Effects of Ovulen-21, Demulen, Enovid-E and Ovral on zinc and copper levels in human plasma and endometrium during the menstrual cycle have been studied. Both plasma and endometrium copper levels were significantly elevated above the control values (P less than 0.001) in women taking oral contraceptives while the zinc levels remained reasonable constant.
In order to consider possible environmental factors related to the development of benign hepatic tumors, all reports of this disorder in the case records of five different hospitals in Rochester, New York, during the past 10 years were reviewed. Seven patients with benign hepatic tumors of the liver and two with peliosis hepatis were identified. Among the seven with a benign hepatic tumor, four were women currently receiving oral contraceptives; one had been receiving this medication in the past, and two had no history of exposure to any steroid whatsoever. The two patients with peliosis hepatis were receiving long-term androgenic anabolic steroids. Dilated, thin-walled vessels and vascular spaces were a more prominent feature of the tumor seen in four patients receiving oral contraceptive pills. It is emphasized in this report that benign hepatic tumors do occur in men and in patients with cirrhotic liver without the use of any kind of steroids.

An intravenous glucose tolerance test (GTT) was given to 116 women--82 nondiabetic subjects (NDS) and 34 diabetes suspects--before they received Ovulen for oral contraception. Subjects were followed for 1 to 4 years during Ovulen therapy. Twenty women using an intrauterine device showed no changes in glucose tolerance. Prompt, significant decline in tolerance was noted in NDS, persisting for the duration of the study. At least one abnormal test, indicating chemical diabetes, was noted in 13 per cent of the NDS. Similarly, prompt decline in tolerance, although not statistically significant, was observed in the suspect group. Fifteen per cent of suspects had at least one abnormal test. Chemical diabetes persisted during Ovulen therapy in suspects but not in NDS. No overt diabetes occurred. Based on greater concern regarding suspects, a procedure for monitoring carbohydrate tolerance is proposed for this group.
Serum prolactin concentrations were measured serially in eight normal women who used continuous chlormadinone acetate treatment (500 mug per day) for six months as part of an investigational new drug contraceptive study. Circulating prolactin was also measured in women using several additional oral contraceptive steroid preparations including a sequential mestranol-mestranol + chlormadinone acetate regimen. No increase in basal serum prolactin concentration was produced by chlormadinone acetate treatment, either alone or when added to estrogen therapy. These results suggest that chlormadinone acetate may not alter human mammary tissue through a prolactin mechanism.
Ergonovine maleate (EM) is a powerful stimulant of human tubal motility. It has been therefore thought that EM could interfere with normal ovum transport and serve as a contraceptive. The tube activating effects of EM and methyl EM were evaluated in 14 women and results confirmed that both are powerful stimulants of the human Fallopian tube. EM immediately post coitus significantly reduces the conception rate.
