There has been growing concern about the potential threat of hormone-disrupting chemicals like bisphenol A to various aspects of animal and human health. We studied the effects of bisphenol A on the Cl(-) secretion in human airway epithelial Calu-3 cells. Pretreatment with bisphenol A (IC(50) = 60 microM, for 30 min) prevented isoproterenol (10 nM)-generated short-circuit current (I(sc)) more potently than 17beta-estradiol or tamoxifen (IC(50) = 1 mM). 5'-Nitro-2-(3-phenylpropylamino) benzoate-sensitive apical conductance potentiated by isoproterenol was not affected by the pretreatment with either of these estrogenic compounds. The effects of bisphenol A were simulated in I(sc) responses to forskolin (10 microM) and 8-bromo-cAMP (1 mM). Nystatin permeabilization of Calu-3 monolayers revealed that bisphenol A attenuated 8-bromo-cAMP-induced basolateral K+ current, which is sensitive to clotrimazole (30 microM) and insensitive to charybdotoxin (100 nM), without affecting the apical Cl(-) current. Bisphenol A, but neither 17beta-estradiol nor tamoxifen, interrupted the charybdotoxin-sensitive component of I(sc) stimulated by 1-ethyl-2-benzimidazolinone (1-EBIO; 500 microM). The inhibitory effects of bisphenol A on these Cl(-) secretory stimuli were remarkable when applied to the apical rather than the basolateral membrane. Alternatively, long-term incubation of bisphenol A (1 microM; 12-72 h) had no discernible effect on isoproterenol- and 1-EBIO-induced Cl(-) secretion. These findings indicate that short-term exposure to bisphenol A attenuates transepithelial Cl(-) secretion through inhibition of both cAMP- and Ca(2+)-activated K+ channels on the basolateral membrane, interacting from the cytosolic surface in Calu-3 cells.

Migration from multi-layer laminated film pouches intended for retort foods was investigated through HPLC analysis with a fluorescence detector, and measurements of residue on evaporation, consumption of potassium permanganate and total organic carbon. HPLC analysis revealed that the levels of migrants in oil and the water which were heated in the pouches (121 degrees C, 30 min) were ten times of those in the corresponding official simulants under the official conditions; n-heptane (25 degrees C, 60 min), and water (95 degrees C, 30 min). Bisphenol A diglycidyl ether and related compounds were found in the oil and the water heated in the pouches, as well as in the simulants. These compounds were thought to have been present in the adhesive between the laminated films, and migrated through the food-contact film of the package. Consumption of potassium permanganate and residue on evaporation of the heated water were ten times of those of the water simulant, while the total organic carbon level of the heated water was several-hold greater than that of the water simulant. In addition, migrant levels per surface area of the pouch were one-fourth of the concentrations per content volume of the pouch. Since compliance with the legal limits is evaluated based on the concentration per surface area, real migration into foods would be underestimated by a factor of another four.
Composite carbon/epoxy resin techniques for restorative dentistry have improved with the development of various composite resins classified according to fiber reinforcement, such as short fibers, woven materials, or long unidirectional fibers. This study of the mechanical properties with three-point flexion enabled comparison of the flexural strengths. The modulus of elasticity of different composite resin materials was determined so that the appropriate reinforced composite resin could be selected for specific clinical conditions.
To characterize the specificity of synthetic compounds for peroxisome proliferator-activated receptors (PPARs), three stable cell lines expressing the ligand binding domain (LBD) of human PPARalpha, PPARdelta, or PPARgamma fused to the yeast GAL4 DNA binding domain (DBD) were developed. These reporter cell lines were generated by a two-step transfection procedure. First, a stable cell line, HG5LN, expressing the reporter gene was developed. These cells were then transfected with the different receptor genes. With the help of the three PPAR reporter cell lines, we assessed the selectivity and activity of PPAR agonists GW7647, WY-14-643, L-165041, GW501516, BRL49653, ciglitazone, and pioglitazone. GW7647, L-165041, and BRL49653 were the most potent and selective agonists for hPPARalpha, hPPARdelta, and hPPARgamma, respectively. Two PPAR antagonists, GW9662 and BADGE, were also tested. GW9662 was a selective PPARgamma antagonist, whereas BADGE was a low-affinity PPAR ligand. Furthermore, GW9662 was a full antagonist on PPARgamma and PPARdelta, whereas it showed partial agonism on PPARalpha. We conclude that our stable models allow specific and sensitive measurement of PPAR ligand activities and are a high-throughput, cell-based screening tool for identifying and characterizing PPAR ligands.

Prenatal exposure to bisphenol A (BPA), an estrogenic compound, has been shown to alter postnatal development at an environmentally relevant exposure level. To elucidate these low dose effects of preimplatation exposure to BPA, two-cell mouse embryos were cultured with 1 nM BPA. More embryos exposed to 1 nM BPA than controls reached the blastocyst stage. When the blastocysts with or without BPA exposure were transferred to uterine horns of pseudopregnant recipient mice not treated with BPA, the number of pups per litter and body weight at birth did not differ. At weaning on postnatal day 21, however, pups treated with 1 nM BPA during the preimplantation period were significantly heavier than controls. These findings suggest that BPA may not only affect early embryonic development even at low, environmentally relevant doses, but also may exert late effects on postnatal development.

The reactions of four epoxides used as monomers for food contact plastics were studied in the food simulants distilled water, 15% aqueous ethanol, 3% aqueous acetic acid and olive oil. Loss of the parent substance and formation of products was monitored to establish the transformation products to be expected in each simulant following migration testing of plastics. Each epoxide was stable in olive oil but suffered extensive loss in the three aqueous simulants. Reaction half-lives were from < 1 to 10 h in aqueous acetic acid, 25-63 h in distilled water, and 33-87 h in aqueous ethanol simulant. Hydrolysis to the diol was the main reaction pathway. Epoxide ring opening in aqueous ethanol simulant gave the diol and also the diol monoethyl ether. It is concluded that, for aqueous simulants and by implication for most foods, testing plastics against specific migration limits for epoxides is not likely to give reliable results due to their reactivity. The present EC mode of control for these reactive monomers, via compositional limits in food contact plastics, is more practical since the hydrolysis products are less toxic than the parent epoxide.

The chemiluminescent reaction of lucigenin with various biological substances has been studied. Chemiluminescence of lucigenin is produced by the addition of organic reducing compounds to lucigenin in alkaline solution. On the basis of this reaction, we have developed highly sensitive chemiluminescent methods for the detection of enzyme immunoassay, especially using alkaline phosphatase (ALP) as a label enzyme. The detection limits of enzyme was 10(-19)-10(-20) mol per assay.
