Circadian rhythm of neuropeptide Y-like immunoreactivity in the iris-ciliary body of the rat.

Neuropeptide Y (NPY)-like immunoreactivity (LI) in the iris-ciliary body of rats kept under constant darkness (DD) and in 12 h light-12 h dark cycle (LD) was determined by enzyme immunoassay. NPY-LI contents in the iris-ciliary body were found to oscillate in circadian fashion under DD and LD conditions, with a peak at about circadian time 12 (CT 12) and a trough at around CT 0. Unilateral superior cervical ganglionectomy caused a significant decrease in NPY-LI levels in the sympathectomized eye compare to the contralateral intact eye, independent of lighting phase. These results suggest the presence of an endogenous circadian rhythm in NPY-LI content in the rat iris-ciliary body, and the possible involvement of a sympathetic input from the superior cervical ganglion.

Characteristics of [3H]5-hydroxytryptamine binding to iris-ciliary body tissue of the rabbit.

PURPOSE: This study sought to identify, characterize, and localize subtypes of 5-hydroxytryptamine (HT) receptors in rabbit iris-ciliary body. METHODS: Radioligand binding assays were performed with [3H]5-hydroxytryptamine on membranes prepared from rabbit iris-ciliary bodies and on tissue sections subsequently developed by autoradiography. RESULTS: [3H]5-HT appeared to bind to a single population of receptors in membrane preparations of rabbit iris-ciliary body. The apparent affinity of the ligand (KD) was 2.19 nM, and the density of binding sites was 58.3 fmol/mg protein. Binding of [3H] 5-HT exhibited guanosine-5-triphosphate sensitivity. Competitive inhibition experiments were performed to differentiate between 5-HT receptor subtypes. A relative potency order of 5-CT > 5-HT = 8-OH-DPAT > ipsapirone > RU24969 > sumatriptan > ritanserin > ketanserin was demonstrated. The apparent inhibitory constants for the ligands tested fit with the profile expected of binding to 5-HT1A receptors. Inhibition studies with [3H] 5-HT plus 100 nM 8-OH-DPAT (which inhibits binding to 5-HT1A receptors only) representing total binding, indicated that no further displacement occurred when ligands preferentially selective for 5-HT1B, 5-HT1D alpha,1D beta, or 5-HT2C were tested. Total binding of [3H] 5-HT in tissue sections developed by autoradiography was displaced completely by 100 nM 8-OH-DPAT. Melatonin showed little affinity for the [3H] 5-HT binding sites. CONCLUSIONS: A population of 5-HT1A receptors is present in rabbit ciliary processes. There is no evidence to suggest the presence of 5-HT1D alpha, 5-HT1D beta, 5-HT2B, or 5-HT2C receptors in the iris-ciliary body.

Specific glucocorticoid receptor in the iris--ciliary body of the rabbit.

The cytoplasm of the iris--ciliary body of the rabbit contains a receptor capable of specifically binding dexamethasone. This binding protein has a high affinity for dexamethasone (average KD = 2.0 X 10(-8) M), a low capacity (average 4.8 X 10(-13) mol of steroid bound per milligram of protein), and extreme heat sensitivity; it exhibits a pattern of competition virtually identical to that obtained with glucocorticoid receptors from other tissues and shows characteristic physicochemical behavior in various salt concentrations. The demonstration of a specific dexamethasone receptor in the iris--ciliary body provides the first direct biochemical evidence that these tissues may function as a target organ for glucocorticoids.

Contact transscleral ciliary body photodynamic therapy in pigmented rabbits using verteporfin and diode laser: evaluation of treatment parameters.

