[Application of confocal microprobe Raman spectroscopy in the analysis of interfaces].

Confocal microprobe Raman spectroscopy has been applied to the study of liquid/gas and solid/liquid interfaces. With the help of the resonance Raman effect, the aggregation process of TSPP has been monitored at the liquid/gas interface using Raman spectroscopy, which is shown to have very different behavior compared with that in the bulk. The solution layer analysis has been performed in the interfacial region above a Pt electrode under reaction, and the solution composition is monitored in situ during the electrochemecial reaction. Taking the advantage of high spatial resolution of the confocal microscope, the surface enhanced Raman imaging has been performed on the electrodeposited gold surface.

Vibrational spectroscopic study of the contents of a chest excavated from the wreck of the HMS Pandora.

The FT-IR and Raman spectroscopic analysis of a red powder found in a chest from an officer's cabin during the excavation of the wreck of the 18th Century frigate HMS Pandora have confirmed that the pigment is cinnabar, mercury(II) sulphide. Weaker signals in the Raman spectrum are assignable to a proteinaceous material, such as collagen, typical of a degraded vellum or parchment. Comparison of the Raman spectra with that of a pigmented seal from a 1786 Lieutenant's commission demonstrated that the beeswax component of the seal was not observable.

The monothiocyanate complexes of chromium ion(III) on the silver electrode by the surface enhanced Raman scattering.

Two adsorbate forms of the monothiocyanate complex of chromium ion on the silver electrode are identified in the surface enhanced Raman scattering. The spectroscopic, especially the electronic, properties of these two forms under different applied voltages on the electrode and under both 632.8 and 514.5 nm excitations are studied by the bond force constants (bond orders) and the bond polarizability derivatives which are retrieved from the Raman intensities by an algorithm developed by Wu and co-workers. The work shows the potential of this approach to the surface enhanced Raman scattering and other fields like resonance Raman that involve vibronic coupling.

Off-resonant fifth-order response function for two-dimensional Raman spectroscopy of liquids CS2 and H2O.

The off-resonant fifth-order response functions for two-dimensional (2D) Raman spectroscopy of molecular liquids CS2 and H2O are investigated by using molecular dynamics calculation. This spectroscopy, able to deal with a phase space dynamics, shows the existence of nodal lines in several polarization tensor elements [see L. Kaufman et al., Phys. Rev. Lett. 88, 207402 (2002) for experimental results]. The nodal property is found to arise from the dynamical couplings among rotational modes, not accounted for in a normal mode picture. The effects of anharmonicities and "mode coupling through polarizability" are also investigated by comparing the 2D Raman signal with a constant temperature velocity reassignment echo method.

The environment of [2Fe-2S] clusters in ferredoxins: the role of residue 45 probed by site-directed mutagenesis.

The biochemical and biophysical properties of the Ala45Ser mutant of the [2Fe-2S] ferredoxin from vegetative cells of the cyanobacterium Anabaena sp. 7120 are described. This novel protein, which incorporates the residue present in many higher plant ferredoxins into the analogous position of a typical cyanobacterial ferredoxin, was prepared to probe the origin of the characteristic spectrochemical and functional differences between the ferredoxins from these two sources. The variant protein was produced by site-directed mutagenesis and was expressed as the holoprotein in Escherichia coli. Although the UV-vis spectrum of the Ala45Ser mutant was indistinguishable from that of the wild-type (WT) protein, the circular dichroism (CD) spectrum of the mutant was distinct and similar in appearance to that of spinach ferredoxin, which possesses a Ser residue at the analogous position. The values of the principal g factors of the EPR spectrum of the dithionite-reduced mutant protein differed from those of the WT spectrum and resembled those of plant ferredoxins containing serine at position 45. Analysis of the mutant EPR spectrum according to the method of Blumberg indicated greater covalent interactions between the localized ferrous site of the cluster and the protein matrix relative to the WT protein. The resonance Raman spectrum of Ala45Ser Anabaena ferredoxin was distinct from the spectrum of the WT protein and showed exceptional similarity to the spectrum of higher plant ferredoxins, such as spinach ferredoxin. The mutant protein spectrum displayed considerably greater deuterium dependent isotope shifts for bands ascribed to terminal Fe-S stretching modes than did the WT spectrum.(ABSTRACT TRUNCATED AT 250 WORDS)

[Analysis of water content in rat brain using Raman spectroscopy].

