We examined the potential for some of the abnormalities of vascular endothelium found in diabetes mellitus to cause neuropathic changes. Non-diabetic rats were treated for 2 months with the cyclo-oxygenase inhibitor flurbiprofen (5 mg.kg-1.day-1) to reduce prostacyclin production, the nitric oxide synthase inhibitor NG-nitro-L-arginine (5 or 25 mg.kg-1.day-1), or combined treatment. There were dose-dependent reductions in sciatic motor and saphenous sensory conduction velocity. The two inhibitors acted synergistically, thus, the 5-6% motor conduction deficits (p < 0.01) produced by either flurbiprofen or NG-nitro-L-arginine (5 mg.kg-1.day-1) increased to 17% (p < 0.001) for combined treatment. With NG-nitro-L-arginine (25 mg.kg-1.day-1) and flurbiprofen, motor and sensory conduction velocity were reduced by 23% (p < 0.001) and 12% (p < 0.001), respectively, matching the deficits following 2-month streptozotocin diabetes. NG-nitro-L-arginine (25 mg.kg-1.day-1) and flurbiprofen together produced a 13% prolongation of the time taken for 80% hypoxic conduction failure in vitro (p < 0.05) and a 10% reduction in sciatic capillary density. A second investigation tested an alternative hypothesis that overproduction of nitric oxide was responsible for vascular-related complications in diabetes, the prediction being that NG-nitro-L-arginine (5 mg.kg-1.day-1) would prevent nerve dysfunction. However, rather than prophylaxis during 2-month streptozotocin diabetes, treatment exacerbated nerve abnormalities.(ABSTRACT TRUNCATED AT 250 WORDS)

1. Fiber types, capillary supply and other morphometrical parameters were analysed in pectoral, gastrocnemius and pronator muscles of homing and urban pigeons. 2. The two kinds of birds were analysed before and after a restrainment period of at least 5 months. 3. Only slight differences in fiber type frequencies were noted between urban and homing pigeons in control conditions. 4. The effect of restrainment on the different parameters studied was unclear in gastrocnemius and pronator muscles and negligible in M. pectoralis. 5. Mean diffusion distances for oxygen from capillaries were smaller in oxidative fibers; also, vascularization indexes were higher for these fiber types. 6. The contribution of each fiber type to total sectional ara of muscle remains stable in spite of fiber type frequencies heterogeneity.

We consider a system composed of two fluids in contact with a solid where one of these fluids dissolves the solid material. Both the dissolution process and the capillary phenomena play a role in the system evolution, which is analyzed on the basis of stability arguments for a two-dimensional geometry.
The special features of liver sinusoidal endothelium (LSE) are crucial for normal liver physiology. Cirrhotic livers, especially in primary biliary cirrhosis (PBC), are characterized by transformation of the LSE into a continuous, vascular type. The transformation is important for disease progression and explains some of the pathological hallmarks of the cirrhotic liver. Here, we investigated the presence of liver sinusoidal endothelial cell (LSEC)-reactive autoantibodies (Abs) in the sera of patients with autoimmune liver diseases, and assessed the ability of these Abs to transform LSE into vascular endothelium. Compared to healthy individuals (9%), significantly higher numbers of patients with PBC (59%; P < 0.001) and autoimmune hepatitis (AIH) (32%; P < 0.05) had Abs against LSECs. Incubation of primary LSEC cultures with F(ab')(2) fragments of anti-LSEC Abs isolated from sera of patients with PBC and AIH, induced 1) cell surface expression of vascular endothelium-associated markers, CD31, and factor VIII-related antigen; 2) significant production of fibronectin, laminin and collagen type IV; 3) loss of fenestrae, formation of tight junctions and Weibel-Palade bodies. Deposition of immunoglobulins on LSECs were found in liver biopsies of AIH and PBC patients. Thus, anti-LSEC autoAbs transform LSE into a vascular type and may therefore play an important role in the development of hepatocellular failure and portal hypertension in PBC and AIH patients.
General solutions of the capillary pressure for liquids as a function of contact angle and volume in planar close-packed spheres have been calculated numerically using Surface Evolver software. Applied pressure differences between liquid and vapor result in undulating (puckered) menisci exhibiting anticlastic curvature in the narrower spaces near particle contacts. The corresponding capillary pressures exhibit maxima with infiltration volume (minima with drainage), corresponding to critical pressures for engulfment of the spheres by the liquid (vapor). The analysis also reveals the formation of residual pendular rings of the wetting phase around particle contacts. Pendular ring formation is explored further by analyzing hexagonally packed spheres separated by 1/10 their radius. The results are discussed relative to the wide range of approximate solutions available in the literature.