PURPOSE: To evaluate the treatment parameters necessary for achieving ciliary body photodynamic damage, enough to significantly reduce IOP, using verteporfin and a diode laser. DESIGN: Animal study. METHODS: The right eye ciliary body of 30 pigmented rabbits was irradiated using verteporfin (Visudyne) and a diode laser. Photosensitizer dose ranged from 0.375 to 2 mg/kg. Three adjacent laser spots were applied 0.5 mm behind limbus at 12 o'clock position using a contact transscleral technique. The laser power was ranging from 10 to 70 mW and the duration of irradiation from 1 to 5 min per spot. The left eyes of the rabbits were used as controls. Animals were sacrificed 24 hours after the procedure and their eyes were evaluated by means of light and electron microscopy. A step-by-step approach was adopted with adjustment of experimental parameters according to histological findings. The end point was to identify the irradiation parameters necessary for induction of photodynamic damage while minimizing thermal damage. Subsequently, 10 more animals were used in order to verify the effectiveness of these irradiation parameters in reducing the intraocular pressure. RESULTS: The therapy parameters that led to photodynamic effect avoiding thermal damage were laser power of 25 mW, irradiation time of 3 min per spot, and verteporfin dose of 1 mg/kg. Transscleral ciliary body irradiation using these parameters resulted in vascular thrombosis of ciliary vessels and in substantial edema, resulting in separation of the two ciliary epithelium layers. These parameters were applied to 4 rabbits, resulting in a mean IOP reduction of 1.8 mmHg +/- 1.2 that lasted for 4 days. An increase of the laser power to 35 mW tested in 6 additional animals, resulted in mean IOP reduction of 2.2 mmHg +/- 1.2, lasting 6 days; some minimal thermal damage was seen with the later settings. CONCLUSION: The combination of verteporfin and 690 nm diode laser is effective for the induction of ciliary body photodynamic damage, which results in significant but temporary IOP reduction, after transscleral PDT in pigmented rabbits. With appropriate parameter selection, intraocular pressure reduction can be achieved while thermal damage is kept to a minimum.

[Existence of atrial natriuretic peptide in choroid, retina and ciliary body in rabbits].

The presence of atrial natriuretic factor (ANF) in choroid, retina and ciliary body of rabbit eyes has been studied. The peptide was extracted by reverse phase chromatography using Sep-Pak C18 cartridges. Immunoreactive ANF (Ir-ANF) was measured by a specific RIA. In the choroid, retina and ciliary body the ir-ANF concentrations were 444 +/- 94, 67 +/- 12 and 223 +/- 75 pg/g tissue weight (n = 16), respectively. The biological activity of the acid extract ANF was evaluated by a 125I-ANF binding assay to a retinal particulate preparation. Results demonstrate the existence of biological active ANF in choroid, retina and ciliary body of rabbit eyes. The presence of ANF and its specific receptors in the ocular tissues suggests that this hormone might play a physiological role in the intraocular fluid homeostasis.

Iris atrophy, serous detachment of the ciliary body, and ocular hypotony in chronic phase of Vogt-Koyanagi-Harada disease.

PURPOSE: To describe iris atrophy, serous detachment of the ciliary body, and ocular hypotony in a patient with chronic phase of Vogt-Koyanagi-Harada (VKH) disease. METHODS: Ocular examination and follow-up including digital infrared transillumination imaging of the iris was done in a 52-year-old woman with chronic phase of VKH disease. RESULTS: Infrared transillumination imaging showed extensive atrophy of the iris stroma and occasional pigment clumps both in the pupillary and ciliary zones of the iris, and detachment of the ciliary body in both eyes. Conventional transpupillary transillumination using white light showed only minute patchy atrophy of the pigment epithelium in the pupillary zone. Treatment did not normalize bilateral shallow retinal detachment of the posterior pole, serous detachment of the ciliary body, or severe ocular hypotony. CONCLUSIONS: Severe atrophy of the iris stroma, retinal detachment of the posterior pole, serous detachment of the ciliary body, and ocular hypotony may occur in chronic phase of VKH disease.

[Angiomorphology of the ciliary body in diabetic disease].

The authors describe the pathological changes in the vascular structures of the ciliary body occuring in diabetes mellitus. The lesions consist of a quantitative reduction in the number of vessels and in modifications of diameter and arborisation. Changes include sectoral thinning, isolated and grouped aneuryms and marked tortuosity. These findings are thought to be constant in the diabetic patient and are probably involved in the various aspects of diabetic eye disease affecting other structures besides the ciliary body. The group of vascular changes in the ciliary body in diabetes may aptly be termed "diabetic ciliopathy".

Muscarinic acetylcholine receptors in the rabbit ciliary body smooth muscle: spare receptors and threshold phenomenon.

