design principle might be more effective at preventing injuries than are currently available designs.

Endothelial lipase provides an alternative pathway for FFA uptake in lipoprotein lipase-deficient mouse adipose tissue.

Lipoprotein lipase (LPL) is thought to be the only enzyme responsible for the catabolism of triglycerides (TGs) associated with TG-rich lipoproteins in adipose tissue (AT). However, LPL deficiency in humans and induced mutant mice is not associated with decreased fat mass. We investigated whether endothelial lipase (EL), a recently discovered phospholipase, might represent an alternative mechanism for the uptake of phospholipid-derived fatty acids in murine lipoprotein-deficient AT. When LPL was expressed in AT and isolated murine adipocytes, EL mRNA was not detectable. In contrast, mouse AT and isolated adipocytes that lacked LPL expressed large amounts of EL mRNA. The cellular phospholipase activity in LPL-deficient fat pads was increased 4-fold compared with control fat pads and could be inhibited to control levels by a specific EL antibody. Fatty acids produced by EL activity were absorbed by adipocytes and incorporated into the TG moiety of AT. Our results suggest that EL activity in AT and other peripheral tissues might contribute to the tissue uptake of free fatty acids, which could have important implications for the metabolism of plasma lipoproteins.

Assay of Delta9-tetrahydrocannabinol (THC) in oral fluid-evaluation of the OraSure oral specimen collection device.

Oral fluid is considered to be an alternative to urine testing for the detection of acute ingestion of drugs. The OraSure Intercept DOA Oral Specimen Collection Device (OSCD) has been used in studies for the quantitation of Delta9-tetrahydrocannabinol (THC), but concerns have been raised. In the present study, we investigated whether the volume of oral fluid can be determined and how much THC remains adsorbed on the device. It was found that THC is markedly adsorbed onto the absorptive pad. The recovery using the standard elution procedure was only 37.8 +/- 9.4% for 10 ng/mL and 55.6 +/- 1.0% for 100 ng/mL of THC in oral fluid (n = 5 each). With an additional methanol wash, a further 25% could be eluted. Therefore, a modification of the procedure was evaluated, consisting of the addition of 2 mL of methanol to the elution buffer. THC could be completely recovered over a range of concentrations (1 to 1000 ng/mL). For the determination of the amount of oral fluid absorbed, a gravimetric approach was evaluated as the weights of the devices vary only by 0.6% relative standard deviation. After application of 0.5 mL oral fluid to pads and evaluation of the weight differences, the applied amount could be estimated with a precision of 7.5% (n = 8) and an accuracy of 6.1%. From these results it can be concluded that the OraSure OSCD is useful to collect oral fluid for reliable quantitative THC assay applying a modified elution procedure and gravimetric determination of the amount of oral fluid.

The effects of padded surfaces on the risk for cervical spine injury.

STUDY DESIGN: This is an in vitro study comparing cervical spine injuries produced in rigid head impacts and in padded head impacts. OBJECTIVES: To test the hypothesis that deformable impact surfaces pose a greater risk for cervical spine injury than rigid surfaces using a cadaver-based model that includes the effects of the head and torso masses. SUMMARY OF BACKGROUND DATA: It is widely assumed that energy-absorbing devices that protect the head from injury also reduce the risk for neck injury. However, this has not been demonstrated in any experimental or epidemiologic study. On the contrary, some studies have shown that padded surfaces have no effect on neck injury risk, and others have suggested that they can increase risk. METHODS: Experiments were performed on 18 cadaveric cervical spines to test 6 combinations of impact angle and impact surface padding. The impact surface was oriented at -15 degrees (posterior impact), 0 degree (vertex impact), or +15 degrees (anterior impact). The impact surface was either a 3-mm sheet of lubricated Teflon or 5 cm of polyurethane foam. RESULTS: Impacts onto padded surfaces produced significantly larger neck impulses (P = 0.00023) and a significantly greater frequency of cervical spine injuries than rigid impacts (P = 0.0375). The impact angle was also correlated with injury risk (P < 0.00001). CONCLUSIONS: These experiments suggest that highly deformable, padded contact surfaces should be used carefully in environments where there is the risk for cervical spine injury. The results also suggest that the orientation of the head, neck, and torso relative to the impact surface is of equal if not greater importance in neck injury risk.