Laser Raman spectroscopy has not been applied to brain tissue. However, this method can possibly analyze the constituent elements of living tissue of cellular and molecular level easily, and this might be essential for further brain research. I attempted to analyze the water content of brain tissues, which was measured by Raman spectrum of CH and OH groups, in normal and edematous rat brain tissues. The experimental results indicated that the peak intensity of CH group originated from protein, 2935 cm-1 in wave number, was constant in normal, dried and edematous brain tissue. Therefore it could be used as internal standard. As the peak intensity of OH group, 3390 cm-1 in wave number, was originated from water, the relative amount of water content in the brain tissues could be evaluated by OH/CH value. The OH/CH value was not significantly different between the cortex and the thalamus, but it was significantly higher in the gray matter (thalamus) than in the white matter (pons). The OH/CH value was decreased in dried specimens. In experimental edematous brain tissues, which was produced by intraperitoneal injection of organic tin, the OH/CH value was evidently increased in the thalamus but not much changed in the pons. This results were well compatible with cytotoxic edema, which was reported to be produced by organic tin. It is suggested that laser Raman spectroscopy can be used for brain edema research as one of the strong weapon, in analyzing water content of brain tissues on a cellular level.

Conformation of pardaxin, the toxin of the flatfish Pardachirus marmoratus.

Pardaxin is the toxic component isolated from the secretion of the Red Sea flatfish Pardachirus marmoratus. Pardaxin has attracted considerable attention recently because of its use as shark repellent. Conformational information regarding the peptide backbone, disulfide bonds, and tyrosine chromophores was obtained using Raman and circular dichroism (CD) spectroscopy. Analysis of the Raman spectra indicates that the toxin consists of 39% alpha-helix, 23% beta-structure, and 39% random coil. The occurrence of a band at 510--512 cm-1 indicates the conformational geometry of the disulfide C-C-S-S-C-C linkages to be gauche-gauche-gauche. From the lack of a peak in the 2500--2700 cm-1 region we concluded that all half-cystines are involved in disulfide linkages. The far-ultraviolet CD spectrum of pardaxin showed positive 196 and negative 208 and 222 nm bands. The best fit secondary structure based on the CD ellipticities was found to be: 23% alpha-helix, 21% beta-structure, and 56% random coil. Under highly acidic and alkaline conditions the far-ultraviolet pardaxin spectra showed extensive, reversible loss of its helical structure. Due to certain biological characteristics of pardaxin the possible influence of salt on pardaxin's conformation was examined. No evidence of conformation perturbation was detected in either the CD or Raman spectra of pardaxin.

Glass distilling collector applied for HCN recovery from submerged culture broth and fruiting body of Pleurotus eryngii for identification and quantification.

Detection and surveillance of food commodities containing cyanide is a crucial issue of food safety. In this study, five strains of Pleurotus eryngii (P. eryngii) were grown in submerged culture of yeast malt broth (YMB) with the suspected production of HCN. A safety-warranted U-bent glass distilling collector with three enlarged bulbs on each arm was designed to recover the broth vapor. When AgNO(3) solution was used as an absorbent to interact with the vapor, a white precipitate was formed. The precipitate was isolated and identified as AgCN by FT-Raman spectroscopic analysis. When the absorbent was substituted by KOH, after evaporation to dryness, dissolved in D(2)O, and followed by (13)C-NMR analysis, a KCN spectrum was achieved. Formation of AgCN and KCN confirmed HCN production in the broth by P. eryngii. When a sodium picrate solution (1.4%) was used as an absorbent and various authentic KCN solutions were applied for distillation and followed by absorbance determination at 510 nm, a linear dose-dependent relationship was obtained and the procedure was applied for HCN quantification of the marketed P. eryngii mushrooms (fruiting body). As estimated, 67.3% of the products contained HCN less than 1.0 mg/kg, 17.3% between 1.0 and 2.0 mg/kg, and 15.4% higher than 2.0 mg/kg. When the mushrooms were sliced and cooked in water at 95 degrees C for 6 min, 89.1% of the original HCN was lost. When the P. eryngii strains were respectively grown by submerged cultivation in YMB or YMB supplemented with 2.5% glycine for 16 days, HCN content was slightly higher in the latter than in the former for each strain.