Interactions of several muscarinic drugs with their receptors were studied in the ciliary body smooth muscles of the rabbit. The ciliary body smooth muscles responded to carbachol, a muscarinic full agonist, with concentration-dependent contractions, and the pD2 value of carbachol was 5.16 +/- 0.05. Atropine, a competitive antagonist of muscarinic receptors, produced a parallel shift to the right in the concentration-response curves for carbachol. Pilocarpine which is a well-known partial agonist on muscarinic receptors in most smooth muscles did not cause any contraction in this tissue. The drug, however, behaved as a competitive antagonist on the muscarinic receptors in the ciliary body smooth muscles. The pA2 values of atropine and pilocarpine versus carbachol obtained from the Schild plot are 8.97 +/- 0.25 and 5.17 +/- 0.09, respectively. On the other hand, arecoline and oxotremorine acted as a partial agonist in this tissue. The intrinsic activity, and pD2 and pA2 values were 0.41 +/- 0.02, 4.93 +/- 0.05 and 5.32 +/- 0.05 for arecoline respectively, and were 0.26 +/- 0.02, 5.64 +/- 0.08 and 6.12 +/- 0.16 for oxotremorine, respectively. The pA2 values of these drugs were significantly larger than the corresponding pD2 values of the drugs. The values of the negative log molar dissociation constant of carbachol, arecoline and oxotremorine estimated by the method of partial irreversible blockade of spare receptors with 3 X 10(-6) M phenoxybenzamine were 4.53 +/- 0.08, 5.20 +/- 0.09 and 6.02 +/- 0.06, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Localization and properties of an immunoreactive protein in bovine ciliary body similar to retinal S antigen.

Immunocytochemical studies showed that monoclonal antibodies to bovine retinal S-antigen recognize an immunoreactive protein present in the bovine ciliary body epithelium. This protein was purified by DEAE-agarose chromatography and found to have a M.W. of 28,000 daltons by SDS polyacrylamide gel electrophoresis. Crude and purified preparations of this protein cross-react with polyclonal antibodies to retinal S-antigen. The relative reactivity of S-antigen monoclonal antibodies to ciliary body protein was determined by ELISA. Lewis rats immunized with crude and purified ciliary body protein failed to develop experimental uveitis. The results indicate that the ciliary body protein contains an antigenic peptide domain which is similar to that of S-antigen but lacks the immunopathogenic domain.

Immunomorphologic studies of macrophages and MHC class II-positive dendritic cells in the iris and ciliary body of the rat, mouse, and human eye.

PURPOSE: To establish the presence of distinct populations of macrophages and MHC class II (Ia)-positive dendritic cells (DC) in the iris and ciliary body of the rat, mouse, and human eye. METHODS: Iris-ciliary body wholemounts from a variety of rat strains, balb/c mice, and human eyes were investigated by single and double immunohistochemistry, immunoelectron microscopy, and confocal microscopy to determine the phenotype, density, distribution, and location of macrophage and DC populations. RESULTS: Dendritiform and pleiomorphic macrophages were distributed in a regular array within the rat iris and ciliary body stroma (600 to 700 cells/mm2 or 7000 cells per iris). Ia+ DC were distributed in a similar regular network (400 cells/mm2 or 5500 cells per iris) within the iris stroma and ciliary epithelium. In the rat, a strain-dependent variation in the numbers of DC was noted, F344 rats displaying highest numbers of DC (962 +/- 398 cells/mm2) and WAG strain the lowest numbers (285 +/- 218 cells/mm2). Double color immunoperoxidase staining using anti-Ia and anti-pan specific macrophage monoclonal antibodies revealed that macrophages and Ia+ DC are distinct populations with only 5% to 15% overlap. Single immunoperoxidase of mouse iris and ciliary body using anti-pan macrophage and anti-Ia antibodies produced findings identical to those in rat. Preliminary studies of human tissue using confocal microscopy of immunostained whole irides also revealed a regular array of macrophages and MHC class II (HLA-DR)+ dendritiform cells. CONCLUSIONS: The mammalian iris contains rich networks of dendritiform-pleiomorphic macrophages and MHC class II+ DC. These findings suggest that the DC in the tissues lining the anterior chamber represent a rich network of putative antigen presenting cells and are the most likely candidates for transmitting antigen-specific signals from the anterior chamber in vivo and in experimental models such as ACAID: These observations have wide implications for the understanding of the pathogenesis of anterior and posterior uveitis.