UNLABELLED: The value of measuring projected area of transverse segment of hand nailfold capillary loops using computerized videophotometry was studied in 17 healthy individuals (10 women and 7 men) group A and 17 patients with diabetes type 2 (10 women and 7 men) group B with comparable ages (49.94+/-8.62 x 49.11+/-14.63 years old P=0.49). Videocapillaroscopy (VC) was performed under controlled temperature environment (24-26 degrees C). The test of post-occlusive reactive hyperemia was performed using a sphygmomanometer attached to the fourth left hand finger, 20 mmHg above maximum arterial pressure during 1 min. Images were captured each 2 s during 1 min. Time averages to reach the maximum post-ischemia area were determined as well as the area's increment percent in both groups. Both averages were compared by the Mann-Whitney test. There was no differences between area increments percent averages among controls and patients with diabetes (52.49+/-22.6 x 67.99+/-47.48% P=0.67), but the time to reach it during reperfusion was significantly increased among diabetics (5.52+/-2.96 x 18.58+/-8.08 s P<0.0001). CONCLUSIONS: changes of capillary response to ischemia may be observed among patients with diabetes through bi-dimensional measures of projected area of capillary loops transverse segment in VC using a computerized system of image analysis. Patients with Diabetes mellitus do not have significantly different maximum area increments than controls, but spend more time to reach it during reperfusion.

Rapid, robust, and economical fabrication of fluidic microchannels is of fundamental importance for the successful development of disposable lab-on-a-chip devices. In this work, we present a solvent-actuated bonding method for fabricating polymeric microfluidic devices at room temperature. A PMMA sheet with an imprinted microchannel was clamped to a blank PMMA sheet, and then 80 +/- 5 muL of acetone (bonding solvent) was introduced at one end of the fluidic channel and aspirated out at the other end. As the solvent moved down the channel, capillary forces drew a fraction of the solvent into the interstitial space between the two polymeric substrates. After aspiration, the assembly was incubated in the clamp for 5 min for effective bond formation. The quantity of the bonding solvent, its water content and flow rate, along with residence time in the channel were found to have significant impact on the bond quality and the channel integrity. Microfluidic electrophoretic separations of a 400-base DNA ladder were performed in devices fabricated using this method in less than 8 min with efficiencies routinely between 2 x 10(6) and 3 x 10(6) plates/m. The simplicity and economy of this technique make it amenable for automation and mass production, which could make polymeric substrates more attractive for single-use chemical analysis devices.

We report an investigation of the water-hydrophobic interface in well-defined nanochannels (R approximately 2-4 nm). Wetting in these systems cannot be described by classical (macroscopic) capillary theory: (1) water occupies only a fraction ( approximately 60%) of the pore volume, and (2) the capillary pressures are approximately 60-90% greater than predicted by the Laplace equation. The results suggest the presence of approximately 0.6 nm layer of low-density fluid (vapor) separating water from the hydrophobic solid.
The growing demand for new solutions to pursue the trend of micro- and nanoelectronics predicted by Moore's law is stimulating the development of new high-resolution, low-cost lithographies. Here we demonstrate that several bottom-up approaches can be used to increase the throughput of soft lithography by exploiting the enhanced hydrophilicity, the low viscosity, and the fragility of the employed materials. In particular, the customized functionalization of the involved surfaces to improve the wettability to polymer fluids and the dramatic decrease of the viscosity of polymer compounds as the temperature is increased, together with the good thermal stability of the functionalized surfaces, allow a faster filling of elastomeric channels, up to almost an order of magnitude with respect to conventional microfluidics.