An ultrastructural study and stereological analysis of the colon wall in the cockroach Periplaneta americana.

The colonic epithelium has been examined for ultrastructural evidence of physiological activity. The cells show extensive folding of the apical plasma membrane, associated with mitochondria and an internal coating of particles about 120 A diameter. Anteriorly many apical infoldings are dilated at the tip to form substantial extracellular spaces up to 0.8 micron wide. Narrow intercellular channels are present, opening to the haemolymph side of the epithelium. Pinocytosis is seen frequently at the basal surface. The surface densities of apical plasma membranes were not significantly different in the posterior mid-gut and colon. Similarly the volume densities of mitochondria were equal in the colonic epithelium and rectal pads, but the surface density of outer mitochondrial membranes was greater in the colon. It is suggested that the colon may absorb organic solutes from the gut lumen.

[Assessment of exposure to organic aromatic compounds and PAH in asphalt industry: the PPTP-POPA Study results].

Asphalt is a mixture of mineral matter and bitumen, its fumes contain about 1% of Polycyclic Aromatic Hydrocarbons (PAH), some of which are carcinogens. In the PPTP-POPA Study of Lombardy Region, a group of 100 asphalt workers (exposed to bitumen fumes and diesel exhausts) and a group of 47 ground construction operators (exposed only to diesel exhausts) were investigated to assess PAH exposure in Italy, by means of environmental-air monitoring (the 16 most relevant, according to the American Environmental Protection Agency, EPA) and biological monitoring (urinary 1-hydroxypyrene excretion). Our results show that PAH exposure in these workers is not higher than that observed in traffic policemen working in urban areas. Since dermal exposure has been suggested as a major determinant of the total PAH dose absorbed by road pavers from bitumen fumes, we assessed skin contamination by organic aromatic compounds and by sixteen PAH: in both groups, six pads were applied to each subject in different parts of the body, during the workshift. The results show that the dermal contamination in road pavers is higher than in ground construction operators and that cutaneous dose rate is higher than respiratory dose rate, whereas the amount of absorption the ratio is inverted.

The bovine digital cushion--a descriptive anatomical study.

The properties of the suspensory and supporting structures of the bovine claw are of particular importance in the pathogenesis of claw lesions since both must function optimally to prevent soft tissue compression and trauma. An essential component is the shock-absorbing digital cushion situated under the distal phalanx. The sound claws of 54 slaughtered cows were dissected. The digital cushion consisted of three parallel pads--axial, middle and abaxial--that ran longitudinally from the heel and underneath the distal phalanx with numerous transverse finger-shaped branches that connected the axial and abaxial pads cranial to the flexor process of the distal phalanx. The middle fat pad frequently ended just at the apical end of the flexor process of the distal phalanx, which may result in an inferior cushioning effect under the distal phalanx and increase the likelihood of ulcers. There were differences in the structure of the digital cushion with age and loose connective tissue in heifers' pads was first replaced by fat which, after the third lactation, was gradually supplanted by collagenous connective tissue.

A pilot study to evaluate reusable absorbent body-worn products for adults with moderate/heavy urinary incontinence.

OBJECTIVE: The purpose of this pilot study was to evaluate the performance of reusable absorbent body-worn products for adults with moderate/heavy urinary incontinence, compare their performance with a group of equivalent disposable products, and establish the need for a larger statistically robust study. SETTING AND SUBJECTS: Ten men and 4 women living in the community were recruited from locations throughout the United Kingdom. METHODS: A randomized multiple crossover design was used in which all subjects were given the opportunity to test each product. All the products were available on the UK market in January 2001. The following tools were used in the evaluation: a product performance questionnaire and a pad leakage diary. Overall opinion was used as the primary outcome indicator. RESULTS: The reusable Paddy T was the best performing product overall, outperforming the disposable products. For low leakage and good absorbency (the most important product attributes identified by the subjects), the disposable all-in-one product performed best during the day. However, the Paddy T performed best for night use. The remaining reusable products performed poorly overall. CONCLUSIONS: Reusable products for moderate/heavy incontinence remain unpopular for use in isolation. Surprisingly, a product manufactured from terry toweling, a traditional material, performed relatively well. Reusables may provide a useful alternative to disposable products in certain circumstances. The results from this study do not support a more comprehensive costly study.