[Trans-scleral resection of the ciliary body and choroidal melanoma].

PURPOSE: To present our preliminary results of trans-scleral resection of ciliary body and choroidal melanoma. MATERIAL AND METHODS: 16 patients aged 20-73 years (mean 56 years) were treated from February 1999 to March 2000. There were 10 patients with ciliary body tumors, 2 with choroidal tumors and in 4 patients the tumor involved ciliary body and choroid. The mean basal diameter of tumors was 9.4 mm and the mean thickness was 7.2 mm. Trans-scleral resection was performed in all cases. Adjunctive radiotherapy using 106Ru plaques was applied in 7 cases. RESULTS: Histopathologically the diagnosis of melanoma was confirmed in all cases. In 10 cases there were spindle cell melanomas and in 6 mixed type. The intra-operative complications were vitreous hemorrhage in 3 patients, vitreous loss in three and both complications in one patient. Post-operative complications occurred in 3 cases of single instances each of retinal detachment, retinal edema and haemophthalmus. After surgery visual acuity did not change in 9 eyes, improved in 2 and deteriorated in 5 eyes. CONCLUSIONS: Our preliminary results confirm the opinion that trans-scleral resection could be a valuable method in the treatment of intraocular melanoma.

Experimental autoimmune anterior uveitis. Induction with melanin-associated antigen from the iris and ciliary body.

PURPOSE: This study was designed to investigate an animal model of uveitis that resembles anterior uveitis in humans after immunization with iris-ciliary body antigen. METHODS: Male Lewis rats 6 to 8 weeks of age were immunized with the buffer- and detergent-insoluble bovine iris-ciliary body antigen mixed with complete Freund's adjuvant and pertussis toxin. Antigen was digested with various proteolytic enzymes and tested in different rodent strains for a uveitogenic response. RESULTS: Acute iridocyclitis developed in both eyes of the Lewis rat during the second week after immunization, and the pattern of inflammation was similar to acute anterior uveitis in humans, with sudden onset, localization to the anterior uvea, and spontaneous resolution. Among the strains tested, F344 rats were susceptible to experimental autoimmune anterior uveitis but Long-Evans rats were not. Experimental autoimmune anterior uveitis did not develop in any of the mice studied, nor was it induced by immunization with synthetic melanin, amelanotic bovine tissues, pigmented bovine skin, or pigmented rat and rabbit iris-ciliary body. A soluble fraction derived from bovine melanin-associated antigen (BMAA) after digestion with the proteolytic enzyme V8 protease resulted in a disease similar to that observed with intact BMAA. CONCLUSIONS: A model of anterior uveitis has been induced in the Lewis rat after immunization with bovine uveal antigen, and it resembles the acute iridocyclitis observed in humans. These results suggest that the pathogenic antigen is a melanin-associated protein(s) present within the iris-ciliary body.

Sigma1 recognition sites in rabbit iris-ciliary body: topical sigma1-site agonists lower intraocular pressure.

