Coordinate expression and trans presentation of interleukin (IL)-15Ralpha and IL-15 supports natural killer cell and memory CD8+ T cell homeostasis.

The high affinity interleukin (IL)-15 receptor, IL-15Ralpha, is essential for supporting lymphoid homeostasis. To assess whether IL-15Ralpha's role in vivo is to trans present IL-15, we generated mixed bone marrow chimera from IL-15Ralpha- and IL-2/15Rbeta-deficient mice. We find that IL-15Ralpha-competent, IL-2/15Rbeta-deficient cells are able to support IL-15Ralpha-deficient natural killer (NK) and memory CD8+ T cells, thus ruling out secondary signals on these cells and demonstrating that IL-15Ralpha-mediated presentation of IL-15 in trans is the primary mechanism by which IL-15Ralpha functions in vivo. Surprisingly, using IL-15- and IL-15Ralpha-deficient mixed chimera, we also find that IL-15 and IL-15Ralpha must be expressed by the same cells to present IL-15 in trans, indicating that IL-15Ralpha is required on a cellular level for the elaboration of IL-15. These studies indicate that IL-15Ralpha defines homeostatic niches for NK and memory CD8+ T cells by controlling both the production and the presentation of IL-15 in trans to NK and CD8+ memory T cells.

PLURIPOTENT STEM CELL FUNCTION OF THE MOUSE MARROW "LYMPHOCYTE".

Bone marrow from normal and polycythemic mice was filtered through glasswool columns to remove cells other than "lymphocytes." For a given number of nucleated cells, filtered marrow was more efficient than the original marrow in repopulating the spleen of an isogenic recipient previously exposed to lethal irradiation. The proliferative capacity of both the filtered and unfiltered marrow suspensions appeared to be a constant function of the number of small and medium "lymphocytes" present and not of any other cell type.

BLEPHARISMA INTERMEDIUM: ULTRAVIOLET RESISTANCE OF PIGMENTED AND ALBINO CLONES.

An albino mutant of Blepharisma intermedium is much more sensitive (on the basis of time to regeneration of transected cells) to short-wavelength ultraviolet radiation than the wild type (containing a reddish pigment) from which the mutant derived. In the wild type, absorption of ultraviolet light by the pigment present in the outer surface of the cell presumably reduces the intensity of the radiation impinging on the vulnerable interior.

Immunologic inhibition of ultraviolet radiation-induced tumor suppressor cell activity.

Long-term exposure of C3H mice to ultraviolet radiation resulted in the formation of suppressor T cells that recognize ultraviolet radiation-induced regressor skin cancers as a class before the appearance of overt tumors. Administration of monoclonal antibodies to the product of the I-Jk subregion of the major histocompatibility complex or low doses of cyclophosphamide in vivo inhibited the development or activity of these cells. This activity of the monoclonal antibody was eliminated by adsorption on B10.BR (I-Jk) but not B10.D2 (I-Jd) splenocytes. These findings provide evidence that elements expressing the I-J determinant are important in regulating the host response prior to the overt development of ultraviolet radiation-induced skin cancers and suggest novel therapeutic approaches to malignancies or other diseases involving suppressor T cells in their pathogenesis.
