[Syndrome differentiation through drug effects in mapping the two regulatory pathways of gene networks in Shen deficiency syndrome].

OBJECTIVE: To study the regulatory pathways and rules of the gene networks in Shen deficiency syndrome. METHODS: Tissues of hypothalamus, pituitary, adrenal, lymphocyte, bone, liver and kidney were taken as samples from 4 months' and 24 months' old SD rats and rats after treatment with Epimedium flavonoids (EF), differences of gene expression profile in Shen deficiency syndrome were studied repeatedly with gene chip rat expression set U230 2.0 array from Affymetrix Co. RESULTS: Gene expressions in the aged rats all decreased including neurotransmitter of gamma-aminobutyric acid (gammaGABA), gonadotropin releasing hormone (GnRH), thyrotropin-releasing hormone (TRH), thyroid stimulating hormone (TSH), growth hormone-releasing hormone receptor (GHRH), insulin-like growth factor (IGF) and binding proteins (IGFBP) in hypothalamus, pituitary and adrenal (HPA axis), cell growth-related gene, growth factor related protein, and immune regulatory genes such as interferon gamma (IFN-gamma), interleukin 4 (IL-4) and interleukin 6 (IL-6) in lymphocytes, parathyroid hormone (PTH), calcitonin, procollagen, collagen, connective tissue growth factor in bone, and oxidative phosphorylation genes such as cytochrome P450 and NADH dehydrogenase, glutamate dehydrogenase related with protein metabolism, and glucose-6-phosphatase related with glucose metabolism in liver, most of which were up-regulated after treatment with EF as well as genes related with ageing and cell cycle, such as cyclin B, metabolism related genes and proteins of sodium and chloride channel in kidney. CONCLUSION: Dysfunction of the two regulatory pathways of gene networks as nerve-endocrine-immunity and nerve-endocrine-bone metabolism exists in Shen deficiency syndrome differentiated by effects of drugs, which could be improved by strengthening Shen therapy.

[The influence of acupuncture on interleukin 2 interferon-natural killer cell regulatory network of kidney-deficiency mice].

The influence of acupuncture on IL2-IFN-NKC regulatory network was investigated, experiments were performed on kidney-deficiency mice received the treatments of acupuncture of "Zusanli" point. The results showed that the interleukin: (IL:), natural killer cell (NKC) activity and interferon (IFN) from the kidney-deficiency mice were lower than that from the normal mice. Acupuncture increased the levels of all the IL2 and NKC activity, acupuncture promoted Newcastle disease virus (NDV) inducing the IFN of kidney-deficiency mice, but also induced the IFN directly. The results suggests that acupuncture could make positive adjustment to IL2-IFN-NKC regulatory network. It provided new theoretical basis for the principles of acupuncture theory on IL2-IFN-NKC regulatory network put forward in recent years.

[Plasma cortisol concentration and blood leukocyte content of glucocorticoid receptors in patients with deficiency-cold vs deficiency-heat syndromes].

Plasma cortisol concentration and blood leukocyte content of glucocorticoid receptors (GCR) were assayed in 20 patients with deficiency syndromes, 10 cold in property (deficiency-cold), the other 10 hot in property (deficiency-heat), and also in 10 healthy individuals as normal control for the purpose of investigating the nature of cold and heat syndromes. As a result, the cases of deficiency-cold syndrome (DCS) had a normal concentration of plasma cortisol but a lowered content of GCR in leukocytes when compared with the normal control (P less than 0.05); the cases of deficiency-heat syndrome (DHS) had a higher concentration of plasma cortisol than the normal control (P less than 0.05) and a slightly higher content of GCR in leukocytes. It was concluded that the DCS is characterized by diminished biological effects of adrenocortical activity, while the DHS, by augmented biological effects of adrenocortical activity.

[The beneficial effect of qigong on the ventricular function and microcirculation in deficiency of heart-energy hypertensive patients].