In this study, we examined the presence of sigma1 and sigma2 sites in the rabbit iris-ciliary body by receptor binding and investigated their effects on intraocular pressure (IOP) in albino rabbits. The iris-ciliary body has binding sites for the sigma1-site agonist [3H](+)-pentazocine (Kd = 4.6 nM; Bmax = 212 fmol/mg protein) and sigma2 sites labeled with [3H]1,3-di-o-tolylguanidine (DTG) (Kd = 8. 2 nM; Bmax = 1120 fmol/mg protein). In competition binding studies, (+)-pentazocine and the sigma antagonist NE-100 displayed high affinity for sigma1 sites (Ki = 2.1 and 2.4 nM, respectively), whereas (+)-N-allylnormetazocine (NANM) was less potent (Ki = 178 nM). Unilateral topical (+)-pentazocine (0.01-0.1%) caused a significant dose-related reduction of IOP in ocular normotensive rabbits and in the alpha-chymotrypsin model of ocular hypertension. (+)-NANM was less potent than (+)-pentazocine. Neither compound altered the IOP of the contralateral eye, and their hypotensive activity was blocked by NE-100 that, by itself, had no effect on IOP. (-)-Pentazocine, (-)-NANM, and DTG had no effect on IOP. DTG prevented the hypotensive effect of (+)-pentazocine, suggesting that it acts as a sigma1-site antagonist. sigma-Site ligands did not affect pupil diameter or cause ocular inflammation. Topical [3H](+)-pentazocine reaches the intraocular tissues within 30 min, and its uptake in the iris-ciliary body and retina was significantly reduced by topical pretreatment with NE-100, as expected for a receptor-specific agent. Reverse-phase HPLC confirmed the presence of intact (+)-pentazocine in iris-ciliary body homogenates. sigma1-Site agonists may offer a novel class of agents potentially effective in the control of ocular hypertension.

Plaque radiation therapy for malignant melanoma of the iris and ciliary body.

PURPOSE: To report on plaque radiation therapy for malignant melanomas involving the iris and ciliary body. METHODS: Twenty-two eyes (22 patients) with anterior uveal melanomas were treated with (103)Pd ophthalmic plaque radiation therapy. Transillumination and ultrasonography were used to evaluate ciliary body involvement and posterior iris extension. Plaques were placed onto the cornea to treat the anterior tumor margins. The targeted-zone included the tumor and a 2 to 3 mm tumor-free margin. After plaque removal, patients were examined at 1 day, 7 days, 4 weeks, and then every 3 to 6 months thereafter. Systemic evaluations for possible metastatic disease were performed every 6 months. RESULTS: After plaque radiation therapy, the melanomas decreased in thickness (mean 47%) in all 22 eyes, and no secondary enucleation was performed. One patient died of metastatic melanoma 5 years after radiation therapy. Despite anterior plaque placement that covered portions of the cornea, no epiphora, eyelash loss, or visually significant corneal opacities were noted. Whereas 15 of 21 phakic eyes (71%) developed secondary cataract, no eyes developed ischemic or neovascular radiation maculopathy. Four eyes were noted to have glaucoma before treatment, and two developed it after irradiation. Twenty of 22 eyes (91%) were within 2 lines of their pretreatment visual acuity. After radiation, the mean +/- SD follow-up was 56 +/- 34.4 months (range, 9 to 117 months). CONCLUSIONS: Plaque radiotherapy of melanomas involving the iris and ciliary body resulted in excellent local control with preservation of vision. Although there was high incidence of secondary cataracts, (103)Pd plaque radiation therapy resulted in no visually significant corneal opacity or radiation retinopathy.

Cholic acid accumulation by the ciliary body and by the iris of the primate eye.

Cholic acid accumulates in both the ciliary body and the iris of the primate eye during in vitro incubations at 37 degrees C for 1 hr. Incubation at 0 degrees C depresses uptake in both tissues. The washout of preaccumulated cholic acid occurs some 3.4 times faster from the iris than from the ciliary body. The mechanism of cholic acid accumulation in both tissues is less sensitive to inhibition by high iodipamide concentrations and also is less sensitive to inhibition by high hippurate concentrations than the mechanism of p-aminohippurate (PAH) accumulation. Therefore, although overlap may exist, the cholic acid--uptake mechanism differs from the PAH-uptake mechanism in both the primate ciliary body and the primate iris.

The ciliary body--the third organ found to synthesize indoleamines in humans.