Metastatic calcinosis circumscripta treated with an oral charcoal absorbent in a dog.

A five-year-old West Highland white terrier dog was admitted to the teaching hospital of Nippon Veterinary and Animal Science University due to swelling and pain of the foot pads. Examinations revealed that the dog had renal failure and calcinosis circumscripta on its foot pads. The diagnosis was metastatic calcinosis circumscripta secondary to renal failure. An oral charcoal adsorbent (Kremezin) was used to treat this condition. Following this treatment, a significant decrease in the Ca x P value (the serum calcium level x the serum phosphorus level) was observed, and the dog's condition improved dramatically. This case suggests that charcoal adsorbent (Kremezin) may be useful for treating metastatic calcinosis circumscripta in dogs.

Characterizing worker exposure to bitumen during hot mix paving and asphalt mixing operations.

A survey of bitumen exposure was carried out during hot mix paving and asphalt mixing. Four methods of characterizing and quantifying bitumen fume in inhalable particles were used: gravimetric, infrared spectroscopy, gas chromatography, and total absorbance (of the particle filter surface). Dermal deposition of bitumen was assessed by determination of cyclohexanesoluble matter in the material collected by paper or cotton pads worn on the inside of a person's wrists. The researchers studied the correlation patterns between results of these methods using pairwise correlation coefficients and principal component analysis (for air concentrations only). For hot mix paving, within- and between-worker variance components were evaluated, adjusting for area paved. Collected fume was dark in color and mainly consisted of aliphatic hydrocarbons of less than 20 carbon atoms. Inconsistent correlation patterns between dermal deposition and air levels were observed. Quantification of organic matter using either infrared spectroscopy or gas chromatography appeared to be good methods for monitoring inhalable bitumen fume. Cotton pads seem to be more promising than paper pads for measuring dermal exposure during paving. Paving workers were uniformly exposed to bitumen fume. Assessment of the uniformity of dermal exposure depended on the type of pad used. Day-to-day variability and intensity of inhalable and dermal bitumen exposures depended on area paved. The small size of the study limits the potential application of the results to the asphalt industry in general. More research is needed to develop a valid and inexpensive method of assessing total bitumen exposure.

A new dressing design for use with maggot therapy.

Maggot therapy has been used since the 1930s for treating soft-tissue wounds. Despite decades of experience with this therapy, selecting appropriate dressing materials continues to be a problem. Before initiating our maggot therapy service, we needed to develop a dressing that would (1) prevent the maggots from escaping, (2) permit oxygen to reach the maggots, (3) facilitate drainage, (4) allow inspection of the wound, (5) require minimal maintenance, and (6) be of low cost. The optimal dressing design proved to be a two-layered cagelike dressing, the bottom layer of which comprised a hydrocolloid pad, applied to the surrounding healthy skin and covered by a fine chiffon or nylon mesh. Liquefied necrotic tissue drained through the mesh and was absorbed in a top layer of gauze, which was replaced periodically. Thus it was possible to contain the maggots within the wound by means of readily available materials.

Oral cadmium chloride intoxication in mice: effects of penicillamine, dimercaptosuccinic acid and related compounds.

The antidotal efficacies of chelators during acute cadmium intoxication has previously been examined in experiments where both a soluble cadmium salt and the chelator were administered parenterally. In the present study, PA, DMSA and related compounds were studied as oral antidotes during oral CdCl2 intoxication. According to the antagonistic effects noted on mortality, peristaltic toxicity and intestinal cadmium uptake, the relative efficacies of the compounds tested were: DMSA greater than PAD greater than DMPS greater than MSA greater than PA greater than NAPA. None of the chelators induced major changes in the organ distribution of absorbed cadmium, in particular no increased cerebral deposition of cadmium. This study indicates that, in oral cadmium intoxication in humans, orally administered DMSA would be likely to offer protection against the local toxicity of cadmium in the gastrointestinal tract as well as to reduce the risk of systemic toxicity of absorbed cadmium.
