[Isolation of a concentrated purified preparation of the transmissible gastroenteritis virus of swine and a study of its antigenic properties].

Pretreatment of the transmissible gastroenteritis virus of swine (TGE) (coronavirus) with trypsin increased the yield of the infectious virus approximately 100-fold when propagated in roller cultures of SPEV cells, the time required for the development of marked CPE being twice shorter than without treatment. The method of concentration and purification of TGE virus is described which yields a preparative amount of highly purified virions retaining their structure. Such preparations were used for immunization of guinea pigs. It is concluded that purified preparations of TGE virus possess marked antigenic potency and may be used for generation of highly active and specific hyperimmune sera.

[Isolation of the Fomédé virus from Chiroptera, Nycteris nana, in the Republic of Guinea].

In October 1978, a new virus named Fomédé was recovered from the bat, Nycteris nana, during the studies of arbovirus infections in Guinea (West Africa). When the virus was tested, it did not fit any prototype patterns. These findings have led to a presumption that the ch. s. 654 strain of virus is a previously unknown virus due to its biological, physico-chemical and antigenic patterns.

[Dependence of the synthesis of virion structures in the tick-borne encephalitis virus on the cell culture type].

Variations in synthesis of antigenic structures are observed in tick-borne encephalitis virus replication in cell cultures of different origin. In a number of cell cultures: pig and Syrian hamster embryo kidney cells, as well as in Chinese striped hamster cells and Tasmanian rat kangaroo cells, virion antigens (VA) are synthesized which differ in the direction of movement in the electric field, namely, anode and cathode VA. In other cell cultures: green monkey kidney, barking deer kidney, and human fibroblasts, only cathode VA is synthesized. In chick embryo fibroblast cultures, in addition to the above-mentioned VA, considerable amounts of a VA which does not move in the electric field are synthesized. In all the cultures, a low molecular nonvirion antigen (NA) is actively produced, the virus-containing fluids of human fibroblast cultures and Chinese striped hamster kidney cell cultures contain lower amounts of high molecular NA, while rat kangaroo and green monkey kidney cell cultures have no high-molecular NA of tick-borne encephalitis virus.

[Experimental induction of lympholeukemia in calves by a purified preparation of bovine leukemia virus].

The leukemogenic properties of bovine leukemia virus (BLV), isolated from blood plasma, milk and short-term cultures of blood-circulating leukocytes were studied. A complex life-time examination of 14 calves inoculated with a preparation of BLV established leukemia in 4 animals; it was suspected in 3 calves. By the present time leukemia has been morphologically established in 6 out of 13 calves and one was a suspect. Positive results point directly to the viral etiology of bovine leukemia.

Antigenic characterization of the inclusion body hepatitis virus.

Proof that inclusion body hepatitis of chickens (IBHC) is caused by a virus (IBHV) was established in 1973. Characterization of IBHV has been conducted by employing the standard criteria for virus classification. The results have indicated that IBHV is a member of the avian adenovirus group. However, no antigenic relationship was found between IBHV and two other avian adenovirus isolates (CELO and Indiana C) as observed by hemagglutination (HA) and virus-serum neutralization tests. Another adenovirus isolate was found to be serologically different from IBHV (Tipton strain) but it incited typical IBHC in susceptible chicks. A novel antigen, detectable by the agar gel immunodiffusion test, was demonstrated in livers from chicks infected experimentally with IBHV. This antigen was able to give a specific precipitin reaction with serums from chickens previously exposed to IBHV. Repeated embryo passage of IBHV lowered its pathogenicity for susceptible chickens. Protection of chickens against IBHV challenge was obtained by vaccination with high embryo passage IBHV (Tipton strain). It was also found that subcutaneous administration of the vaccine virus induced a better immune response than the eye-drop route as observed by the virus-serum neutralization index obtained.