Indoleamines are associated with circadian rhythms in pineal gland and retina. Because the ciliary epithelium has an embryonic origin similar to that of pineal gland and retina, and intraocular pressure shows circadian variations, indoleamines were searched for in aqueous humor and ciliary body in humans. In aqueous humor, serotonin, 6-hydroxymelatonin, and melatonin were simultaneously detected and measured using high-performance liquid chromatography with electrochemical detection. The concentration was 48.7 +/- 10.9 ng/ml for serotonin, 0.47 +/- 0.8 ng/ml for melatonin, and 13.9 +/- 7.7 ng/ml for 6 hydroxymelatonin. In ciliary bodies from freshly enucleated human eyes, tryptophan, 5-hydroxytryptophan, serotonin, and 5-hydroxyindoleacetic acid were detected using high-performance liquid chromatography with simultaneous fluorescence- and electrochemical detection. Finally, the enzymatic activities of arylalkylamine N-acetyltransferase (NAT) and hydroxyindole-O-methyltransferase (HIOMT), enzymes indispensable in the synthesis of melatonin, were measured. The NAT activity was 273 +/- 25 pmol/mg protein/hour and that of HIOMT, 13520 +/- 50 pmol/mg protein/hour in ciliary body. Comparison of these activities (NAT versus HIOMT) permits the suggestion that NAT is a limiting enzyme in serotonin metabolism in this tissue. These findings indicate that a circadian rhythm of indoleamines exists in human aqueous humor and that the human ciliary body is the third organ, after the pineal gland and the retina, found to synthesize indoleamines in humans.

Participation of pigment epithelium in ocular immune privilege. 3. Epithelia cultured from iris, ciliary body, and retina suppress T-cell activation by partially non-overlapping mechanisms.

PURPOSE: The ocular microenvironment is immunosuppressive and anti-inflammatory. Since various ocular pigmented epithelia contribute to this microenvironment, we studied the relative capacities of pigment epithelial (PE) cells cultured from the iris, ciliary body, and retina of mouse eyes to suppress T-cell activation in vitro. METHODS: Pigment epithelium was cultured from iris, ciliary body, and retina for 14 days, then assayed for the capacity, directly or across transwell membranes, to suppress mixed lymphocyte reactions and anti-CD3 stimulation of T cells. Potential molecules responsible for suppression were examined by attempting to block suppression with appropriate reagents, and by using mice with pertinent mutant or disrupted genes. RESULTS: We found that PE cells from all three ocular tissue sources profoundly suppressed T-cell activation in vitro. While iris PE suppressed poorly when separated from T cells by a transwell membrane (implying that cell contact is necessary), retina PE suppressed fully even in the presence of such a membrane (implying that soluble factors were responsible). Ciliary body PE used both soluble factors as well as cell contact to achieve suppression. Suppression could not be ascribed to TGFbeta, IFNgamma, TNFalpha, CD48, or ICAM-1, or to interactions between CD40 and CD154, or CD95 and CD95 ligand. Galectin-1, a galactoside-binding protein, was found to be expressed on all cultured PE cells, but only retinal pigment epithelium (RPE) from galectin-1 KO mice showed reduced capacity to inhibit T-cell activation. CONCLUSIONS: Cultured pigment epithelia from iris, ciliary body, and retina comparably suppress T-cell activation in vitro, but by partially different mechanisms. Although RPE cells suppress in part through expression of galectin-1, the molecular mediators of suppression by iris and ciliary body PE remain to be identified.

The effect of indocyanine green pretreatment on the parameters of transscleral diode laser thermotherapy-induced threshold coagulation of the ciliary body.

BACKGROUND AND OBJECTIVE: To study the effect of indocyanine green (ICG) pretreatment on threshold parameters of transscleral diode laser thermotherapy-induced threshold coagulation of the ciliary body. The procedure was termed 'cyclothermotherapy' based on the long duration (15-60 seconds) of diode laser application. STUDY DESIGN/MATERIALS AND METHODS: The right eyes of nine young adult New Zealand white rabbits underwent transscleral cyclothermotherapy (TCT, Group 1), TCT following ICG pretreatment (Group 2), and external manipulation of the ciliary body alone (Group 3). Rabbits were sacrificed after 24 hours; specimens were evaluated with gross examination and light microscopy. RESULTS: Thresholds were 30 J/cm2 (TCT) and 4.5 J/cm2 (TCT with ICG). Widespread structural damage was seen in the ciliary processes and the ciliary body in Groups 1 and 2. In Group 3, external manipulation of the ciliary body caused hemorrhage and structural damage confined to the ciliary processes. CONCLUSION: ICG pretreatment reduced the energy necessary to cause a threshold lesion with TCT in nonpigmented rabbits.
