Chronic haloperidol or fluphenazine: effects on dopamine metabolism in brain, cerebrospinal fluid and plasma of Cercopithecus aethiops (vervet monkey).

Chronic treatment with haloperidol for 3 to 5 weeks (0.5 mg/kg, daily) resulted in significant increases of homovanillic acid (HVA) content in dorsal and orbital frontal cortex and in cingulate cortex. No change in HVA was seen in the olfactory cortex, basal ganglia, cisternal cerebrospinal fluid or plasma of animals chronically treated with haloperidol. Treatment with a single weekly dose of fluphenazine decanoate (5 mg/kg) for 3 weeks resulted in increased HVA levels in all the above brain regions, cisternal cerebrospinal fluid and plasma. Moreover, the fluphenazine-treated group had a significantly higher incidence of extrapyramidal side effects than the haloperidol-treated group. It is concluded that chronically increased dopamine metabolite production in the basal ganglia but not in cortex is reflected by increases in the HVA level of cerebrospinal fluid and plasma and is accompanied by severe extrapyramidal side effects.

Nutritional effects on blood lipid and HDL cholesterol concentrations in two subspecies of African green monkeys (Cercopithecus aethiops).

The African green monkey has previously been found to be a promising model for the study of atherosclerosis. We have compared the plasma and HDL cholesterol response to dietary manipulation in the two subspecies of African green monkeys (vervets and grivets) most often imported for biomedical research purposes. Twenty vervets and 20 grivets were fed, in succession, diets containing safflower oil, butter, or lard as the principal dietary fat at a level of 40% of calories. Ten animals of each subspecies were fed the diets without added cholesterol (control groups) and 10 were fed diets with either added crystalline cholesterol (safflower oil and butter diets) or egg yolk (lard diet) to raise the diet cholesterol level at least five-fold. The effect of the type of dietary fat was that total plasma cholesterol (TPC) and HDL cholesterol concentrations were lowest while the safflower oil diet was fed, were significantly higher when butter fat diets were fed, and were highest when the egg yolk-lard based diets were fed. In addition, a significant effect of the elevated level of dietary cholesterol, independent of the type of dietary fat, was seen: a statistically significant negative correlation between TPC and HDL cholesterol concentrations was induced. In contrast, a positive correlation between TPC and HDL cholesterol concentrations was found at the lower dietary cholesterol level. Thus, the different factors (type of fat versus cholesterol) influenced lipoprotein metabolism in distinct yet related ways. Although average values for both plasma and HDL cholesterol concentrations were significantly higher in the grivet subspecies than in the vervet subspecies, the data for both subspecies fit the same regression lines. This outcome suggested that the subspecies differed in the magnitude of response rather than in the mechanism of response.

Electrocardiographic studies in African green monkeys (Cercopithecus aethiops).

Electrocardiograms obtained as part of a pre-experimental clinical examination were analyzed for a newly purchased group of 46 male African green vervets. In general, they were similar to electrocardiograms reported for other Old World species. An unexpectedly high prevalence (17%) of electrocardiogram abnormalities was found in this young (4--6 years of age) vervet population. The main abnormalities were nonspecific T wave flattening, left axis deviation, peaked P waves, RSR' patterns, and abnormally deep Q waves.

Reinfection of vervet monkeys (Cercopithecus aethiops) with Borrelia hermsii.

Two of 4 untreated vervet monkeys infected with Borrelia hermsii survived and were resistant to reinfection for at least 1 year. Seven of 9 vervets receiving tetracycline could be reinfected after 12 to 36 weeks. Immobilizine could be demonstrated for a shorter time in treated than in untreated vervets.

An experimental study of the central gustatory pathways in the monkey, Macaca mulatta and Cercopithecus aethiops.

1. The cortex at the base of the central fissure, that is the fronto-parietal operculum, represents primary sensory receptive cortex for gustatory modalities. 2. This primary receptive cortex for taste is linked to the anterior Island of Reil by short inter- and intracortical association fibers. Thus the anterior island is a gustatory association area involved in the subjective recognition of gustatory modalities. 3. The nucleus ventralis posteromedialis pars parvocellularis of the dorsal thalamus is the thalamic receptive nucleus for gustatory impulses in the macaque. 4. Fibers which originate at rostral glossopharyngeal levels from the dorsal visceral gray terminate in the contralateral mucleus ventralis posteromedialis pars parvocellularis.

Ultrastructure of sensory nerve terminals in the penis in green monkey (Cercopithecus aethiops sabaeus).

Tje paper describes the ultrastructure of axons in the endings of various types from the corium in the glans penis in green monkey. In the Meissner's endings the axons are mostly completely enveloped in the plasma of Schwann cells. They contain numerous mitochondria which are partially vacuolated or are quite converted into vacuoles. Next, there are pseudomyelinated figures, light vesicles and further organelles. In the papillar simple glomerular endings with accumulation of Schwann cells there are axons irregular in shape, eccentrically placed in the plasma of Schwann cell, rounded smaller axons either completely or partially surrounded by the plasma of Schwann cell and finally axons with a concentric system of lamellae up to four in number, In the complicated glomerular endings the axons vary in appearance and are enveloped in one to five lamellae of Schwann cells, which is typical of those formations. About some of these systems there is a sign of a capsule formed by an elongated lamella probably of the perineurium. When the axons are not enveloped in the plasma of Schwann cell, they are covered by the basement membrane. In close neighbourhood of the epidermis so-called free endings forming groups were found. The plasma of Schwann cell covers them either partially or completely or it again forms around them a lamellar system amounting up to four layers. It is noticeable that these axons are very poor in organelles. A comparison of the simple sensory corpuscles in the nose skin in hedgehog, the funtional properties of Meissner's endings and the simple corpuscles results in the view that the complexes having a larger amount of lamellae correspond to an extent to the simple sensory corpuscles ant that the Meissner's endings and the complicated glomerules are probably a morphological and functional equivalent of simple sensory corpuscles in the non-primate mammals and that the gloverular endings may also be the first (developmental) stage of the simple sensory corpuscles.

In vitro fertilization and preimplantation embryo development of African green monkeys (Cercopithecus aethiops).

Ovaries of five adult female African green monkeys were stimulated by repeated administrations of equine chorionic gonadotrophin (eCG), followed by a single administration of human chorionic gonadotrophin (hCG). Oocytes were collected from enlarged follicles 28 h after hCG administration and incubated in vitro for 288 h. Oocytes that had extruded the first polar body were inseminated with spermatozoa that had been incubated for 4 to 6 h in medium with caffeine and dibutyryl cyclic AMP. Of these oocytes, 66% were fertilized and the incidence of polyspermy was 37%. Eighty-two percent of the fertilized eggs cleaved, with some developing into expanded blastocysts.