Ultrasonic cardiogram was performed on 120 aged subjects. Experiment showed that the left ventricular function in the hypertensive aged group (n = 80) was lower than that in the aged group (n = 40), while the left ventricular function in the deficiency of heart-energy hypertensive patients (n = 46) was the lowest in the non-deficiency of heart-energy hypertensive patients (n = 34). After practising Qigong for 1 year, the cardiac output (CO) was increased, the total peripheral resistance (TPR) was decreased, ejection fraction (EF) mitral valve diastolic closing velocity and mean velocity of circumferential fiber shortening (mvcf) tended to be increased. The results indicated that Qigong had a regulatory effect on haemodynamic alteration as well as on improvement of left ventricular function. Nailfold microcirculation detection of 120 aged subjects was made. It found that hypertension had an accelerating effect on the disturbance of microcirculation. The incidence of disturbance of microcirculation was 73.91% in the deficiency of heart-energy hypertensive patients, after 1 year Qigong practice, the incidence of disturbance of microcirculation was 39.13% (P less than 0.01). The result suggested that Qigong had an effect to improve the disturbance of microcirculation. The above data indicate that Qigong can benefit heart-energy and regulate the blood channel.

[A study of classification and treatment of combined traditional Chinese medicine and Western medicine of 69 cases of aplastic anemia].

Hemopoietic progenitor cells (BFU-E, CFU-E and CFU-GM) of 69 cases of aplastic anemia were cultured in vitro while the sensitivity of CFU-E and BFU-E to androgen was assayed. The suppressive effect of peripheral blood mononuclear cell (PBMNC) from patients on normal CFU-GM growth was also studied. Hence 69 cases of aplastic anemia were divided into three types: 10 cases of stem cell deficiency type, 30 cases of immuno-mediated type and 29 cases of androgen response type. According to above classification, different treatment programs were practiced using both TCM and western medicine. In the androgen response type, androgen and TCM were used and the effective rate reached 92.3%. In the immuno-mediated type, in addition to TCM. Immunosuppressive agents were cautiously used and the effective rate was 70.6%. As for the stem cell deficiency type, 10 patients' conditions were usually very severe so comprehensive therapeutical means were adopted and 3 cases were improved. The overall response rate of these three was 73.2%. These results showed that the treatment program under the direction of classification have greatly improved the curative effect.

[Pharmacokinetic characteristics of ferulic acid in patients with different syndromes of deficiency of spleen qi, stagnation of liver qi and spleen deficiency, and excess of stomach heat].

OBJECTIVE: To investigate the nature of syndrome of traditional Chinese medicine by means of pharmacokinetic (PK) method. METHODS: Twenty-one healthy volunteers, 20 patients with syndrome of stagnation of liver qi and spleen deficiency, 22 patients with syndrome of deficiency of spleen qi and 19 patients with syndrome of excess of stomach heat were included and administered to take Jiawei Xiaoyaosan Recipe (JWXYSR). The serum PK parameters of ferulic acid (FA) were examined by high performance liquid chromatography (HPLC) method. RESULTS: The absorption rate constant (alpha) and the elimination rate constant (beta) were both decreased while the apparent first-order absorption constant (K(a)) was enhanced significantly in the patients with syndrome of deficiency of spleen qi; the alpha, beta and Ka were all reduced in the patients with syndrome of stagnation of liver qi and spleen deficiency; the beta and K(a) were increased in the patients with syndrome of excess of stomach heat, as compared with the corresponding PK parameters in the healthy volunteers (P<0.01). CONCLUSION: The PK analysis of FA in the patients with syndrome of deficiency of spleen qi shows that the absorption rate is accelerated, and both the distribution and elimination rates are slowed down. The absorption, distribution and elimination rates of AF are all slowed down in the patients with syndrome of stagnation of liver-qi and spleen deficiency, while the absorption and elimination rates of AF are both accelerated in the patients with syndrome of excess of stomach heat. There are obvious differences in the PK characteristics among these three syndromes.
